Background. The purpose of this study was to verify the effect of whole-body cryotherapy (WBC) in rats on their antioxidant systems, lipid peroxidation products, and their total oxidative status at different exposure times and temperatures.Methods. Antioxidants in serum, plasma, liver, and erythrocytes were evaluated in two study groups following 1 min of exposure to −60°C and −90°C, for 5 and 10 consecutive days.Results. WBC increased the activity of superoxide dismutase, catalase in the group subjected to 5 and 10 days exposure, −60°C. The glutathione S-transferase activity increased in the groups subjected to 10 days WBC sessions. Total antioxidant capacity increased after 5 and 10 days of 1 min WBC, −60°C; a decrease was observed at −90°C. A decreased level of erythrocyte malondialdehyde concentration was observed at −60°C after 5 and 10 days of cryostimulation. An increased concentration was measured at −90°C after 10 days, and increase of erythrocyte malondialdehyde concentration after 5 days, −90°C.Conclusions. To the best of our knowledge, this is the first research showing the effect of WBC in rats at different exposure times and temperatures. The effect of cryotherapy on enzymatic and nonenzymatic antioxidant systems was observed in the serum of animals exposed to a temperature of −60°C in comparison to control.

Background. We examined the effectiveness of the systemic cryotherapy in terms of the temperature and duration of the therapeutic series measured by oxidative stress markers in the rat animal model. Methods. Antioxidants in serum, plasma, liver, and erythrocytes were evaluated in two study groups following 1 min exposure to − 60°C and − 90°C, for 5 and 10 days. Results. Superoxide dismutase activity in the tissues was lower than in the serum. The glutathione peroxidase was significantly higher in − 60°C than in − 90°C, in both 5 and 10 days of exposition. The liver catalase CAT were significantly lower in − 60°C when compared to − 90°C for 5 and 10 sessions of exposure. In all analysed tissues, the sessions of cryotherapy, – 60/5 and – 60/10, were more effective in reduction malondialdehyde than sessions of − 90/5 and − 90/10. The highest total antioxidant capacity was observed in the − 60/5 group. Conclusions. Whole body cryotherapy based on temperature − 60°C may be considered as more beneficial than − 90°C for most of the oxidative stress (OS) markers measured in the selected tissues. The temp. − 60°C is more beneficial than − 90°C when measured by activity of Total SOD, CAT, and GPx. The therapeutic sessions − 60/10 and − 60/5 were the optimal schemes of WBC model in terms of TAC and MDA amount.

Background: The activity of antioxidant enzymes and sirtuins (Sirt) decreases along with age, which is counteracted by aerobic training. Sirtuins increase antioxidant defence. Whole-body cryotherapy (WBC) increases total antioxidant capacity (TAC) in young men. The aim of our study was to assess the impact of 24 WBC treatments on the blood concentration of selected sirtuins and the level of antioxidant defence as well as oxidative stress index of training and non-training men depending on age. Methods: The study involved 40 males. In each group, there were 10 non-training older and young men (60 NTR and 20 NTR), and 10 older and young long-distance runners (60 TR, 20 TR). During an 8-week period, participants underwent 24 WBC treatments (3 min −130 °C), which were performed three times a week (Monday, Wednesday, Friday). The concentrations of Sirt1, Sirt3, TAC, total oxidative status and the activity of superoxide dismutase (SOD), catalase and glutathione peroxidase (GPx) in the blood were determined before 1 WBC and after 1 WBC, 12 WBC and 24 WBC. Results: After 1 WBC, the activity of GPx and the concentration of Sirt1 and TAC in 60 TR and TAC in 60 NTR increased. After 12 WBC, the level of Sirt1 in 20 NTR and SOD in 20 TR increased. After 24 WBC, the level of Sirt1 increased in 60 TR and in 20 NTR, Sirt3 in 60 TR and SOD in 20 TR. Conclusions: Cryogenic temperatures increase blood levels of Sirt1 and Sirt3 and systemic antioxidant defence in men, but the effect is dependent on age, level of performed physical activity and the number of applied treatments.

Background and aims. Periodontitis is a chronic inflammation that involves nitro-oxidative stress with damaging periodontal structural effects. We aimed to evaluate the consequences of low-dose cimetidine on nitro-oxidative stress in periodontitis. Methods. A rat model of ligature-induced periodontitis was used. After two weeks, the periodontitis groups were treated with cimetidine, aminoguanidine, N-nitro-L-arginine methyl ester and trolox for one week. On day 21, blood was drawn and the serum analyzed for measurement of total nitrites and nitrates, total oxidative status, total antioxidant response, and oxidative stress index. Results. Cimetidine had an inhibitory effect on the synthesis of nitric oxide (p=0.001), total oxidative status (p=0.01) and oxidative stress index (p=0.01). Total antioxidant reactivity was increased by cimetidine (p=0.01). The effects of cimetidine were almost like those of aminoguanidine, NG-nitro-L-arginine methyl ester, and trolox. Conclusions. Low-dose cimetidine can be used as adjunctive host modulatory therapy in chronic periodontitis because it reduces nitro-oxidative stress.

CeO2 nanoparticles (CNPs) as effective ROS scavengers exhibit potent antioxidant activity. In this study the effect of CNPs investigated was on HO-1, NQO1, and GCLC expression in the streptozotocin (STZ)-induced diabetic rats. Twenty-four male Wistar rats were divided into 4 groups: controls did not receive any treatment; diabetic rats received STZ (60 mg/kg daily); CNPs group received CNPs 30 mg/kg daily for 2 weeks; and rats in STZ + CNPs group received CNPs 30 mg/kg daily for 2 weeks following STZ injection. Oxidative stress was evaluated by measurement of total antioxidant capacity (TAC) and total oxidative status (TOS levels). HO-1, NQO1, and GCLC expression was measured using quantitative real-time PCR. Following STZ injection, significant lower levels of TAC and higher levels of TOS were observed. CNPs could alleviate deleterious effects of diabetes through the enhancement of TAC levels and a significant decline in TOS levels. HO-1, NQO1, and GCLC expression in the diabetic rats were lower than controls. HO-1, NQO1, and GCLC was upregulated in the diabetic rats treated with CNPs. There were significant correlations between NQO1 and GCLC, NQO1 and HO-1, and between HO-1 and GCLC expression. Moreover, Nrf2 was associated with NQO1, GCLC, and HO-1 expression. CNPs as Nrf2 upregulator confer protection against oxidative stress in the testes of STZ-induced diabetic rats by upregulating HO-1, GCLC, and NQO1 cytoprotective genes.

Objective. The aim of the study was to estimate the impact of whole-body cryotherapy (WBC) and subsequent kinesiotherapy on oxidative stress and lipid profile when performed in a closed cryochamber on healthy subjects. Material and Methods. The effect of ten WBC procedures lasting 3 minutes a day followed by a 60-minute session kinesiotherapy on oxidative stress and lipid profile in healthy subjects (WBC group, n=16) was investigated. The WBC group was compared to the kinesiotherapy only (KT; n=16) group. The routine parameters of oxidative stress (antioxidant enzymatic and nonenzymatic antioxidant status, lipid peroxidation products, total oxidative status (TOS), and oxidative stress index (OSI)) and lipid profile were estimated one day before the beginning and one day after the completion of the research program. Results. After treatment, in the WBC group, a significant decrease of oxidative stress markers (TOS and OSI) and a significant increase of total antioxidant capacity were observed. The activity of plasma SOD-Mn and erythrocyte total SOD increased significantly in the WBC group. In the KT group, the erythrocyte activity of total SOD, CAT, and GR decreased significantly after the treatment. The levels of T-Chol and LDL-Chol decreased significantly after treatment in both groups, but the observed decrease of these lipid parameters in the WBC group was higher in comparison to the KT group. The level of TG decreased significantly after treatment in the WBC group only. Conclusion. WBC performed in a closed cryochamber followed by kinesiotherapy improves lipid profile and decreases oxidative stress in healthy subjects.

The aim of the study was to determine the effect of single whole-body cryotherapy (WBC) session applied prior to submaximal exercise on the activity of antioxidant enzymes, the concentration of lipid peroxidation products, total oxidative status, and the level of cytokines in blood of volleyball players. The study group consisted of 18 male professional volleyball players, who were subjected to extremely cold air (−130∘C) prior to exercise performed on cycloergometer. Blood samples were taken five times: before WBC, after WBC procedure, after exercise preceded by cryotherapy (WBC exercise), and before and after exercise without WBC (control exercise). The activity of catalase statistically significantly increased after control exercise. Moreover, the activity of catalase and superoxide dismutase was lower after WBC exercise than after control exercise (P<0.001). After WBC exercise, the level of IL-6 and IL-1βwas also lower (P<0.001) than after control exercise. The obtained results may suggest that cryotherapy prior to exercise may have some antioxidant and anti-inflammatory properties. The relations between the level of studied oxidative stress and inflammatory markers may testify to the contribution of reactive oxygen species in cytokines release into the blood system in response to exercise and WBC.

<b>Introduction:</b> Oxidative stress is largely responsible for numerous skin complications that occur in the course of various diseases as well as accelerated skin aging. A high amino acid diet, supplemented with whey protein concentrate (WPC), is well-balanced and has well-absorbing proteins, which are an ideal source of essential amino acids. <br/><b>Purpose:</b> To assess what changes will occur in the antioxidant barrier of unharmed skin of rats on a high amino acid diet. <br/><b>Materials and methods:</b> The study was conducted on sexually mature male Wistar rats (160-180g): 1. control (standard feed), 2. high amino acid diet (WPC-80 80% whey protein) administered for 7 days at a dose of 0.3g/kg of body weight, 3. WPC-80 for 7 days at a dose of 0.5g/kg of body weight, 4. WPC-80 for 14 days at a dose of 0.3g/kg of body weight, 5. WPC-80 for 14 days at a dose of 0.5g/kg of body weight. Total antioxidant capacity, total oxidative status and oxidative stress index were determined. <br/>Results:</b> Enrichment of a standard diet with WPC-80 did not affect the total oxidative status of undamaged healthy rat skin. This study shows that a diet rich in amino acids in rats caused an increase in total antioxidant capacity, but statistically significant values were obtained after 14 days of administering WPC at a dose of 0.5mg/kg of body weight. <br/><b>Conclusions:</b> Enrichment of a standard diet with WPC-80 strengthens the antioxidant barrier in unwounded healthy rat skin.

Background: ‪Osteoarthrosis (OA) is the most common disease affecting mankind. There are many physiotherapeutic methods of treating OA symptoms. One of them is whole body cryotherapy (WBCT). The aim of this study was to present the impact of WBCT combined with kinesiotherapy on functional efficiency and changes in plasma concentrations of serotonin and melatonin in patients with lumbar spine OA.. Material and methods: ‪60 patients underwent a series of WBCT combined with kinesiotherapy (group A). 60 patients underwent a series of kinesiotherapy alone (group B). Before and after the therapy patients were examined by Rolland Morris Disability Questionnaire (RMDQ), and blood samples were collected for laboratory assessment. Results: ‪Following therapy, the functional efficiency (RMDQ) improved by 33% in both study groups (p < 0.0001). After the therapy, both in the group of patients undergoing WBCT combined with exercises (group A, p = 0.032) and the cryotherapy alone (group B, p < 0.0001), the plasma melatonin concentration significantly increased. The rise of plasma serotonin concentrations was higher in group B as compared to group A, but in both study groups the increase in plasma serotonin was statistically significant (p < 0.0001).. Conclusions: ‪WBCT combined with therapeutic exercises as well as kinesiotherapy used alone resulted in an increase in melatonin and serotonin plasma levels and an improvement in functional efficiency in patients with lumbar spine OA.

Metabolic syndrome (MS) is the association of three or more pathologies among which obesity, hypertension, insulin resistance, dyslipidemia, and diabetes are included. It causes oxidative stress (OS) and renal dysfunction. Hibiscus sabdariffa L. (HSL) is a source of natural antioxidants that may control the renal damage caused by the MS. The objective of this work was to evaluate the effect of a 2% HSL infusion on renal function in a MS rat model induced by the administration of 30% sucrose in drinking water. 24 male Wistar rats were divided into 3 groups: Control rats, MS rats and MS + HSL rats. MS rats had increased body weight, systolic blood pressure, triglycerides, insulin, HOMA index, and leptin (p ≤ 0.04). Renal function was impaired by an increase in perfusion pressure in the isolated and perfused kidney, albuminuria (p ≤ 0.03), and by a decrease in clearance of creatinine (p ≤ 0.04). The activity of some antioxidant enzymes including the superoxide dismutase isoforms, peroxidases, glutathione peroxidase, glutathione-S-transferase was decreased (p ≤ 0.05). Lipoperoxidation and carbonylation were increased (p ≤ 0.001). The nitrates/nitrites ratio, total antioxidant capacity, glutathione levels and vitamin C were decreased (p ≤ 0.03). The treatment with 2% HSL reversed these alterations. The results suggest that the treatment with 2% HSL infusion protects renal function through its natural antioxidants which favor an improved renal vascular response. The infusion contributes to the increase in the glomerular filtration rate, by promoting an increase in the enzymatic and non-enzymatic antioxidant systems leading to a decrease in OS and reestablishing the normal renal function.
