Pathology of five species of Gyrodactylus Nordmann, 1832 (Monogenea)

Attachment-site pathology of the ectoparasites Gyrodactylus adspersi Cone and Wiles, 1983 on Tautogolabrus adspersus, Gyrodactylus avalonia Hanek and Threlfall, 1969 on Gasterosteus aculeatus, Gyrodactylus bullatarudis Turnbull, 1956 on Poecilia reticulata, Gyrodactylus sp. on Carassius auratus, and Gyrodactylus salmonis (Yin and Sproston, 1948) on Salmo gairdneri was examined using optical microscopy and scanning electron microscopy. In the first four species, the disc-shaped haptor attached itself to the host skin surface by means of blade tips of 16 peripherally located marginal hooks, with the two ventrally located hamulus blades compressing the underlying epithelium into a small depression. Tissue damage from attachment and feeding was minimal. However, G. salmonis lodged its marginal hook sickles deep into the host epidermis and appeared to cause extensive fin damage and skin discoloration. There was little evidence of bacterial involvement in the wounds, but bacterial microcolonies were observed on the body surface and haptor of G. salmonis. The latter results are discussed in relation to our understanding of gyrodactyliasis.

Gyrodactylus colemanensis Mizelle and Kritsky, 1967 and Gyrodactylus salmonis (Yin and Sproston, 1948) parasitized Salvelinus fontinalis, Salmo gairdneri, and Salmo salar at a fish farm in Nova Scotia. Gyrodactylus colemanensis occurs on the edges of fins and clings delicately to the skin surface. Gyrodactylus salmonis embeds its marginal hooks deeply into the epidermis and occurs all over the body surface except on the fin edges. The parasites gain entrance to the hatchery on infected stocks arriving from other facilities and possibly through infected wild salmonids entering via the water supply. Brood stocks serve as reservoir hosts within the farm. Young-of-the-year S. fontinalis became infected within 8 weeks of transfer to the outside raceways, with hatchery buckets and nets likely serving as vehicles of transmission. Intensity of infection increased during winter to a spring peak followed by a decrease during the summer months. Intensity generally decreased with host age. Stocks were initially infected with mixed infections; subsequently, single species infections occurred which involved first G. salmonis, then G. colemanensis, and then G. salmonis. There were no clinical signs of disease associated with infections and the parasites did not harbor bacterial or viral pathogens.

Between 1985 and 1989, 133 of 136 (98%) bowfin (Amia calva) collected from Lakes Ontario, Erie and Huron, Ontario, Canada, and from Sugg's Lake, Florida, U.S.A., were infected with a new species of trypanosome. Trypanosoma phaleri n.sp. was pleomorphic, developed to infective metatrypanosomes in the leech Desserobdella phalera, and was transmitted by D. phalera to laboratory-reared bowfin but not to Catostomus commersoni, Anguilla rostrata, Ictalurus nebulosus, Gasterosteus aculeatus, Salmo gairdneri, Perca flavescens, Stizostedion vitreum, or Carassius auratus. Trypanosoma phaleri n.sp. developed poorly in the leech Hemiclepsis marginata.

Abstract Human-induced growth of macro-algae is often assumed to increase trematode infections in fishes by increasing the abundance and condition of the parasite’s intermediate host – snails – as this can boost the release of trematode larvae, cercariae, from the intermediate hosts. However, macro-algae can also impose barriers to the transmission of cercariae and reduce infections. We investigated whether an increased growth of filamentous algae affects the transmission of Diplostomum pseudospathaceum cercariae to the threespine stickleback Gasterosteus aculeatus, a common fish in eutrophied shallow waters. We exposed sticklebacks to trematode cercariae in the absence and presence of artificial filamentous algae, and recorded effects on the proportion of sticklebacks infected and the number of encysted metacercariae per fish. No significant effect of artificial algae on cercariae transmission was detected. However, the body size and the sex of the sticklebacks were strongly correlated with the number of encysted metacercariae per infected fish, with females and larger individuals being more infected. We discuss different factors that could have caused the difference in parasite transmission, including sex-related differences in body size and behaviour of sticklebacks.

We tested the hypothesis that birds can rapidly change the conductance of water vapor at the skin surface in response to a changing need for evaporative heat loss. Mourning doves (Zenaida macroura) were placed in a two-compartment chamber separating the head from the rest of the body. The rate of cutaneous evaporation was measured in response to dry ventilatory inflow at three ambient temperatures and in response to vapor-saturated ventilatory inflow at two ambient temperatures. At 35 degrees C, cutaneous evaporation increased by 72 % when evaporative water loss from the mouth was prevented, but no increase was observed at 45 degrees C. For both dry and vapor-saturated treatments, cutaneous evaporation increased significantly with increased ambient temperature. Changes in skin temperature made only a minor contribution to any observed increase in cutaneous evaporation. This indicates that Z. macroura can effect rapid adjustment of evaporative conductance at the skin in response to acute change in thermoregulatory demand.

The rectal temperature of normal healthy camels at rest may vary from about 34°C to more than 40°C. Diurnal variations in the winter are usually in the order of 2°C. In summer the diurnal variations in the camel deprived of drinking water may exceed 6°C, but in animals with free access to water the variations are similar to those found in the winter. The variations in temperature are of great significance in water conservation in two ways. a) The increase in body temperature means that heat is stored in the body instead of being dissipated by evaporation of water. At night the excess heat can be given off without expenditure of water. b) The high body temperature means that heat gain from the hot environment is reduced because the temperature gradient is reduced. The effect of the increased body temperature on heat gain from the environment has been calculated from data on water expenditure. These calculations show that under the given conditions the variations in body temperature effect a considerable economy of water expenditure. The evaporative heat regulation in the camel seems to rest exclusively on evaporation from the skin surface (sweating), and there is no apparent increase in respiratory rate or panting connected with heat regulation. The evaporation from isolated skin areas increases linearly with increased heat load. The critical temperature at which the increase sets in is around 35°C. The fur of the camel is an efficient barrier against heat gain from the environment. Water expenditure is increased in camels that have been shorn.

Larvae of the lamprey, Entosphenus lamottei (Le Sueur), and rainbow trout, Salmo gairdneri Richardson, were exposed to the sodium salt of 3-trifluormethyl-4-nitrophenol (TFM) in concentrations of 0.75 p.p.m., 3.00 p.p.m., and 6.00 p.p.m. Microscopic examination of changes induced in the gills, liver, cloacal region, and musculature were made on 7-micron sections stained with Harris' haematoxylin and Bowie's eosin, and in the gill region with periodic acid Schiff reagent. A comparison of the degree of the effects in the two species was made by planimetry of the vascular, cellular, and edematous areas from enlarged drawings of sections.Upon exposure to lethal concentrations of TFM, the body of the larval lamprey becomes distended at the pharyngeal level and heavy cords of mucus emerge from the external gill clefts. A deep red coloration is evident in the pharyngeal region consequent upon vasodilatation of the arterioles and capillaries of the gill filaments. Trout exhibit a similar vasodilatation of the gills together with increased mucous secretion. Edema in the connective tissue between the respiratory epithelium and the vascular endothelium is induced in both species. After prolonged exposure to TFM, the mucous cells in the lining of the branchial chamber and covering the tips of the gill filaments are actively discharging their secretions or completely spent.Certain effects induced by TFM in the larval lamprey are not evident in the trout. The cloacal region takes on a deep red coloration due to dilatation of the venous sinuses and the liver becomes reddish because of sinusoidal dilatation. Extensive edema of the fibrous connective tissue of the skeletal musculature is characteristically present. A slightly increased secretory activity of mucous-secreting cells may occur in the epidermis.With the techniques employed in this study, there was no evidence in either species of cytological or histological changes in the nervous tissue, cardiac musculature, notochord, alimentary canal (including the haemopoietic typhlosole of the lamprey), or mesonephros.

ABSTRACT The sterilization chamber is designed to sterilize the body using a filler formulation that is not harmful to the skin surface. Formulation, namely the composition of filler fluids (type, dose and amount) using substances that are safe for the body accordingly, certainly provides benefits. The use of the sterilization booth uses the correct and safe filling fluid. The sterilization chamber uses a mist nozzle sprayer (with the appropriate discharge) to spray the filling liquid so that the user is safe. This sterilization booth is named "White Box". This "White Box" research aims to design (functional and structural) and test (function test and performance test) on a system and mechanism for condensing the condensation of a humidifier that can fill the sterilization room optimally. The design method of the "White Box" sterilization booth uses a qualitative method with the following phases: (1) The preparation stage, namely the functional design and the structural design of the "White Box"; (2) The implementation stage is analyzing the use of tools and materials and making sterilization booths; (3) Post-implementation stage, namely testing the sterilization chamber, including the function test and performance test of the sterilization chamber. The design activity of the "White Box" sterilization booth was carried out by testing various variations of the prototype, including variations in the dimensions of the distribution pipe, the shape of the distribution pipe, and variations of the piping system. The validation process includes: (1) The diameter of the pipe against humidifier dew bursts; (2) The shape of the piping against humidifier dew spray; and (3) the length of the pipe passage to the humidifier dew spray. ABSTRAKBilik sterilisasi didesain untuk mensterilisasi tubuh dengan menggunakan formulasi zat pengisi yang tidak berbahaya bagi permukaan kulit. Formulasi yaitu komposisi cairan pengisi (jenis, takaran dan jumlah) menggunakan zat yang aman bagi tubuh yang sesuai, tentu memberi manfaat. Penggunaan bilik sterilisasi menggunakan cairan pengisi yang benar dan aman. Bilik sterilisasi menggunakan mist nozzle sprayer (yang debitnya sesuai) untuk menyemprotkan cairan pengisinya sehingga pengguna aman. Bilik sterilisasi ini diberi nama "White Box". Penelitian "White Box"ini bertujuan merancang (fungsional dan struktural) dan menguji coba (uji fungsi dan uji kinerja) pada sebuah sistem dan mekanisme penyaluran pengembunan dari alat humidifier yang mampu memenuhi ruang bilik sterilisasi secara optimal. Metode rancang bangun bilik sterilisasi "White Box" menggunakan metode kualitatif dengan tahap kegiatan meliputi: (1) Tahap persiapan yaitu rancangan fungsional dan rancangan struktural"White Box"; (2) Tahap pelaksanaan yaitu analisa penggunaan alat dan bahan dan Pembuatan bilik sterilisasi; (3) Tahap Pasca Pelaksanaan yaitu pengujian bilik sterilisasi antara lain uji fungsi dan uji kinerja bilik sterilisasi. Muatan kegiatan rancang bangun bilik sterilisasi "White Box" dilakukan dengan cara menguji coba berbagai variasi prototipe, meliputi variasi dimensi pipa penyaluran, bentuk pipa penyalur, variasi sistem perpipaan. Proses validasi yang dilakukan meliputi: (1) Diamater pipa terhadap semburan embun humidifier; (2) Bentuk pipa penyalur terhadap semburan embun humidifier; dan (3) Panjang lintasan pipa terhadap semburan embun humidifier.

Tetrahymena pyriformis has been firstly isolated from brood Goldfish(Carassius auratus) Common, Fantail, Ryukin, Veiltail and Moor, Black molly(Poecilia sphenops), Gourami (Trichogaster trichopterus), red Swordtail(Xiphophorus helleri), Guppy (Poecilia reticulata), deep Angelfish(Pterophyllum scalare) and brood Algae eater (Hypostomus plecostomus) inIraq. The infected fish suffered from mucus hypersecretion large hyperemicareas on the skin, and slight shedding of the scales. The incidence percentage ofinfection was 100 %. Rising water temperature was up to 29 – 30cْ for one timeand methylene blue was at a concentration of 5 ppm for 24 hours. Sodiumchloride at a concentration of 2 % for one minute and two minutes has noobvious effect on Tetrahymena pyriformis. Treatment with NaCl at theconcentration of 1% for 20 minutes for three consecutive days achieved the bestresults.

OBJECTIVE: To report a case of cephalexin-induced Stevens-Johnson syndrome (SJS), a devastating adverse drug reaction that involves the entire skin surface and mucosal areas of the body. DATA SOURCES: MEDLINE search (key terms cephalosporins, Stevens-Johnson syndrome, erythema multiforme, and systemic lupus erythematosus) and references identified from bibliographies of pertinent articles. DATA SYNTHESIS: Clinical presentation and manifestations of SJS include the skin, eyes, gastrointestinal tract, and pulmonary system. Infectious complications are the leading cause of mortality. Early intervention is important to prevent progression of SJS. The case described is consistent with the features of this syndrome. The patient presented with fever, arthralgias, and malaise. Skin lesions included a diffuse violet macular rash with erythema and multiple bullous lesions on her neck and abdomen. The skin biopsy was consistent with SJS. Multiple mucocutaneous ulcers were found in her mouth, but no evidence of lower gastrointestinal tract involvement was documented. She remained relatively free of pulmonary complaints except for the presenting bronchitis. CONCLUSIONS: Cephalexin should be added to the list of agents to consider as iatrogenic causes of SJS.

A technique has been elaborated that enabled the plerocercoid larvae of Schistocephalus solidus to be removed from the body cavity of Gasterosteus aculeatus without bacterial contamination. Larvae were cultured in plugged test-tubes under completely aseptic conditions in a variety of balanced salines, glucose salines and nutrient peptone broth. The most successful results were obtained with peptone broth at room temperatures (16-19° C) in which plerocercoids remained active and showed normal behaviour for periods up to 300 days. In ¾ strength Locke's solution, which was found by experiment to be approximately isotonic with Schistocephalus (δ = -0.44 ± 0.02° C), the mean period of normal behaviour was 114 days. In the remaining saline and saline-glucose media, the mean viability and period of normal behaviour was considerably less. In the plerocercoid, histological examination revealed that the genitalia are in an immature condition. During cultivation at room temperatures, the genitalia remained in this undifferentiated condition and showed no signs of undergoing spermatogenesis, oogenesis or vitellogenesis. Plerocercoids were induced to develop into sexually mature adults by raising the temperature of cultivation in peptone broth to 40° C. (i.e. the body temperature of the final host in the natural life cycle). Oviposition took place after 48-60 hr. at this temperature, and histological examination revealed that spermatogenesis, oogenesis, vitellogenesis and shell formation had taken place in a normal manner. The viability of artificially matured Schistocephalus was 4-6 days in vitro--a period equivalent to the viability of the adult in vivo. The eversion of the cirris was observed in each proglottid after 40 hr. cultivation at 40° C. During the sexual process the cirris everted and invaginated at the rate of about once per second. Cross-fertilization between segments of the same worm or with segments of another worm was not observed. Except for one specimen in ¾ strength Locke's solution which underwent spermatogenesis and partial vitellogenesis, larvae cultured in salines or glucose salines at 40° C. died within 1-3 days without further development. Attempts to hatch out the eggs produced by the cultivation of larvae in peptone broth at 40° C. proved unsuccessful. Histological examination revealed that spermatozoa had not been taken into the vagina. It was concluded that the eggs were not fertilized owing to the failure of normal copulation to take place.
