Moderate body hypothermia alleviates behavioral and EEG manifestations of audiogenic seizures in metaphit-treated rats

We investigated the effects of hypothermia on the incidence and EEG signs of audiogenic seizures in rats treated with metaphit (1-[1(3isothiocyanatophenyl)-cyclohexyl] piperidine), an experimental model of generalized reflex epilepsy. After i.p. injection with metaphit (10 mg/kg) Wistar rats were exposed to audiogenic stimulation at hourly intervals during the time course of the experiment. After intermittent use of an ice pack 8 h after the metaphit treatment, when seizure was fully developed, the body temperature was reduced to 30 ± 0.5 °C in one half of the rats, and maintained at 37 ± 0.5 °C in the other half. Saline-injected rats served as a control group. In the hypothermia group, the incidence of audiogenic seizures induced by metaphit was completely suppressed during the 3 consecutive testing times, while no signs of epileptiform activity were noted in EEG tracings. The termination of hypothermic treatment resulted in the recovery of seizure susceptibility, and during audiogenic stimulation, bursts of spiking activity were recorded in the EEGs of metaphit-treated rats. These findings indicate that moderate body hypothermia is an effective anticonvulsant treatment for audiogenic seizures in metaphit-treated rats.

The aim of this study was to investigate the clinical efficacy of vanadium complexes on triglycerides (TG), total cholesterol (Chol), uric acid (UA), urea (U), and antioxidant parameters: nonenzymatic (FRAP—ferric reducing ability of plasma, and reduced glutathione—GSH) and enzymatic (glutathione peroxidase—GPx, catalase—CAT, and GPx/CAT ratio) activity in the plasma of healthy male Wistar rats. Three vanadium complexes: [VO(bpy)2]SO4⋅2H2O, [VO(4,4′Me2bpy)2]SO4⋅2H2O, and Na[VO(O2)2(bpy)]⋅8H2O are administered by gavage during 5 weeks in two different diets such as control (C) and high fatty (F) diets. Changes of biochemical and antioxidants parameters are measured in plasma. All three vanadium complexes statistically decrease the body mass growth in comparison to the control and fatty diet. In plasma GSH was statistically increased in all vanadium complexes-treated rats from control and fatty group in comparison to only control group. Calculated GPX/CAT ratio was the highest in the control group in comparison to others.

The aim of this study was to evaluate the influence of ligature-induced periodontal disease in pregnant rats on their newborn's health parameters. Twenty-four female adult Wistar rats were divided into two groups: the control group (G1) and the group that was submitted to dental ligatures around second upper molars (G2). After the four week period of development of periodontitis, the female animals were mated with male adult Wistar rats. There were no differences in the body weight of females between the two groups during mating and pregnancy. No differences were observed among the groups in relation to the viable newborn index. However, there were differences in newborn birth weight, explained by the diverse size of the litters. In this study, ligature-induced periodontal disease did not promote changes during pregnancy that resulted in low birth weight in newborn Wistar rats.

The aim of this research was to investigate the effects of extract ethanol of pegagan (Centella asiatica) after treated with restraint stress by measuring the eight arms radial maze performance of rats. Pre-test and post-test controlled groups design was applied in this research. Male Wistar rats (Rattus novergicus) with the mean age of 8 weeks and the body weight of 150-200 grams which used for sample were divided randomly into 4 groups, each group consisted of 6 rats, i.e. (i) control group (without any treatment), (ii) stress group (it was given by restraint stress for 2 hours/day for each rat), (iii) pegagan group (it was given by 0.3 mg/g BW/day/rat extract ethanol of pegagan), and (iv) pegagan and stress group (it was given by 0.3 mg/g BW/day/rat extract ethanol of pegagan and restraint stress for 2 hours/day for each rat). The treatments were given for 21 days. Within 12 days for each pre-treatment and post-treatment, a test on the eight arm radial maze was conducted on individual rat to observe its performance. The assessment of rat performance in the eight arms radial maze test was conducted based on error type B. Kruskal-Wallis and Mann-Whitney tests with SPSS for Windows 16 version were applied to analyze statistically the difference between four groups. Kruskal-Wallis test was used to show the significant performance level difference between four groups of rats with p=0.001, while Mann-Whitney test was used to determine the significant difference between stress group and pegagan group (p=0.001), also stress group and pegagan and stress group (p=0.001). The result of research showed that there was no significant difference between control group and stress group (p=0.051), control group and pegagan group (p=0.143), control group and pegagan and stress group (p=0.143), also pegagan group and pegagan and stress group (p=0.952). It was concluded that extract ethanol of pegagan improved the performances of rats on the eight arms radial maze after treated with restraint stress.

Objective: Potash known as potassium carbonate (K2CO3) is a mixture of salt with other components, including impurities which coexist in mineral and salt is highly consumed in various forms by pregnant women. The aim of this research is to determine the effect of potash on the weight index of pregnant Wistar rats. Methods: A total of 25 albino Wistar rat with weights ranging from 180 to 300 g were used and allocated into five groups of five animals each (four females and one male) designated as Groups A, B, C, D, and E. The experimental Groups B, C, D, and E were administered through oral route different doses of potash of 300 mg/kg, 600 mg/kg, 900 mg/kg, and 1200 mg/kg, respectively, after pregnancy was detected by checking for mucus plug in the vagina. Group A served as the control group and was administered distilled water only. The animals were allowed for 1 week for acclimatization under normal temperature (270–300°C), which they were being fed with normal feed (grower’s mash) and water ad libitum for 1 week. Results: The result showed a significant (p˂0.05) reduction in weight with the highest level seen with the 1200 mg/kg group when compared to the control. Conclusion: Therefore, the effect of potash alters the physical activity and decreases weight, by implication may induce growth retardation of the Wistar rats which is not healthy for a pregnant animal.

PURPOSE: Thermal injury causes catabolic processes as the body attempts to repair the damaged area. This study evaluated the effects of a scald injury on the morphology of muscle fibers belonging to a muscle distant from the lesion. METHODS: Thirty Wistar rats were divided into control (C) and scalded (S) groups. Group S was scalded over 45% of the body surface, standardized by body weight. Rats in both groups were euthanized at four, seven and 14 days following the injury. The middle portions of the medial gastrocnemius muscles were sectioned, stained with hematoxylin and eosin and Picrosirius, and submitted to histological analysis. RESULTS: Control group sections exhibited equidistantly distributed polygonal muscle fibers with peripheral nuclei, characteristic of normal muscle. The injured group sections did not consistently show these characteristics; many fibers in these sections exhibited a rounded contour, variable stain intensities, and greater interfiber distances. A substantially increased amount of connective tissue was also observed on the injured group sections. CONCLUSION: This experimental model found a morphological change in muscle distant from the site of thermal injury covering 45% of the body surface.

Abstract: Liver is the largest organ in the abdominal cavity. As the center of metabolism in the body, liver is potentially damaged by exposure of toxic substances, inter alia carbon tetrachloride (CCl4). Metabolism of carbon tetrachloride (CCl4) produces CCl3 free radicals that can damage the liver. In Indonesia, there are a lot of natural ingredients that have antioxidant properties, such as tomato. Lycopene in tomatoes contains antioxidant compounds that can prevent damages due to free radical. This study aimed to obtain liver histopathological changes of wistar rats fed with tomato juice after being induced of carbon tetrachloride (CCl4). This was an experimental study, using 10 wistar rats which were divided into 4 groups. Group I was the negative control; group II was induced with CCl4 0,05 cc/day and was terminated on day 6; group III was induced with CCl4 0,05 cc/day and was given tomato juice 3 ml/day, and terminated on day 13; group IV was induced by CCl4 0,05 cc/day, given regular pellets, and terminated on day 13. The results showed that group II had histopathological changes of the liver indicating fatty liver, meanwhile group III showed regeneration of nearly all liver cells. Conclusion: Administration of tomato juice after the induction of 3 ml carbon tetrachloride (CCl4) for 7 day showed regeneration of almost all liver cells. Keywords: histopathological changes of the liver, carbon tetrachloride, tomato juice.   Abstrak: Hati merupakan organ terbesar dalam rongga abdomen, dan pusat metabolisme tubuh dengan fungsi yang sangat kompleks dan sangat berpotensi mengalami kerusakan akibat terpapar oleh bahan-bahan toksik, salah satunya yaitu karbon tertraklorida (CCL4). Metabolisme CCl4 menghasilkan radikal bebas CCl3 yang dapat merusak hati. Di Indonesia terdapat  banyak sekali bahan-bahan alami yang mempunyai kandungan antioksidan, salah satunya yaitu tomat. Tomat mengandung senyawa likopen sebagai antioksidan yang dapat mencegah kerusakan jaringan akibat radikal bebas. Penelitian ini bertujuan untuk mendapatkan gambaran histopatologi hati tikus wistar yang diberi jus tomat pasca induksi karbon tetraklorida (CCl4). Metode penelitian ialah eksperimental. Sampel sebanyak 10 ekor tikus wistar yang dibagi dalam 4 kelompok. Kelompok I sebagai kontrol negatif; kelompok II diinduksi CCl4 0,05cc/perhari dan diterminasi hari ke-6; kelompok III diinduksi CCl4 0,05 cc/hari kemudian diberikan jus tomat 3ml/hari;  dan kelompok IV diinduksi CCl4 0,05 cc/hari kemudian diberikan pelet biasa dan diterminasi hari ke-13. Hasil penelitian menunjukkan pada pemberian CCl4 pada tikus wistar selama 5 hari terdapat gambaran morfologik perlemakan sel hati. Pemberian jus tomat dosis 3 ml pasca induksi karbon tetraklorida (CCl4) menunjukkan terjadinya regenerasi pada hampir seluruh sel-sel hati. Simpulan: Pemberian jus tomat dosis 3 ml pasca induksi karbon tetraklorida (CCl4) selama 7 hari menunjukkan regenerasi pada hampir seluruh sel-sel hati. Kata kunci: gambaran histopatologi hati, karbon tetraklorida, jus tomat.

Introduction: Goko cleanser is one of the popular herbal mixtures used by the Nigerian populace. This study was carried out to evaluate the effects of this herbal mixture on some haematological parameters of adult female Wistar rats. Methodology: A total of twenty five Wistar rats weighing about 160 – 280g were divided into five groups according to their corresponding weights. The groups were designated as groups 1 – 5. Group 1 served as the control group, while groups 2, 3, 4, and 5 received 1000mg/Kg, 1500mg/Kg, 2000mg/Kg and 4000mg/Kg respectively. Results: The TWBC, PCV, PLATELET count, and RBC and Hb showed a statistically significant (p<0.05) increase for the test groups 2 – 5 when compared to the control group. The body weight had a statistically significant (p<0.05) decrease in groups 2, 4, and 5; while group 3 had a statistically insignificant (p>0.05) decrease. Conclusion and Recommendation: This study revealed that Goko Cleanser contains some haemopoietic benefits, but inhibit appetite centres. Hence, this study recommends that further study be carried out on the effects of this herbal mixture on the brain so as to reveal the extent of its effect on the appetite centres.

This study aims to determine the effects of open and closed formulated standard diets supplied from different sources on growth performance and internal organ development of laboratory rats. Five-week-old 32 Wistar rats were used. A special control group diet was produced in accordance with the criteria determined by the National Research Council (NRC) (1995). Three different most preferred commercial open and closed-formula diets produced by international and local companies were used as trial groups’ diets. The experiment was carried out for 12 weeks. Weekly feed consumption, body weight change, internal organ weight, intestinal organ weigths and lengths, intestinal villi heigth and crypt depth were measured in groups. The body weight values of the control group and the first group fed with open-formula diet were found at the highest level (P <0.05). The control group diet had a positive effect on small intestine villi heigth and crypt depth (P <0.05). The nutrient contents and energy values of the diets of experimental groups were determined as different from the commercial firm notifications. As a result of the research, it is concluded that the diets prepared with open-formula give more reliable results in the growth performance and development of internal organs of Wistar rats.

The general method for synthesis the η6-(arene)-triscarbonyl-chromium(0) complexes was modified and applied for preparation of η6-(anisole)-triscarbonyl-chromium(0) and the study of its analgesic activity was undertaken. A significant analgesic activity was observed after intraperitoneal injection, in Wistar rats. Two doses (30 and 50 mg/Kg of the body weight) of η6-(anisole)- triscarbonyl-chromium(0) were injected and the analgesic activity was evaluated by the Hot Plate Test method. They showed a significant analgesic effect in comparison with the control group and the group treated with dipyrone standard, but not so high when compared with the group treated with morphine standard. Overall, it was observed that the η6-(anisole)- triscarbonyl-chromium(0) was easily obtained by the modified synthetic method and was effective in increasing the pain threshold.

The increasing number of tobacco users will directly impact to an increase in the number of passive smokers. Various harmful substances that enter the respiratory tract can cause health problems. The airway will recognize these substances as pathogen and activate immune response accordingly. Inflammatory response becomes one of the effects of body compensation due to an increased pathogen. This effect is indicated by the elevation of C-Reactive Protein as one systemic inflammatory response parameters in the body. This study aimed to determine the efficacy of red mulberry juice to reduce levels of C-Reactive Protein in Wistar rats because exposure to cigarette smoke. This is an experimental study with a post-test control group design. Animals were divided into negative control group, positive control group, and 3 treatment groups with different red mulberry doses. The positive control group showed the highest increase   in C-Reactive Protein levels compared with the red mulberry intake group (p<0,001). In conclusion, the administration of red mulberry juice can reduce inflammatory response in rats because of exposure to cigarette smoke.
