Purpose.This study aimed to evaluate the effect of chronic treatment with chaetomellic acid A (CAA) on oxidative stress and renal function in a model of renal mass reduction.Methods. Male Wistar rats were subjected to 5/6 nephrectomy (RMR) or sham-operated (SO). One week after surgery, rats have been divided into four experimental groups: RMR: RMR rats without treatment(n=14); RMR + CAA: RMR rats treated with CAA(n=13); SO: SO rats without treatment(n=13); and SO + CAA: SO rats treated with CAA(n=13). CAA was intraperitoneally administered in a dose of 0.23 µg/Kg three times a week for six months.Results.RMR was accompanied by a significant reduction in catalase and glutathione reductase (GR) activity(p<0.05)and a decrease in reduced glutathione (GSH)/oxidized glutathione (GSSG) ratio. CAA administration significantly increased catalase and GR activity(p<0.05)and increased GSH/GSSG ratio, but no significant difference between the treated and nontreated groups was found in this ratio. No significant differences were found between the RMR groups in any of the parameters of renal function. However, CAA administration slightly improves some parameters of renal function.Conclusions.These data suggest that CAA could attenuate 5/6 RMR-induced oxidative stress.

Introduction: Cnidoscolus aconitifolius is considered to be an important and effective medicinal plant in folklore remedies where it has been applied as an alternative therapy for the treatment of various ailments. Aim: The present study aims to determine the effects of Cnidoscolus aconitifolius on lipid profile and some oxidative stress markers of male Wistar rats. Methodology: A total of 15 male wistar rats were procured for the study and randomly assigned into three groups of 5 rats each. Group 1 served as control and received distilled water only. Group 2 received 200 mg/kg and group 3 received 400mg/kg of the hydromethanolic (1:4) extract of Cnidoscolus aconitifolius which was administered as single daily dose using oral cannula. On completion of treatment, blood samples were collected by cardiac puncture for determination of some serum lipid parameters and oxidative stress markers. Results: Results showed that there were no significant difference in the serum level of total cholesterol, triglyceride, low density lipoprotein cholesterol and high density lipoprotein cholesterol at both doses of the extract, compared to control. However, compared to control, there was a significant (p<0.05) increase in the activity of superoxide dismutase and glutathione reductase but significant reduction in malondialdehyde level. The catalase enzyme activity was not significant. Conclusion: The result obtained suggest that the extract may be useful in reducing oxidative stress by improving some antioxidant enzyme activities and may also prevent cell death due to lipid peroxidation.

The aim of the present study was to evaluate the subchronic effects of 3,4-methylenedioxymethamphetamine on several oxidative stress markers: index of lipid peroxidation (ILP), superoxide dismutase (SOD) activity, superoxide radical (O2.-) levels, and reduced glutathione (GSH) levels in the frontal cortex, striatum and hippocampus of the rat. The study included 64 male Wistar rats (200-250g). The animals were treated per os with of 5, 10, or 20 mg/kg of 3,4-methylenedioxymethamphetamine (MDMA) every day for 15 days. The subchronic administration of MDMA resulted in an increase in ILP, SOD and O2.-, and a decrease in GSH, from which we conclude that oxidative stress was induced in rat brain.

Aim. This study aimed to assess the antioxidant activity of quercetin (Q) in an experimental model of cirrhosis induced by CCl4 inhalation. Materials and Methods. We used 25 male Wistar rats (250 g) that were divided into 3 groups: control (CO), CCl4, and CCl4+Q. The rats were subjected to CCl4 inhalation (2x/week) for 16 weeks, and they received phenobarbital in their drinking water at a dose of 0.3 g/dL as a P450 enzyme inducer. Q (50 mg/Kg) was initiated intraperitoneally at 10 weeks of inhalation and lasted until the end of the experiment. Statistical analysis was by ANOVA Student Newman-Keuls (mean±SEM), and differences were considered statistically significant when P<0.05. Results. After treatment with quercetin, we observed an improvement in liver complications, decreased fibrosis, as analyzed by picrosirius for the quantification of collagen, and decreased levels of matrix metalloproteinase 2 (MMP-2) compared with the CCl4 group. It also reduced oxidative stress, as confirmed by the decrease of substances reacting to thiobarbituric acid (TBARS), the increased activity of antioxidant enzymes, and the reduced glutathione ratio and glutathione disulfide (GSH/GSSG). Conclusion. We suggest that the use of quercetin might be promising as an antioxidant therapy in liver fibrosis.

The study was conducted on male Wistar rats. We used 4 mathematically related series of doses prepared by sequential dilution of water from a man-made lake according to the scheme: arsenic: 0.15 - 0.05 - 0.016 - 0.0055, antimony: 0.68 - 0.227 - 0.075 - 0.025 mg/l. The animals were examined before inoculation (background), on days 40 and 90 of the experiment, taking into account the processes of cumulation of chemical elements in the kidneys. New data on the nature of nephrotoxic effects of arsenic and antimony in the long-term combined intake into the body of laboratory animals have been obtained. Key words: water from a man-made lake, arsenic and antimony compounds, Wistar rats, nephrotoxicity.
