The objective of this study was to investigate the effect of L-carnitine (LC), hypotaurine (HT), and taurine (T) on the quality of frozen-thawed chicken semen. Pooled semen samples were divided into seven aliquots (control, 1 mM LC, 5 mM LC, 1 mM HT, 10 mM HT, 1 mM T, and 10 mM T) and subjected to cryopreservation. Postthaw sperm motility was determined by IVOS system and sperm characteristics were assessed with fluorochromes and flow cytometry. The highest sperm motility and the highest percentage of viable sperm were in the HT1 group (P<0.01andP<0.05) following cryopreservation. After thawing, we observed a higher percentage of sperm without apoptosis and membrane reorganization changes in the LC1 and T1 group when compared to the control (P<0.05). There was a higher percentage of live sperm without lipid peroxidation (LPO) in all treatments (P<0.01;P<0.05), when compared to the control group. The percentage of sperm with high mitochondrial potential significantly increased with LC1, T1, and T10 (P<0.05). Supplementation of the diluent with LC1, LC5, and T1 significantly (P<0.05) reduced DNA susceptibility to fragmentation, compared to the control and HT1 groups. These results indicate that the addition of examined antioxidants improves the quality of cryopreserved chicken semen.

Plasma membrane composition has impact on phase transition from liquid crystal to gel state of cooled sperm cell. The incorporation of polyunsaturated fatty acids increases its fluidity and can contribute to sperm motility. The aim of this study was to compare the effect of adding docosahexaenoic acid (DHA) and ?-tocopherol (?-Toh) to the cooling extender, singly or combined, to the equine sperm parameters, submitted to cooling, up to 72 hours. Two ejaculates of ten stallions collected with artificial vagina were used, and evaluated for motility, plasma membrane integrity, chromatin fragmentation, mitochondrial activity and lipid peroxidation, according to the following treatments: C; DHA; ?-Toh; DHA/?-Toh; EtOH 100: and EtOH 140 (corresponding to control; 10 ng mL-1 of DHA; 2 mM of ?-Toh; : 10 ng mL-1 of DHA + 2 mM of ?-Toh; 100 µL of ethanol and 140 µL of ethanol respectively). DHA treatment showed higher motility (68.2 ± 12.3; p < 0.05) when compared to control (62.1 ± 16.2), DHA/?-Toh (61.3 ± 12.7) and EtOH (58.1 ± 8.6) groups. In lipid peroxidation assay, the control group showed 2,506.2 ± 796.4 ng of MDA 108 spermatozoa-1, being significantly higher (p < 0.05) than the groups treated with DHA (2,036.0 ± 687.0), ?-Toh (1,890.8 ± 749.5) and DHA/?-Toh (1,821.1 ± 627.2). In conclusion, ?-Toh was effective in diminishing lipid peroxidation of equine sperm subjected to cooling, and DHA improved sperm motility and, in spite of being a polyunsaturated fatty acid with high susceptibility to peroxidation, reduced lipid peroxidation.

This study was conducted to investigate the effect of docosahexanoic acid (DHA) supplementation in BioXcell extender on the quality of frozen–thawed bull semen. Twenty-four ejaculates were collected from three bulls (eight from each bull). Ejaculates with motility ≥70% and normal morphology ≥80% were extended into BioXcell extender to which 0 (control), 3, 5, 10 or 15 ng mL–1 DHA was added. The supplemented semen samples were incubated at 37°C for 15 min for DHA uptake by spermatozoa. Later, samples were cooled for 2 h at 5°C and packaged into 0.25-mL straws, frozen in liquid nitrogen for 24 h and subsequently thawed for evaluation. Results are presented as percentages ± s.e.m. Supplementation with DHA at 3 ng mL–1 significantly improved sperm functional parameters including sperm motility, normal morphology, viability, acrosome integrity and membrane integrity when compared with other supplemented groups and the control. Lipid peroxidation increased as the incorporation of DHA supplementation increased. In conclusion, 3 ng mL–1 concentration of DHA resulted in superior quality of frozen–thawed bull spermatozoa and is suggested as the optimum level of DHA to be added into BioXcell extender.

Background: Hesperetin is a bioflavonoid compound, largely used in Chinese traditional medicine and found plenty in citrus fruits. Hesperetin has beneficial effects against different diseases. The sperm cryopreservation process is a common method that is used in infertility laboratories. It has been reported that during the cryopreservation process, the quality of sperm is significantly reduced. Objective: To investigate the effect of hesperetin on the quality of human spermatozoa during the cryopreservation process. Materials and Methods: In this experimental study, 22 sperm sample of normozoospermia men who referred to the infertility department of the Shariati Hospital (Tehran, Iran) Between October and November 2019 were collect and divided into three groups as: 1) fresh, 2) control (frozen-thawed group without treatment), and 3) treatment group as frozen-thawed samples supplemented with 20 μM hesperetin. Motility, Viability, morphology, Apoptotic-like changes, intracellular H2O2, intracellular O2−, and lipid peroxidation (LPO) was measured. Results: Hesperetin treatment during the cryopreservation process of human sperm significantly improved the viability, motility, and morphology rates of the spermatozoa after frozen-thawed process in control group (p < 0.01). In addition, it significantly reduced the reactive oxygen species (ROS) level, LPO level and increased the percentage of viable sperm cells with intact plasma membrane (p < 0.01) after frozen-thawed process. Conclusion: Hesperetin can improve the quality of human sperm and also protect human sperm against reactive oxygen species, LPO, and apoptosis during the cryopreservation-thawing process. The aim of this study was to investigate the effects of different extenders viz. Tris citric acid fructose egg yolk (TCFEY), Tris citric acid glucose egg yolk (TCGEY), Egg yolk citrate fructose (EYCF) and Egg yolk citrate glucose (EYCG) on the quality of ram spermatozoa during preservation at 4°C. Semen samples showing more than 3+ mass motility and 70% progressive motility were pooled and subsequently divided into four aliquots. Each aliquot was extended separately in four different extenders viz. TCFEY, TCGEY, EYCF and EYCG and stored at 4°C up to 72h. The quality of spermatozoa on the basis of percentage of sperm motility, live sperm, morphological abnormalities, intact acrosome and hypoosmotic swelling test (HOST) reacted spermatozoa was evaluated immediately after extension in particular extenders (0 h), 24 h, 48 h and 72 h after preservation at 4°C. The percent sperm motility was significantly (P<0.01) higher for TCFEY and TCGEY than EYCF and EYCG at 72 h of preservation at 4°C. The percent HOST reacted spermatozoa and intact acrosomes were significantly (P<0.01) higher and morphological abnormalities were significantly (P<0.01) lower for Tris based fructose extender than other three extenders at 72 h at 4°C. In conclusion, Tris citric acid fructose egg yolk (TCFEY) was found the best in maintaining the quality of ejaculated ram spermatozoa during preservation for 72 h at 4°C.

In the process of cryopreservation of dairy goat semen, it will face many threats such as oxidative damage, which will affect the motility and plasma membrane function of sperm. As an endogenous antioxidant in animals, glutathione (GSH) can significantly improve the quality of thawed sperm when added to the frozen diluent of semen of pigs and cattle. In this study, different concentration gradients of GSH [0 mmol/L (control), 1, 2, 3, 4 mmol/L] were added to the frozen diluent of Guanzhong dairy goat semen. By detecting the sperm motility parameters, acrosome intact rate and plasma membrane intact rate after thawing, the effect of GSH on the cryopreservation of dairy goat semen was explored. Sperm motility parameters were measured with the computer-aided sperm analysis (CASA) system (total power, TM; forward power, PM; linearity, LIN; average path speed, VAP; straight line speed, VSL; curve speed, VCL; beat cross frequency, BCF). The sperm acrosome integrity rate after thawing was detected by a specific fluorescent probe (isothiocyanate-labeled peanut agglutinin, FITC-PNA), and the sperm plasma membrane integrity rate after thawing was detected by the hypotonic sperm swelling (HOST) method. Reactive oxygen species (ROS) kit, malondialdehyde (MDA) kit, superoxide dismutase (SOD) kit, glutathione peroxidase (GSH-PX) kit were used to detect various antioxidant indicators of thawed sperm. in vitro fertilization experiment was used to verify the effect of adding glutathione on sperm fertilization and embryo development. The results showed that when the concentration of glutathione was 2 mmol/l, the sperm viability, plasma membrane intact rate, and acrosome intact rate were the highest after thawing, reaching 62.14, 37.62, and 70.87% respectively, and they were all significantly higher. In terms of antioxidant indexes; the values of SOD and GSH-PX were 212.60 U/ml and 125.04 U/L, respectively, which were significantly higher than those of the control group; The values of ROS and MDA were 363.05 U/ml and 7.02 nmol/L, respectively, which were significantly lower than the control group. The addition of 2 mmol/L glutathione significantly improves the fertilization ability of sperm. In short, adding 2 mmol/l glutathione to the semen diluent can improve the quality of frozen Guanzhong dairy goat sperm.

Two groups of five medium Wool Peppin Merino rams were individually penned in an animal house for 18 months. During this time the normal seasonal pattern of changes in day length was reversed for one group of rams (treatment). The control group of rams received normal daylight at Trangie (32�S). From late September 1960, semen was collected once every 14 days from all rams by electro-ejaculation and semen quality was assessed. Significant treatment X months of year interactions found for most semen characteristics. The semen of treated rams had a lower pH during October 1960, and a higher proportion of normal sperm and number of sperm per ejaculate during November 1960, than the semen of control rams. During the period February to June 1961 the semen of treated rams was of a lower quality than the semen of control rams. During November- December 1961 and January to March 1963, the semen of treated rams had a significantly lower proportion of live sperm (P<0.005) than the semen of control rams. From these results it was not possible to determine if the daily photoperiod exerted a direct or indirect effect on the semen quality of Merino rams.

Reactive oxygen species (ROS) are physiologically produced by spermatozoa and leukocytes present in the seminal plasma. In low concentrations, ROS play an important role in sperm function because they are required for sperm fertilizing capacity, mainly during sperm capacitation and hyperactivation (Saleh RA and Agarwal A 2002 J. Androl. 23, 737-752). However, an imbalance between the formation of free radicals and the capacity for defense of the antioxidant mechanisms may lead to cell damage (Rover Jr L et al. 2001 Química Nova 24, 112-119). Spermatozoa are sensitive to lipid peroxidation due to its high content of polyunsaturated fatty acids and low concentrations of protective enzymes (Sharma RK and Agarwa LA 1996 Urology 48, 835-850). The aims of the present study were to compare ROS content among the 3 fractions of canine semen and to correlate these values with sperm variables. Semen samples were collected from 15 healthy dogs of distinct breeds aged 2 to 6 years. Sperm analysis was performed through motility, forward progressive velocity, morphology, and the percentage of viable sperm with the use of the eosin/nigrosin stain. The determination of thiobarbituric acid reactive substances (TBARS) was used to estimate the degree of lipid peroxidation in each sperm fraction. Values were compared using ANOVA and Tukey’s test for multiple comparisons at a significance level of 5%. Pearson correlation was used to calculate the relationship between sperm variables in the second fraction. Sperm motility, velocity, and percentage of viable sperm were within the normal range for canine semen: 84±2%, 3.4±0.1%, and 83 ± 2%, respectively. The sperm-rich fraction presented statistically higher concentrations of TBARS (1474.19 ± 245.78 ng mL-1) compared to the first and third fractions (579.41 ± 171.23 and 399.62 ± 58.08 ng mL-1, respectively; P < 0.05), indicating that spermatozoa and epididymal secretions are the main source of free radicals. No statistical correlation among TBARS and sperm motility and velocity were verified. However, a positive correlation was observed between the percentage of sperm proximal droplets and TBARS (r = 0.44, P = 0.7). This result suggests that a high incidence of sperm proximal droplets can enhance ROS formation in seminal plasma. Hence, canine sperm presenting delayed maturation in the epididymes produce higher concentrations of free radicals. In fact, sperm production of ROS occurs mainly by abnormal cells, especially the ones containing cytoplasm residues (Gomez E et al. 1998 Int. J. Androl. 21, 81-94). However, no protective effect of the sperm distal droplets was verified in canine semen as observed elsewhere for the bovine spermatozoa (Nichi M et al. 2007 Theriogenology 67, 334-340). In conclusion, spermatozoa and epididymal fluids are the primordial source of free radicals in canine seminal plasma, mainly when sperm proximal droplets are present.

Density gradient centrifugation with PureSperm® (PureSperm® 40 + PureSperm® 80; Nidacon International, Mölndal, Sweden) has been satisfactorily used to enhance the quality of dog semen samples; however, no studies have been performed on the effect of single layer centrifugation (SLC) with PureSperm® on frozen–thawed dog semen. The aim of this study was to investigate if SLC with PureSperm® 80 can improve the post-thaw semen quality of dog. Semen from 5 dogs was collected by digital manipulation. Two ejaculates from each dog were centrifuged with Tris-based extender, supernatant was removed, and sperm pellet was suspended to a final concentration of 300–400 × 106 sperm mL–1 with CaniPROTM Freeze A plus 20% egg yolk at 22°C. Extended semen was cooled to 5°C within an hour and then diluted to a final concentration of 150–200 × 106 sperm mL–1 in CaniPROTM Freeze B plus 20% egg yolk at 5°C, loaded in 0.5-mL plastic straws and frozen horizontally in ranks placed 4 cm above the surface of liquid nitrogen vapors for 10 min, after which they were directly placed in liquid nitrogen. After 24 to 48 h of storage, straws were thawed in a water bath at 37°C for 30 s. After thawing, semen samples were divided in 2 aliquots: one of them was used as control and the other one was processed by SLC PureSperm® 80. Assessment of sperm motility (assessed by computerized-assisted semen analysis), morphology (Diff-Quick staining), and viability [triple fluorescent stain of propidium iodine/isothiocyanate-labeled peanut (Arachis hypogaea) agglutinin/Rhodamine 123] were evaluated in control and treated semen samples. Data were studied by ANOVA. Results are expressed as mean ± SEM. Significant (P < 0.001) differences were found between SLC-treated and control semen for sperm motility (percentage of total motile spermatozoa: 93.65 ± 0.05 v. 83.79 ± 0.13; percentage of progressive motile spermatozoa: 79.38 ± 6.66 v. 54.61 ± 16.11), morphology (86.45 ± 0.01 v. 83.51 ± 0.01), and viability (percentage of viable sperm with an intact acrosome: 58.32 ± 0.04 v. 36.50 ± 0.17; percentage of viable sperm with an acrosome reaction: 2.81 ± 0.01 v. 9.74 ± 0.21). Based on our results, we can conclude that SLC with PureSperm® 80 is an alternative and successful method for improving the quality of frozen–thawed dog spermatozoa, selecting good-quality spermatozoa (motile, morphologically normal, viable, and acrosome intact spermatozoa) from the rest of the semen sample.

The article contains information on the state of the processes of lipid peroxidation (LPO) in fresh cows under the influence of stress reaction caused by labor and the beginning of the lactation period. To reduce oxidative stress in the body of fresh cows, antioxidants were used in the form of preparations “E-selenium” and “Butofan”. The aim of the research was to determine the effect of vitamin-containing preparations “E-selenium” and “Butofan” on the productivity of fresh cows, the quality of milk and butter, made from it. The studies were carried out during the winter-stall period on one of the farms of Ryazan district of Ryazan region and involved 12 fresh black-and-white cows aged 4 years. The control group of animals was intact. As a result of the investigations, it was found that due to the action of the above preparations, which have antioxidant properties, the productivity parameters in the experimental groups of animals were higher compared to the control group, because they had an inhibiting effect on lipid peroxidation processes in the organism and activated the work of its own antioxidant system.
