An efficientin vitroregeneration protocol from seed culture has been established successfully forDendrobium chrysotoxum, an epiphytic orchid having tremendous ornamental and medicinal values. Seed germination response was encouraging in Mitra (M) medium enriched with different combinations of auxins and cytokinins. Medium supplemented with 0.4% activated charcoal (AC), 2 mg/L 6-benzyl amino purine (BAP), and 2 mg/L indole-3-acetic acid (IAA) produced best seed germination percentage in 2 weeks of culture. Incorporation of higher concentration of kinetin (KN) or BAP in combination with low auxin in medium induced pronounced shooting and leaf formation. Reduction in leaf development was evident when cytokinins exist singly in medium indicating synergistic effect of auxin and cytokinin in leaf induction. Presence of elevated level of indole-3-butyric acid (IBA) or 1-naphthalene acetic acid (NAA) with low cytokinin content in medium generated morein vitrorooting, though IBA was found to be more effective in rooting induction as compared to NAA. Thein vitroprotocol for asymbiotic seed germination developed from the present investigation can be used for rapid mass propagation of this highly importantDendrobiumorchid species.

In vitro seed germination and seedling development of Phaius tancarvilleae (L’Her.) Blume. was carried out on 0.8%(w/v) agar solidified MS Medium (Murashige and Skoog, 1962) without hormones or supplemented with different concentration and combination of Naphthalene acetic acid (NAA) and 6-benzylaminopurine (BAP). MS medium supplemented with 0.5 mg/l of BAP was the most ideal condition for early seed germination, protocorm formation and development of seedlings. Germination started after 7 weeks of culture and complete seedlings were obtained after 24 weeks of culture. This protocol might be helpful for mass propagation of orchids by asymbiotic seed germination. Orchid seeds are very small, dust like in appearance, fusiform in shape, lacking endosperm and have undifferentiated embryo covered by transparent seed coat. Mycorrhizal association is required for seed germination of naturally growing orchids. In this symbiotic association, orchid species are dependent on mycorrhizal association for supply of mineral nutrients. In in-vitro condition, such demand of minerals may be compensated by external supply of sugar and mineral nutrients that are required for seed germination of orchid. Several orchids are responded by this asymbiotic seed culture and have commercial importance. Therefore, formulation of efficient in-vitro protocol is important for commercially important orchid species as well as endangered orchid species for conservation. This review paper is focused on various aspects of asymbiotic seed germination of orchids and the role of organic additives in successful seed germination.

Peppermint (Mentha piperita), sweet basil (Ocimum basilicum), and coriander (Coriandrum sativum) are important medicinal plants in the pharmacological industry. These plants are produced in commercial scale but their seeds exhibit low germination percentages under favorable germination conditions. Enhancing seed germination is thus crucial for improving the production of these plants. The influence of gibberellic acid (GA3), indole-3-acetic acid (IAA), indol-3-butyric acid (IBA), and naphthalene acetic acid (NAA) on seed germination of the three plants were investigated. The seeds were soaked in each plant growth regulator at 50, 100, and 150 mg·L−1 for 24 hours at 25 ± 2 °C. Seed germination was checked daily for 20 days and germination parameters including final germination percentage (FGP), corrected germination rate (CGRI), and number of days lapsed to reach 50% of FGP (GT50) were recorded. The phosphorus and protein contents were determined in germinated seedlings on day 21 of culture. All plant growth regulators enhanced seed germination as compared with control. However, GA3 improved seed germination more than IAA, IBA, and NAA. GA3 at 100 mg·L−1 significantly increased the FGP from 22.3% and 33.3% (control) to 74% and 65.6% for peppermint and sweet basil, respectively. Low concentration of GA3 at 50 mg·L−1 increased the FGP for coriander from 27% to 52.3%. GA3 also increased CGRI, GT50, phosphorus, and protein contents in germinated seedlings as compared with control. Seeds of peppermint, sweet basil, and coriander possess a physiological dormancy that could be elevated by GA3 presowing treatment. This study established a successful methodology for optimizing seed germination to satisfy the demand for the medicinal parts of these plants in the pharmacological industry.

A comparative study of in vitro seed germination of two endangered orchid species, viz. Cymbidium elegans Lindl. and Dendrobium densiflorum Lindl. ex Wall., was carried out on Murashige and Skoog's (MS) medium, supplemented with different concentrations and combination of 6-benzylaminopurine (BAP) and á-Naphthalene acetic acid (NAA). The hormone-free MS medium and MS medium supplemented with various growth hormones were found effective for in vitro seed germination of both species. However, the seeds of these two species showed variation in their germination behavior. Hormone-free MS basal medium was found most effective for seed germination of D. densiflorum; whereas, basal medium supplemented with BAP (1mg/l) was effective for C. elegans. The seeds of D. densiflorum showed quick response in earlier germination, protocorm formation and further development into seedlings in comparison to C. elegans. In C. elegans, germination of immature seeds started after nine weeks of inoculation; whereas in D. densiflorum, the initiation of germination started after five weeks of culture. The variations in seed germination, protocorm formation and seedling differentiation in the two orchid species might be due to the differences in their genetic constitution and the presence of different endogenous growth stimulating substances present in their seeds. The present study has provided useful information for in vitro clonal mass multiplication of these commercially important orchid species.  The immature seeds of Dendrobiumchryseum, asympodial epiphytic orchid with yellow flowers, were cultured in vitro, and the resultant protocorms were used as explants for seedling development. Protocorms were cultured on½ M.S. medium fortified with Kinetin (Kn), 6-Benzylaminopurine (BAP), and Gibberellic Acid (GA3) in three concentrations (0.5mg/l, 1.0mg/land 2.0 mg/l) both alone and supplemented with 5% and 10% coconut water (C.W.). The highest number of shootsofD. chryseum developed on ½ - M.S. medium fortified with 2.0mg/lofKn and10% C.W. and the longest shootsdeveloped on ½ M.S. media fortified with 1.0mg/lGA3, and 10% C.W. The shoot derived from protocorms were placed in ½ M.S. medium fortified with three different rooting hormones, Indole -3- acetic acid (IAA), Indole -3-butyric acid (IBA) and α-Naphthalene acetic acid (NAA) in different concentrations alone as well as with each 1.0mg/l hormone combined with 10% C.W. The most effective of these media was ½ M.S. medium fortified with 1.5 mg/l IAA for rooting as well as for the production of longest roots. The present study could be useful for standardizing the protocol for mass propagation of the endangered orchid Dendrobiumchryseum.

Annona crassiflora Mart known as 'araticum', 'marolo' or 'field araticum' is a typical fruit from the Cerrado biome of Brazil with socio-economic and medicinal importance. Normally, Annona crassiflora is propagated through seeds. However, due to a deep dormancy that the seeds display at dispersion and the difficulty to obtain uniform plants in a short time period, micropropagation may be a feasible alternative. Concentrations of gibberellic acid (GA3) and naphthalene-acetic acid (NAA) and their interactive effects on in vitro seed germination and seedling development of Annona crassiflora were studied. Mature fruits of Annona crassiflora were depulped and the seeds washed in clear water and dried at room temperature. Seed coat was removed and the seeds were placed on Murashige & Skoog (MS) medium supplemented with gibberellic acid (GA3) and naphthalene-acetic acid (NAA), 30 g L-1 sucrose and 6 g L-1 agar-agar. Seeds were kept under these conditions for 30 days. After this period, seedlings were kept for another 90 days on Wood Plant Medium (WPM) with 20 g L-1 sucrose and 5 g L-1 agar-agar supplemented with the same GA3 and NAA concentrations. Cultures were incubated under controlled conditions at 25 ± 2°C temperature, 16: 8 (light: dark) photoperiod of 32 µmol m-2 s-1 irradiance provided by cool white fluorescent tubes (Philips). Use of WPM medium supplemented with 25-32 mg L-1 GA3 or MS with 26-30 mg L-1 GA3 and 2 mg L-1 NAA promoted rooting and plant growth.

The present study was planned to enable in vitro conservation of Vanda testacea, a highly medicinal orchid species through in vitro asymbiotic seed germination technique in Mitra orchid medium supplemented with cytokinins (Kn - 4.65 μM, BAP - 4.44 μM), and auxin (NAA- 5.37 μM). The germination frequency and initiation of germination was higher in NAA augmented medium and seedlings developed in 12.50 ± 0.50 weeks. Coconut water (20%) proved optimum for the multiplication of protocorm like bodies. Activated charcoal successfully checked the release of brownish exudates in the cultures. The influence of growth regulators, explants and their interactions on in vitro shoot bud formation from callus was studied in <I>Dianthus caryophyllus</I> L. The leaf and internode explants were cultured on Murashige and Skoog (MS) medium containing different concentrations of growth regulators. The highest callus induction was observed with 2 mg/l 2,4-dichlorophenoxy acetic acid (2,4-D) and 1 mg/l benzyl adenine (BA). Out of twenty seven shoot regeneration media tested, only 2 mg/l thidiazuron (TDZ) and zeatin alone or in combination with naphthalene acetic acid (NAA) and/or indole acetic acid (IAA) could differentiate calli. The highest average number of shoots was observed with 2 mg/l TDZ and 1 mg/l IAA. Significant differences were observed in calli producing shoots and number of shoots per callus in the explants of leaf and internode. The shoots were elongated and multiplied on MS medium supplemented with 1 mg/l BA and solidified with 1% agar. The shoots were rooted and hardened with 76% survival success in pots after six weeks of transfer to the pots.
