Linkage of allozyme loci in Japanese red pine (Pinusdensiflora)

Linkage relationships among 29 polymorphic loci were investigated in Japanese red pine, Pinusdensiflora Sieb. et Zucc. Haploid megagametophytes from 12 highly heterozygous plus-tree clones were used. A total of 217 pairs of two-locus combinations were available for the linkage study. Chi-square and Akaike Information Criterion analyses identified 19 pairs of linkages. Close linkages (r = 0–0.10) were seen for Est-2, 3–Aap-2, 3, Dia-1–Acp, and Shd-1–Got-1. Strong linkages (r = 0.10–0.20) were seen for Est-2, 3–Dia-1, Est-2, 3–Acp, Aap-2, 3–Dia-1, Got-2–Lap-1, and Pgm–Aap-1. Moderate linkages (r = 0.20–0.30) were seen for Adh–Sod, Adh–Pgi-1, 2, Adh–Lap-1, G2d–G6p, and Dia-2–Amy pair of linkages. The remaining six pair combinations, namely 6Pg-1–Est-2, 3, 6Pg-1–Acp, Est-1–Acp, Est-2, 3–Shd-1, Sod–Pgi-1, 2, and Adh–Got-2 were weakly linked (r = 0.30–0.40). In all, 22 loci were classified into five linkage groups by means of three-point mapping methods.

The available U. S. Cucumis sativus germplasm collection (754 Plant Introductions) was electrophoretically screened for genetic diversity using 39 enzymes representing a total of 57 loci. Polymorphisms were observed at 18 loci which included g2dh, gpi1, gpi2, gr1, gr2, idh, mdh1, mdh2, mdh3, mpi2, pep-la2, pep-pap2, per4, pgd1, pgd2, pgm1, pgm3, and skdh. Appropriate crosses were set up to verify the inheritance of and test linkages among these loci. Four allozyme linkage groups have currently been identified. Representative linkages and their genetic distances include: gpi1 - mdh3 (20); pgm1 - pgd1 (25); and g2dh - pgd2 (19). Additionally, crosses were made to marker stocks to test for linkages between some allozyme loci and loci coding for resistance to downy mildew and anthracnose, long hypocotyl, divided leaf, short petiole, glabrous, compact plant, determinate, little leaf, and bitter free (bi).

Megagametophyte tissue from seeds of 35 clones of Pinusstrobus L. were subjected to horizontal starch gel electrophoresis. The resulting gels were tested for activity of 10 enzyme systems. Isozymes observed were under control of at least 17 loci. Direct evidence, based on segregation of allozymes in megagametophytes of heterozygous clones, is presented for nine loci in five systems (LAP, GPI, GOT, PGM, and AP). Three polymorphic loci were detected for LAP. No genetic variation was detected for IDH, ADH, GLY-3-PD, GDH, or 6 PDG.Five pairs of linked loci were detected by chi-square analyses of joint segregation. Twenty-five of 36 possible two-locus combinations among 18 clones were evaluated. Recombination estimates suggest relatively strong linkage between LAP1-LAP2 and GPI2-GOT1; weaker linkage between LAP2-GPI2 (based on one clone only); and weak linkage between LAP2-AP1 and GOT3-AP1.

We tested the constancy of linkage relationships of randomly amplified polymorphic DNA (RAPD) marker loci used to construct a population-based consensus map in material from an Italian stand of Picea abies (L.) Karst. in 29 individuals from three Norwegian populations. Thirteen marker loci linked in the Italian stand did show a consistent locus ordering in the Norwegian population. The remaining 16 unlinked marker loci were spread over different linkage groups and (or) too far apart both in the population map and in this study. The limited validity of RAPD markers as genomic "hallmarks" resilient across populations is discussed. We also investigated the reliability of RAPD markers; only 58% of the RAPD markers previously used to construct the consensus map in the Italian population were repeatable in the same material. Of the repeatable ones 76.3% were amplified and found polymorphic in 29 megagametophyte sibships from three Norwegian populations.
