The oxidation and antioxidant status of serum are often determined in serum samples which have been frozen for some time. The oxidative stress process is prone to fast alterations in the sample because of the possible instability of the reactants. Here one oxidation assay (ROM) and three antioxidant assays (FRAP, TAS, and BAP) have been tested on their performance and stability at short-time storage. The most commonly used temperatures for storage and handling of serum samples (+4 and +20°C) were selected. In three short-term studies in which the storage time varied between 3 and 48 hrs the performance of these assays were tested on human serum samples. The general conclusion is that most assays performed well and gave stable results during 2 days of storage of the samples at both temperatures. Only the FRAP and TAS assays showed a small deviation at some storage conditions. In conclusion, handling of serum samples at +4 and +20°C during short-time periods did not affect the quality and performance of the oxidation and antioxidant assays during day-to-day analyses.

The present work aims at accessing the stability of biological material stored for diagnostic and scientific purposes. The influence of the temperature, storage time, and cyclic thawing on concentration stability of selected oxidative stress parameters in human serum was investigated. The study group consisted of 20 serum samples collected from healthy volunteers aged 18–52. The parameters whose reference ranges were not determined and to which validated determination methods did not correspond were examined by manual methods (FRAP and AOPP). Automatic methods were used to determine routine laboratory tests (albumin, total protein, bilirubin, uric acid) using the Konelab 20i® analyzer. The samples were stored at various temperatures (room temperature, 4 °C, −20 °C, −80 °C) for max 6 months and were subjected to cyclic thawing at 1 month intervals. In order to check whether any differences between the concentrations of the studied parameters existed when the samples were stored in various conditions, the paired Student t-test or Wilcoxon test and comparison to desirable bias were applied. Based on the obtained results, it was found that the temperature and time of serum sample storage significantly affected the stability of the analyzed parameters and determined different shelf lives of serum samples for oxidative stress examination. Therefore, continuing the investigation concerning the impact of storage conditions on various serum parameters seems justified due to the discrepancy between the individual results obtained by different researchers and the inconsistencies between the results of scientific research and the applicable recommendations.

AbstractAmino acids analysis or metabonomics requires abundant serum/plasma samples collection and samples storage has become inevitable given the limited capacity for immediate analysis. Currently, most of the existing studies on metabolites stability during sample storage focused on long-term and short-term stability, while many functional amino acids might be ignored due to the poor sensitivity and detection of analysis methods. Here, we attempted to elucidate the stability of amino acids and related amines as comprehensive as possible in human serum following different preprocessing and pre-storage procedures. Pooled, fasting serum samples were collected and stored at 4 °C and 22 °C respectively after a delay in sample processing (0, 1, 2, 4, 8, 12 and 24 hours) and underwent freeze-thaw cycles for three times at −80 °C. The concentration of amino acids and related amines were quantified using isobaric tagging reagent iTRAQ®-LC-MS/MS. Approximately 54.84 %, 58.06 % and 48.39 % of detectable and target analytes altered at 4 °C and 22 °C during pre-treatment and freeze-thaw cycles. Some amino acids which are not stable and relatively stable were found. Our study provided detailed profiles and suggestions for amino acids in human serum corresponding to diverse collection and pre-treatment measures.

Abstract Environmental or metabolic heat can increase oxidative stress in sows and may impact suckling pig growth; yeast cell- and oil-based products are used to alleviate oxidative stress. A study was conducted to assess yeast cell and mint oil supplementation in sow lactation diets on sow anti-oxidant status and offspring performance to weaning. A total of 45 sows and gilts (240.7 ± 38.5 kg BW) were assigned to one of 4 diets: Control diet (CON), control + yeast cell at 0.2% (YC), control + mint oil at 10 ppm (MO), and control + yeast cell and mint oil (YCMO). Diets were provided from d 110 of gestation through weaning (21 ± 4 d). Variables evaluated were: sow and piglet growth, litter characteristics at birth, colostrum and milk oxidative status, sow feed intake. Data analyzed as randomized complete block with sow as experimental unit and Tukey’s adjustment as means separation test. There was no effect of maternal diet on sow BW (204.52 ± 43.5 kg) at weaning, piglet birth weight (1.44 ± 0.96 kg), litter size (13.09 ± 6 pig/sow) or sow feed intake (6.32 ± 3.05 kg). Piglets from CON females had lesser gain (P < 0.05) compared with the other groups during the suckling phase (0.23, 0.24, 0.24, and 0.24 ± 0.005 kg in CON, YC, MO, and YCMO group, respectively). Superoxide dismutase activity in sow serum (d110 and weaning), colostrum, and milk was not different among diets and was 39, 59, 45, and 10% lower at weaning than d 110 in CON, YC, MO, and YCMO sows, respectively. Milk glutathione content tended to be lower (P < 0.10) in MO than YCMO sows with CON and YC intermediate (15.2, 32.0, 20.6, and 21.1 U/ml), respectively. Overall, inclusion of yeast cell or mint oil enhanced piglet growth, which may be related to lower sow antioxidant status.

Oxidative stress and inflammation are implicated in obesity. Therefore, we investigated whether moderate and short-term calorie restriction (CR) reflects a real-life situation, mediates weight loss, and improves oxidative stress markers. We analyzed oxidative stress markers in patients with obesity undergoing moderate CR. Serum oxidative stress markers (myeloperoxidase (MPO), superoxide dismutase (SOD), catalase, total antioxidant status (TAS), and reactive oxygen species (ROS) (generation by endothelial cells in vitro)) were measured in 53 subjects (mean BMI 37.8 ± 5.9 kg/m2) who underwent 8 weeks of CR, which included a reduction of 300–500 kcal/day. MPO was the most CR-sensitive parameter. The mean level of serum MPO in patients with obesity was 20% higher than that in post CR intervention (p < 0.001). SOD increased by 12% after CR (p < 0.05), which was largely due to the improvement in glucose tolerance and the reduction in insulin resistance after CR. Other tested parameters were not modified during the treatment. CR resulted in an expected decrease in body weight (by 5.9 ± 4.6 kg, p < 0.0001) and other anthropometric parameters. Additionally, it was accompanied by a significant change in hsCRP, hsTNF alpha, hsIL-6, leptin (all p < 0.0001), and HOMA-IR (p < 0.05). Cardiovascular and metabolic parameters were also partially improved. Short-term, moderate CR partially improves antioxidant capacity but is enough to substantially change anthropometric parameters in obese patients. Our observations indicate that mimicking real-life situations and low-cost dietary intervention can be successfully implemented in obesity treatment with a simultaneous moderate effect on antioxidant status.

Tacrolimus (TAC) is an immunosuppressive drug, optimally used for liver, kidney, and heart transplant to avoid immune rejection. In retrospect, a multitude of studies have reported effects of TAC, such as nephrotoxicity, diabetes, and other complications. However, limited information is available regarding short-term exposure of TAC on the liver. Therefore, the present study was designed to unravel the effects of short-term exposure of TAC on a rat model. The animal model was established by TAC administration for 6, 12, 24, and 48 h time points. Liver histopathological changes were observed with PAS-D, reticulin stain, and immunostaining of PCNA and CK-7 coupled with glycogen quantification in a liver homogenate. TUNEL assay was performed to evaluate the DNA damage in the liver. Concentration of GSH and activities of SOD and CAT in the serum were measured to assess the antioxidant status, whereas liver tissue MDA level was measured as a biomarker of oxidative stress. Hepatic gene expression analysis of IL-10, IL-13, SOCS-2, and SOCS-3 was performed by RT-PCR. Results revealed marked changes in liver architecture of all TAC-treated groups, as evidenced by sinusoid dilation, hepatocyte derangement, glycogen deposition, and collapsed reticulin fibers. Significant increase in PCNA and CK-7 immunostaining along with the presence of TUNEL-positive cells was revealed in treatment groups as compared to the control group. Serum antioxidant enzyme status was markedly decreased, whereas the liver MDA level was increased in TAC treatment groups indicating oxidative stress induction. The gene expression profile of cytokines was significantly upregulated in treatment groups highlighting an inflammatory response. In conclusion, results of the current study propose that even a short-term TAC exposure can induce change in antioxidant status and lipid peroxidation. Therefore, these factors should be considered to avoid and minimize immunosuppression-related issues in a prolonged course of treatment.

Abstract. Background: The pathophysiological basis of chronic kidney disease and its complications, including cardiovascular disease, are associated with chronic inflammation and oxidative stress. We investigated the effects of active vitamin D (calcitriol) and synthetic vitamin D analog (paricalcitol) on oxidative stress in hemodialysis patients. Methods: This cross-sectional study was composed of 83 patients with a minimum hemodialysis vintage of one year. Patients with a history of any infection, malignancy, and chronic inflammatory disease were excluded. Oxidative markers (total oxidant and antioxidant status) and inflammation markers (C-reactive protein and interleukin-6) were analyzed. Results: A total of 47% (39/83) patients were using active or analog vitamin D. Total antioxidant status was significantly higher in patients with using active or analog vitamin D than those who did not use (p = 0.006). Whereas, total oxidant status and oxidative stress index were significantly higher in patients with not using vitamin D when compared with the patients who were using vitamin D preparation (p = 0.005 and p = 0.004, respectively). On the other hand, total antioxidant status, total oxidant status, and oxidative stress index were similar between patients who used active vitamin D or vitamin D analog (p = 0.6; p = 0.4 and p = 0.7, respectively). Conclusion: The use of active or selective vitamin D analog in these patients decreases total oxidant status and increases total antioxidant status. Also, paricalcitol is as effective as calcitriol in decreasing total oxidant status and increasing total antioxidant status in patients with chronic kidney disease.

Objective: To study the effect of short-term use of oral contraceptive pills on intra-ocular pressures of women of childbearing age.   Methods: It was a comparative observational study, conducted at Arif memorial teaching hospital and Allied hospital Faisalabad for a period of six months. Hundred female subjects were divided into two groups of 50 each. Group A, included females, who had been taking oral contraceptive pills (OCP) for more than 6 months and less than 36 months. Group B, included 50 age-matched controls, who had never used OCP. Ophthalmic and systemic history was taken. Careful Slit lamp examination was performed and intraocular pressures (IOP) were measured using Goldman Applanation tonometer. Fundus examination was done to rule out any posterior segment disease. After collection of data, we analyzed and compared the intra ocular pressures between the two groups by using ANOVA in SPSS version 21.   Results: Average duration of using OCP was 14.9 months. There was no significant difference of Cup to Disc ratios between the two groups (p= 0.109). However, significant difference was noted between the IOP of OCP group and controls. (p=0.000). Conclusion: OCP significantly increase IOP even when used for short time period.
