The prevalence of diabetes mellitus has reached epidemic proportion in Malaysia and worldwide. Scientific studies have shown that herbal plantPiper sarmentosumexhibits an antidiabetic property. Despite the extensive usage and studies of this herb as alternative medicine, there is paucity of the literature on the safety information of this plant. Thus, the present study aimed to observe the subacute toxic effects ofPiper sarmentosumaqueous extract (PSAE) on the haematological profile, liver, and kidney in rats. The extract was administered by oral gavage to 6 male and femaleSprague Dawleyrats in daily dose of 50 mg/kg, 300 mg/kg, and 2000 mg/kg for 28 consecutive days. The control group received normal saline. General behavior of the rats, adverse effects, and mortality were observed for 28 days. The haematological and biochemical parameters were determined at baseline and after the treatment. PSAE did not show abnormality on the body weight and gross observation of internal organs. The haematological, biochemical and histopathological profiles showed minimal changes and variation within normal clinical range except for significant increase in serum potassium level that suggests the need of regular monitoring. Nevertheless, these findings suggested that PSAE up to 2000 mg/kg/day did not show subacute toxicity inSprague Dawleyrats.

There is tremendous concern about the potential harmful effects of cell phone microwaves. The results are contradictory. However, the effects of these waves on the body have been reported. In this study, the effect of cellular microwaves on sperm parameters and total antioxidant capacity in relation to the duration of exposure and frequency of these waves were studied. This experimental study was conducted on male Sprague Dawley rats (150-200 g). The animals were randomly assigned to three groups (n = 18). Group 1 Natural control rat were fed to standard laboratory feeding Animals for 14 days. Group 2 animals were exposed to cell phone receiver stimulated waves (915 MHz frequency) for 14 days, and Group 3 animals were exposed to simulated waves of a cell phone antenna (950 MHz frequency) for 14 days. The results indicated that the total antioxidant capacity in all exposure groups decreased significantly compared with the control group (P <0.05). Increased exposure time within two weeks caused a statistically significant decrease in sperm motility and mobility (P <0.05).Effect of phone waves on liver and kidney of male Sprague Dawley rat. The study of pathological anatomy revealed a clear and appropriate deviation. These results found blood enzymes such as amino-aspartase transferase (AST), amino amines (ALT), and creatinine levels. The results showed no significant changes in both liver and kidney physiological functions. Malondialdehyde level (MDA) and total antioxidant contents were performed. The level of glutathione (GST), superoxide dismutase (SOD) was in normal ranges. Exposure to cell phone waves can reduce sperm movement and mobility in rats. These waves can also reduce the total antioxidant capacity in rats and lead to oxidative stress.

Mycotoxins have been formed in feed are secondary metabolites of fungi and are quite stable substances that have teratogenic, mutagenic and carcinogenic effects. An effective way to combat mycotoxins in feed is the use of feed additives that adsorb toxins, prevent their absorption in the gastrointestinal tract of the animal and are excreted from the body. The infl uence of coconut enterosorbent Shelltic Es on fattening and meat qualities of young pigs has been studied. It has been found in the process of fattening that the highest feed digestibility was in pigs of the experimental group, in the diet of which enterosorbent has been added. The results of studies of the chemical composition and technological properties of pork have been provided. The positive eff ect of enterosorbent on precocity, absolute average daily gain of live weight and reduction of feed expenditures per 1 kg of gain has been revealed. In pigs from the experimental group the average thickness of the fat was higher and the area of the “muscle eye” was lower compared with animals of the control group, which indicates that higher rates of carcass yield have been obtained due to faster accumulation of fat tissue. The weight of internal organs of pigs indicates the intensity of metabolic processes in the body. In experimental animals the weight of the lungs was 0,1 kg lower, and the liver and heart were higher by 0,13 and 0,01 kg, respectively. Enterosorbent had no effect on the content of vitamins in the liver of pigs of the compared groups. The content of impurities of organochlorine toxicants and toxic elements in the meat and liver of animals of the experimental group was lower than that of control analogues. Therefore, the use of enterosorbent Shelltic Es promotes more active excretion of them from the body.

Iron (IV) clathrochelate based on a macrobicyclic ligand of the hexahydrazide type is a unique compound that contains iron in a rare high valence IV. Preclinical and clinical studies of this complex, which were started for the first time in Ukraine, have an important theoretical and practical consequence as this complex can be recommended as an active substance in iron-containing drugs with antianemic action. In conducting preclinical studies of new drugs, pathomorphological studies are important because they are a necessary step in studying the biological response of animals to the action of test substances. It was found that some pathological changes develop in the body of white mice under conditions of experimental acute and chronic iron (IV) clathrochelate intoxication. They correlated with the dose of the test compound. During chronic intoxication, the microscopic changes in the liver and kidney of white mice treated with iron (IV) clathrochelate at a dose of 1/10 DL50 were similar to the microscopic changes in the liver and kidney of mice treated with the experimental drug at a dose of 1/5 DL50. However, the severity of these changes was lower, reflecting a lower degree of organ damage. In the myocardium of mice treated with iron (IV) clathrochelate at a dose of 1/5 DL50 on the 10th day, as in acute iron (IV) clathrochelate poisoning, only edema was recorded. The prospects for further research are the study of microscopic changes in the organs of laboratory animals of other species during experimental iron (IV) clathrochelate toxicosis.

There were infected neonatal piglets in the first days of their lives PED virus suspension derived from pigs previously PED patients. Diagnosis for PED in piglets donor virus PED was inserted complex method for clinical and epizootic performance and confirmed the identification PEDV by PCR-RT using the test system «EZ-RED/TGE/PDCoV MPX 1.0 Real time RT-PCR» company Tetracore (USA) Thermocyclers CFX 96 Real-Time System company BIO RAD (USA). Homogenate small intestine of pigs PEDV donor, prepared in a blender for PCR in a thick band of 18 animal carcasses, frozen at -18 °C without cryopreservation and kept 359 days. Before infecting pigs and strip defrost by RT-PCR identified the concentration of the virus genome equivalents (GE) without establishing viable virions quantitative pathogen. For Sample 20 selected analog neonatal piglets, divided them into 3 experimental groups (group 1 – 5 piglets, group 2 – 5 piglets and group 3 – 7 piglets) and one control (3 piglets). Research pigs infected per os virus-containing suspension with a concentration PEDV 1.03×106 GE/cm3. The dose for infection first group was 6 cm3 (6.18×106 GE/cm3), for the second – 5 cm3 (5,15 × 106 GE/cm3), for the third – 4 cm3 (4.12 GE×106/cm3) homogenate. The fourth group – control (not infected). All the pigs were in identical conditions that fully meet the physiological needs of the body. Of the 17 infected pigs only 2 was infected PEDV. PED was confirmed by laboratory methods. In bacteriological examination of internal organs of pigs that came out of a research experiment and control group were diagnosed colibacteriosis. In the control group was isolated from heart and intestinal non-pathogenic for white mice E. coli. From pigs 1 and 2 research groups has been allocated to white mice nonpathogenic E. coli, is set colibacteriosis; 2 experimental group found in one pig hemolytic E. coli; 3 experimental group from the internal organs of pigs in conjunction with non-pathogenic for mice intestinal former cane isolated Klesiella spp., is diagnosed with mixed infection (E. coli, Klesiella spp.). From the intestine of experimental and control pigs do not identified beneficial microflora – aerococcus, lactobacteria, bifidobacteria and cultured putrefactive anaerobic spore facultative and non spore microflora.

The purpose of the study was to compare the effect of feeding broiler chickens with mixtures containing raw or extruded soybean seeds on the performance indices, slaughter value and liver histology of the animals. The research began with an analysis of the chemical composition of selected high protein feeds. A nutritional experiment was then performed on 108 sexed Ross 308 chickens, which were allocated to 3 equal groups (I, II, III). The chickens were reared for 42 days with ad libitum feeding. The birds from the control group (I) received mixtures in which the only high protein component was soybean meal extracted. Extruded soybeans and raw soybeans were fed to chickens in experimental groups II and III, respectively, in quantities that replaced 30% of soybean meal extract protein in the Starter mixture and 50% of this protein in the Grower and Finisher mixtures. During the experiment, the body weight of the birds (BW) and their feed intake (FI) were recorded. At the end of rearing, 12 chicks of body weights representative of their group and sex were slaughtered in each group. The results of the slaughter analysis were used to calculate the percentage of muscles, skin with subcutaneous fat and abdominal fat in the carcass. The weights of selected internal organs were also determined, and then their proportion in the chilled carcass weight was determined. In addition, after slaughter, chicken livers were collected to assess the histological image. Raw soybean meal extrusion resulted in an increase in protein content and a decrease in crude fibre by about 10%. Quantities of individual amino acids in the analysed material were proportional to the protein content. The extrusion of full-fat soybean seeds reduced the content of trypsin inhibitors by more than a half (to 9 mg/g), but the least of these compounds (1.2 mg/g) was found in soybean meal extract. The tannin content of extruded soybeans was about one-third of that of extracted meal and less than one-fourth of that of raw soybean. It was shown that the chickens receiving extruded soybeans reached a weight similar to that of the control group, with similar FCRs for individual rearing periods, whereas the use of raw soybean significantly (P ≤ 0.01) decreased both parameters. In addition, group III had a significantly lower dressing percentage (P ≤ 0.05) and a higher proportion of abdominal fat (P ≤ 0.01) compared to the other groups. The introduction of extruded or raw soybeans into the mixtures increased (P ≤ 0.05) the proportion of total internal organs. The microscopic image revealed that all chickens had a similar, normal histological structure. The results of this study show a beneficial effect of raw soybean extrusion on the nutritional value of soybeans. Extruded soybeans can therefore be recommended as a partial substitute for protein (30% in Starter and 50% in Grower and Finisher) from soybean meal extract in chicken broiler mixtures. Raw soybeans should not be used in the above amounts, mainly because of the clearly worse rearing results....

In this study, we aimed to study the possible preventive effect of docosahexaenoic acid (DHA), a dietary omega-3 fatty acid, on toxicity caused by chlorpyrifos (CPF). Six groups of Sprague Dawley rats (200–250 g) consisting of equal numbers of males and females (n = 8) were assigned to study. The rats were orally given for 5 days. The control group was administered pure olive oil, which was the vehicle for CPF. The CPF challenge groups were administered oral physiological saline, pure olive oil, or DHA (50, 100 and 400 mg/kg dosages) for 5 days. The animals were weighed on the sixth day and then administered CPF (279 mg/kg, subcutaneously). The rats were weighed again 24 h following CPF administration. The body temperatures and locomotor activities of the rats were also measured. Blood samples, brain and liver tissues were collected for biochemical, histopathological and immunohistochemical examinations. A comparison with the control group demonstrated that CPF administration increased malondialdehyde (MDA) levels in blood, brain and liver, while it reduced catalase (CAT), superoxide dismutase (SOD) and glutathione peroxidase (GPx) concentrations ( p < 0.05–0.001). Advanced oxidation protein products (AOPPs) increased only in the brain ( p < 0.001). DHA reduced these changes in MDA and AOPP values ( p < 0.05–0.001), while it increased CAT, SOD and GPx concentrations ( p < 0.05–0.001). Similarly, DHA prevented the decreases in body weight, body temperature and locomotor activities caused by CPF at 100 mg/kg and 400 mg/kg dosages ( p < 0.05–0.001). Similar to the physiological and biochemical changes, the histopathological damage scores, which increased with CPF ( p < 0.05–0.01), decreased at all three dosages of DHA ( p < 0.05–0.01). Our findings suggest that DHA, by supporting the antioxidant mechanism, reduces toxicity caused by CPF.

Heavy metal toxicity contributes to liver and kidney dysfunction and damage through oxidative stress mechanisms; however, from previous studies, extracts from the Vernonia amygdalina plant have shown to possess potent antioxidant properties. This study was aimed at uncovering the potential ameliorative effects of ethanolic extract from Vernonia amygdalina plant in heavy metal toxicity-induced liver and kidney dysfunction. For this study, 44 Sprague Dawley rats were divided into three groups. The control group received a basal diet and water only while the treatment groups received varied dosages of the heavy metals. The copper (Cu) and lead (Pb) groups had five subgroups. The Cu only and Cu recovery subgroups were administered with 16 mg/kg Cu intraperitoneally daily for 14 days, whereas the Pb only and Pb recovery subgroups were administered with 13 mg/kg Pb intraperitoneally daily for 14 days. Subsequently, the Pb only and Cu only subgroups were sacrificed. The three Pb and Cu treatment subgroups received oral graded doses (100 mg/kg, 200 mg/kg, and 300 mg/kg) of the extract for 21 days. The Cu recovery and Pb recovery subgroups were left to recover for 21 days. After histological examinations, the Pb and Cu pretreatment groups showed evidence of focal necrosis accompanied by inflammatory cell infiltrations. The serum levels of liver biomarkers AST, ALT, and GGT, as well as urea and creatinine, were significantly elevated (P=0.01) following copper and lead exposure. Upon posttreatment of the rats with the extract, the physiological levels of the biomarkers were restored and tissue architecture of the organs improved. Thus, the ethanolic extract of Vernonia amygdalina is capable of ameliorating the effects of heavy metal toxicity through potent antioxidative mechanisms.

Lead acetate (300 mg/L) and/or cadmium chloride (50 mg/L) were administered as drinking water to Sprague-Dawley rats for 9 weeks to investigate the effects of concurrent exposure to lead and cadmium on the distribution patterns of five essential trace elements. Inductively coupled plasma mass spectrometry was used to determine the concentrations of zinc, copper, manganese, selenium and iron in the urine at different exposure times, as well as their levels in the renal cortex and serum at the end of treatment. Compared with the control group, exposure to lead and/or cadmium resulted in a significant increase in the urinary excretion of these five elements during the experiment, whereas significant decreased levels of these elements were found in kidney and serum. In conclusion, increased urinary loss of antioxidant trace elements due to lead and/or cadmium exposure induced the deficiency of antioxidants in the body, which could result in further oxidative damage. Moreover, there was an obvious synergistic effect of lead combined with cadmium on the distribution patterns of these essential trace elements, which may be related to the severity of co-exposure to these two metals.

This study was conducted to compare between two types of opportunistic fungi (Aspergillus fumigatus and Penicillium chrysogenum) in concerning their pathogenicity after intraperitoneal inoculation of mice. A total of twenty four male albino mice were used in this study which divided equally into 3 groups, The first and second groups were inoculated with 0.2ml of 1x 107spores/ml of A. fumigatus and P. chrysogenum  intraperitonially respectively, while the third group was inoculated with normal saline which served as control group. All animals were monitored for 2 weeks after infection. The blood samples were collected by heart puncture after 18 days post infection to isolate of serum that used for biochemical analysis of liver and kidney functions. After that, all animals were sacrificed. Some internal organs of infected groups (liver, kidney, intestine, heart, spleen and lung) were taken to study the histopathological changes. It was found that there was severe histopathological changes in studied organs of infected mice particularly liver, kidney, spleen and intestine which corresponding with significant variation (p<0.01) in enzyme activities of liver and kidney like (Alanine Aminotransferase (ALT), Urea and Creatinine). Also, It was found that P. chrysogenum had more impact on these enzymes (15.65 ± 0.78, 135.23 ± 8.75 and 0.928 ± 0.02 respectively) than A. fumigatus (21.70 ± 1.04, 57.91 ± 5.99 and 0.587 ± 0.03 respectively). Therefore, the present study indicated that fungi present in the environment can induce severe inflammation reach to tissue damage in most vital internal organs So, further studies should be performed to determine the specific virulence factors and active components, which are responsible for pathogenesis of A. fumigatus and P. chrysogenum in spite of the fact that P. chrysogenum can produce antibiotic.

Our previous studies have demonstrated that naturally occurring peptide, Nisin possess antibacterial activity and did not interfere with rabbit vaginal mucosa. In this study, the reproductive toxicity of the Nisin in male rats was evaluated. Rats were fed orally with Nisin (10, 25, and 50 mg/kg/day) for 13 weeks. No treatment related mortality was observed. The body weight gain, food consumption and serum biochemical parameters were at par with the control group. Histomorphology of the selected reproductive (testis, epididymis, ventral prostate, and seminal vesicle) and nonreproductive (liver and kidney) tissues was observed to be normal. There was no treatment-related increase or decrease in the expression of testis-specific genes (c-Kit, GATA-1, and HILS-1) and the activity levels of epididymal α-glucosidase, ventral prostate alkaline phosphatase (AlP), liver alanine aminotransferase (AlAT) and aspartate aminotransferase (AAT). Fructose and lactic acid levels in the seminal vesicles also remained unchanged. These studies suggest that Nisin did not affect the normal physiology of these organs. In addition, no adverse effects were observed on the reproductive performance of Nisin-treated male rats and their offspring. In conclusion, the current studies support our earlier studies, which demonstrated suitability of Nisin as a safe and effective microbicide.
