Königinnen von 12 Taxa der Untergattung Bombus sensu stricto (Bombus affinis, B. albocinctus, B. cryptarum, B. lucorum, B. magnus, B. moderatus, B. occidentalis, B. patagiatus, B. sporadicus, B. terrestris, B. terricola and B. tunicatus) wurden im Frühjahr gefangen, um künstliche Kolonien zu züchten. Zusätzlich wurden Männchen von B. franklini und B. lucorum/China gefangen. Mitochondriale Cytochrome Oxidase Untereinheit I (COI) von 53 Proben wurde sequenziert (Teilsequenzen der Länge 1257 bp). GenBank Sequenzen der Ostasiatischen Arten B. hypocrita, B. ignitus und B. lucorum/China wurden einbezogen. Der Unterschied zwischen den Arten beträgt 30 – 92 Basen-Substitutionen und die Tamura-Nei genetische Distanz 0.030-0.103, während der Unterschied innerhalb der Arten nur 1 – 3 Basensubstitution beträgt und die Tamura-Nei genetische Distanz nur 0.001-0.003. Dreizehn Taxa mit Artrang sowie eine neue Art B. minshanicola Bischoff 1936 (= B. terrestris ssp. minshanicola = B. lucorum/China) wurden nachgewiesen. In der Topologie des phylogenetischen Stammbaums gibt es 4 Cluster: ein Artenpaar B. sporadicus – B. ignitus und ein Artenpaar B. terricola – B. occidentalis, ein Cluster für B. magnus, B. patagiatus and B. cryptarum mit den Subspecies B. cryptarum albocinctus und B. cryptarum moderatus sowie ein Cluster für B. hypocrita, B. lucorum, B. minshanicola, B. franklini and B. affinis. Die Arten B. terrestris mit den Subspecies B. terrestris canariensis und B. terrestris sassaricus sowie B. tunicatus sind einzeln abgetrennt. Da sich im Alignment der COI Sequenzen keine Lücken finden, können die einzelnen Nucleotid-Positionen als homolog betrachtet werden. Jede Art besitzt eine Reihe einzigartiger (‘privater’) Positionen, die als diagnostische Merkmale benutzt werden können um die Art zu definieren und zu identifizieren.

Königinnen der vier Taxa Bombus affinis, B. moderatus, B. occidentalis and B. terricola wurden an verschiedenen Orten quer durch Nordamerika gefangen. Zusätzlich wurden Männchen von B. franklini, B. occidentalis und B. terricola gesammelt. Mitochondriale Cytochrome Oxidase Untereinheit I (COI) von 25 Proben wurde sequenziert (Teilsequenzen 1005 bp Länge). Die Divergenz der Sequenzen zwischen den Taxa beträgt 30–50 Basen-Substitutionen und die Tamura-Nei Genetische Distanz 0.05–0.13, während innerhalb der Taxa die Divergenz nur 1 bis 2 Basen-Substitutionen beträgt und die Tamura-Nei Genetische Distanz 0.001–0.002. Da die COI Sequenzen keine Lücken aufweisen, können die einzelnen Nukleotide wie homologe Positionen verwendet werden. Jedes Taxon besitzt 8–20 eigene Substitutionen, die als dia­gnostische Positionen verwendet werden können, um das Taxon zu charakterisieren. Das Phylogramm zeigt drei klar getrennte Cluster: B. moderatus, konspezifisch mit der ostasiatischen B. albocinctus, das Artenpaar B. affinis – B. franklini locker verwandt mit der eurasiatischen B. lucorum, und das Artenpaar B. terricola – B. occidentalis ohne Beziehungen zu irgendwelchen Arten der Alten Welt.

Species of Psithyrus (Hymenoptera: Apidae) are obligate social parasites of bumblebees. They range from monospecificity to species which have many Bombus hosts. Much of the reported variation in host–parasite behaviour which occurs during attempted nest usurpations may be explained in the context of host specificity. Psithyrus ashtoni is a relatively specialized bumblebee social parasite. The ability of P. ashtoni females to successfully enter laboratory colonies of Bombus affinis and Bombus terricola was tested. Female Psithyrus were introduced into more than 100 host colonies containing 0–58 workers. Parasites and host queens never fought, but in both hosts, invasion success decreased with increasing worker numbers. The passive nest usurpation behaviour of P. ashtoni differs from that of other, nonspecialized species of Psithyrus, which may kill or displace the host queen, and from that of completely specialized parasites, which may cohabit with both host queens and host workers. Host specialization may allow Psithyrus females to evolve behavioural and physiological features which minimize host defenses. Thus, while the number of available host species is limited, there is an increased probability of successful usurpation in nests of the appropriate host.

1. Substrate cycling of fructose 6-phosphate through reactions catalysed by phosphofructokinase and fructose diphosphatase was estimated in bumble-bee (Bombus affinis) flight muscle in vivo. 2. Estimations of substrate cycling of fructose 6-phosphate and of glycolysis were made from the equilibrium value of the 3H/14C ratio in glucose 6-phosphate as well as the rate of 3H release to water after the metabolism of [5-3H,U-14C]glucose. 3. In flight, the metabolism of glucose proceeded exclusively through glycolysis (20.4μmol/min per g fresh wt.) and there was no evidence for substrate cycling. 4. In the resting bumble-bee exposed to low temperatures (5°C), the pattern of glucose metabolism in the flight muscle was altered so that substrate cycling was high (10.4μmol/min per g fresh wt.) and glycolysis was decreased (5.8μmol/min per g fresh wt.). 5. The rate of substrate cycling in the resting bumble-bee flight muscle was inversely related to the ambient temperature, since at 27°, 21° and 5°C the rates of substrate cycling were 0, 0.48 and 10.4μmol/min per g fresh wt. respectively. 6. Calcium ions inhibited fructose diphosphatase of the bumble-bee flight muscle at concentrations that were without effect on phosphofructokinase. The inhibition was reversed by the presence of a Ca2+-chelating compound. It is proposed that the rate of fructose 6-phosphate substrate cycling could be regulated by changes in the sarcoplasmic Ca2+ concentration associated with the contractile process.

Genetic variability at eight enzyme loci was assessed in five bumble bee species from southern Ontario: Bombus affinis Cresson, B. fervidus (Fabricius), B. bimaculatus Cresson, B. impatiens Cresson, and B. vagans Smith. Average expected heterozygosity (Hexp ± SE) across all species was 0.089 ± 0.023 (range 0.046–0.139). These values are higher than any previously reported estimates of average expected heterozygosities for bumble bees.
