SummaryLysolecithin is adsorbed to washed blood platelets and, at sufficient concentration, lyses them, inhibits their clot-retracting activity and promotes their thromboplastin-generating activity. Lysolecithin adsorption to the platelet was studied by using P32-labelled lysolecithin obtained from the liver of rats injected with labelled orthophosphate. The amount of lysolecithin adsorbed to the surface of the washed platelet in saline medium is dependent on the concentration of lysolecithin in solution and reaches saturation — 5 × 10-8 jig per platelet — at a concentration of 9—10 µg per ml. Platelet lysis in saline medium begins at a lysolecithin concentration higher than 18 jig per ml. Plasma and albumin prevent adsorption of lysolecithin to the platelet and protect the platelet from damage by lysolecithin. Albumin is able to remove previously adsorbed lysolecithin from the platelet surface. The protective action of plasma explains the lack of platelet damage in blood, the plasma lecithin of which has been converted to lysolecithin by the action of Vipera palestinae venom phosphatidase, in vitro and in vivo.

The aim of this work is to study the effects of hydrogen absorption on mechanical properties of pipe API 5L X70 steel. This study is conducted in special soil solution NS4 with pH 6.7 It show that the tensile properties like yield stress, ultimate strength and elongation at failure reduced under hydrogen embrittlement. Several fatigue tests (three (03) points bending tests) on roman tile specimens with notch are performed. Fatigue initiation is detected by acoustic emission. A comparison between specimens electrolytically charged with hydrogen and specimens without hydrogen absorption is made and it has been noted that fatigue initiation time is reduced when hydrogen embrittlement occurs. The field of elastoplastic stresses near the notch is computed by the finite-element method with the Abaqus software package. Effective distance and stress are calculated with the volumetric approach and the Notch intensity Factor of the roman tile specimen is determined for each loading value used in our tests.

An important problem in embryology is whether the differentiation of cells depends upon a stable restriction of the genetic information contained in their nuclei. The technique of nuclear transplantation has shown to what extent the nuclei of differentiating cells can promote the formation of different cell types (e.g. King & Briggs, 1956; Gurdon, 1960c). Yet no experiments have so far been published on the transplantation of nuclei from fully differentiated normal cells. This is partly because it is difficult to obtain meaningful results from such experiments. The small amount of cytoplasm in differentiated cells renders their nuclei susceptible to damage through exposure to the saline medium, and this makes it difficult to assess the significance of the abnormalities resulting from their transplantation. It is, however, very desirable to know the developmental capacity of such nuclei, since any nuclear changes which are necessarily involved in cellular differentiation must have already taken place in cells of this kind.

Volume-osmotic pressure relationships at equilibrium have been obtained in chick heart fibroblasts grown in slide-coverslip cultures in a fluid medium consisting of heparinized plasma and embryo extract. The refractive index of the fibroblast gives a direct measure of its solid concentration, and the volume is estimated as the reciprocal of concentration. The volume is found to be linearly related to the reciprocal of the osmotic pressure over a range from 130 to 587 m-osm, provided the measurements are carried out rapidly at 38°C. The isotonic water content of the cells derived from the gradient of the regression line on the basis of the simple Boyle-van’t Hoff Law was found to be less than actual water content obtained by direct refractometry, i. e. the value of Ponder’s ℛ was 0⋅94 (s. d. 0⋅04). In cultures grown in a simple saline medium and measured at 22°C the volume was related linearly to the reciprocal of the osmotic pressure only between the limits of 330 and 191 m-osm. Outside these limits the volume was greater than expected and this was attributed to alterations in the semi-permeable properties of the cell membrane. The value of Ponder’s ℛ in these cultures was 1⋅15. The importance of the quantity, ℛ, as applied to cells other than the erythrocyte, is indicated. The value, 0⋅94 (s. d. 0⋅04), obtained in fibroblasts under physiological conditions is not explicable on the basis of the probable osmotic properties in vitro of the cell proteins. The discrepancy is within the experimental error, but it may also be due to abnormal osmotic behaviour of the cell proteins resulting from some form of intermolecular structure in the cytoplasm.

The paper addresses the development of advanced welding technologies with two and three solid wires for joining of HSLA API-5l X70 (High-strength low-alloy) steel plates with thickness of 19.1 mm. The experiments were performed using a multi-wire Submerged Arc Welding (SAW) system that was developed for welding of steels with solid, tubular and cold wires, in different combinations. The main goal of the research was to assess the mechanical performances of the welded joints achieved by multi-wire SAW technology and then to compare them with the single wire variant, as reference system. The welded samples were firstly subjected to NDT control by examinations with liquid penetrant, magnetic particle, ultrasonic and gamma radiation, with the aim of detecting the specimens with flaws and afterwards to reconsider and redesign the corresponding Welding Procedure Specifications (WPS). The defect-free welded samples were subjected to tensile, Charpy V-notch impact and bending testing in order to analyse and report the mechanical behaviour of API-5l X70 steel during multi-wire SAW process. The experimental results were processed and comparatively discussed. The challenge of the investigation was to find the appropriate welding technology which responds simultaneously to the criteria of quality and productivity. Further research on metallurgical behaviour of the base material will be developed, in order to conclude the complete image of the SAW process effects and to understand how the multi-wire technologies affect the mechanical and metallurgical characteristics of the API-5L X70 steel used in pipelines fabrication.

Purpose – This paper aims to clarify the corrosion behavior of two famous structural steels in sour environment. These steels have a vast application in oil and gas industries. The study aims to find the effect of different concentrations of sour solution on the origin of crack in these steels. Design/methodology/approach – After preparation of specimens, different sour solutions were made using the synthetic brine (according to National Association of Corrosion Engineers [NACE], Technical Committee Report 1D182) and various amounts of Na2S.9H2O and CH3COOH. The polarization test was done by Potansiostat apparatus model Zahner-IM6 at two temperatures, 25°C and 50°C. The corrosion current densities were obtained from the polarization curves. Finally, the corrosion products and hydrogen-induced cracking (HIC) were investigated by Tescan Vega II XMU scanning electron microscope (SEM) linked to a Rontec energy-dispersive X-ray spectroscopy (EDS) system. Findings – API 5L-X70 steel showed smaller corrosion current values than A516-Gr70 steel. The HIC cracks propagated parallel to the surface of A516-Gr70 steel in three solutions and confirmed the inappropriateness of this steel for sour environment applications. Originality/value – This paper studies the effect of sour environment on the behavior of two famous industrial steels at two temperatures by new method.
