Production of deoxynivalenol and related compounds in liquid culture by Fusarium graminearum

A liquid culture method for the production of deoxynivalenol and related compounds by Fusarium graminearum was developed. Major factors which stimulate the biosynthesis of these compounds include reduced oxygen levels, depletion of carbohydrate in the medium, pH, and possibly a low concentration of an organic nitrogen source. Isolates of F. graminearum were tested for the yields of four trichothecene mycotoxins and zearalenone in this system. The time course of acetyl deoxynivalenol, deoxynivalenol, and zearalenone in the fermentation was measured over a 21-day period against pH, glucose concentration, protein, fungal biomass, and ergosterol. A new ester of deoxynivalenol, 15-acetyl-deoxynivalenol, is reported from North American isolates of F. graminearum.

Paratuberculosis is a chronic disease of ruminants caused by Mycobacterium avium subsp. Paratuberculosis (MAP). Since isolation of MAP type I (S) is rarely reported in Italy, our research was aimed at isolating, by an inexpensive liquid culture manual method, this type of MAP isolates. At first, we used an ELISA to point out to serologically positive samples from five flocks. Secondly, we used a fecal direct IS900-qPCR on the ELISA positive samples, in order to detect shedder animals. Feces from IS900-qPCR positive samples were inoculated in solid and liquid culture media. IS900-qPCR was further used to test the growth of MAP isolates in liquid medium, which were further confirmed by f57-qPCR and submitted to typing by specific PCR in order to identify the MAP type. Twenty-eight samples (24 fecal and four tissutal samples) were processed by culture methods, resulting in the isolation of six type I MAP field isolates. Notably, no isolates were recovered by solid media, underlining the utility of this liquid method. Few data about this type of MAP are currently available in Italy, and further analyses should be carried out in order to study the origin and epidemiology of type I strains circulating in Italy.

The recent study has demonstrated the effects of different nitrogen sources on vanillin production by Phanerochaete chrysosporium. Primary screening supported maximum biotransformation of ferulic acid (from lemongrass leaves hydrolysate) to vanillin by using ammonium chloride and yeast extract as inorganic and organic nitrogen source, respectively. With the 2-level factorial analysis, the optimum conditions of vanillin production from ferulic acid by P. chrysosporium was achieved at 0.192g/L with a molar yield of 24.5%.

In this study, the antagonistic effect of five bacteria genera namely Bacillus, Pseudomonas, Azotobacter, Azospirillum, and Streptomyces isolated from rhizosphere of wheat were evaluated against Fusarium graminearum as potential biocontrol agents in vitro. F. graminearum was molecularly diagnosed using the Polymerase chain reaction (PCR) technique. Each bacteria were tested for the production of catalase enzyme, oxidase enzyme, analysis of starch, analyze of gelatin, and the motility, where Azotobacter, Azospirillum, and Bacillus subtilis were positive for all tested. Fungal inhibition tests were performed by using the dual culture method and agar well diffusion technique. Among them, Streptomyces and Azospirillum exhibited potent inhibition to the growth of F. graminearum (72.14% and 66.42%) respectively, followed by B.pumillus, P.fluorescens, B. subtilis and Azotobacter ( 58.28%, 43.23%, 39.71% and 35.71%) respectively as compared with the control treatment (0.0%).The dry weight of the fungus biomass was decreased with bacteria P. fluorescens, Streptomyces sp, Azotobacter sp, Azospirillum sp, B. subtilis, and B. pumillus which reached (0.114, 0.103, 0.147, 0.101, 0.143, and 0.107 g) respectively compared to the control treatment that was 0. 665 g.

When symbiotic coelenterates, especially hermatypic corals, were incubated in the light in sea water containing NaH 14 CO 3 , small quantities of fixed 14 C were released from the tissues at a steady rate over 4 h. The rate of release was greatly increased in the presence of glycerol, glucose and alanine; the additional 14 C released was in the same substance as that added to the medium. The following related compounds had little or no effect on 14 C release : ethylene glycol, sorbose, fructose, glucosamine, glycine, proline, serine and glutamic acid. Such results have been previously reported in other symbiotic systems, and the substances causing the specific release of fixed 14 C are believed to be those which move from the autotrophic to the heterotrophic symbiont. This belief is supported here by previous observations that glycerol, glucose and alanine are among the most important organic substances released by freshly isolated zooxanthellae. Ammonium chloride increased the amount of fixed 14 C released by corals into alanine media, possibly due to conversion of ammonia to amino acids by zooxanthellae. Appreciable release of 14 C fixed in the dark also occurred into alanine solutions. These results suggest possible roles of zooxanthellae in supplying organic nitrogen compounds to the host cell at night as well as during the day. The involvement of zooxanthellae in ‘recycling’ nitrogen compounds within the association may help to explain the success of corals in seas poor in nutrients. There was substantial utilization of external glycerol and glucose when supplied at either high or low concentrations. Corals may well be able to utilize some of the small amounts of organic matter dissolved in sea water in the natural environment.

Detection and enumeration of Legionella in water samples is of great importance for risk assessment analysis. The plate culture method is the gold standard, but has received several well-known criticisms, which have induced researchers to develop alternative methods. The purpose of this study was to compare Legionella counts obtained by the analysis of potable water samples through the plate culture method and through the IDEXX liquid culture Legiolert method. Legionella plate culture, according to ISO 11731:1998, was performed using 1 L of water. Legiolert was performed using both the 10 mL and 100 mL Legiolert protocols. Overall, 123 potable water samples were analyzed. Thirty-seven (30%) of them, positive for L. pneumophila, serogroups 1 or 2–14 by plate culture, were used for comparison with the Legiolert results. The Legiolert 10 mL test detected 34 positive samples (27.6%) and the Legiolert 100 mL test detected 37 positive samples, 27.6% and 30% respectively, out of the total samples analyzed. No significant difference was found between either the Legiolert 10 mL and Legiolert 100 mL vs. the plate culture (p = 0.9 and p = 0.3, respectively) or between the Legiolert 10 mL and Legiolert 100 mL tests (p = 0.83). This study confirms the reliability of the IDEXX Legiolert test for Legionella pneumophila detection and enumeration, as already shown in similar studies. Like the plate culture method, the Legiolert assay is also suitable for obtaining isolates for typing purposes, relevant for epidemiological investigations.

ABSTRACT Proteins are a main organic nitrogen source for microorganisms. Many heterotrophic microorganisms secrete extracellular proteases (ex-proteases) to efficiently decompose proteins into oligopeptides and amino acids when exterior proteins are required for growth. These ex-proteases not only play important roles in microbial nutrient acquisition or host infection but also contribute greatly to the global recycling of carbon and nitrogen. Moreover, may microbial ex-proteases have important applications in industrial, medical, and biotechnological areas. Therefore, uncovering the mechanisms by which microorganisms initiate the expression of ex-protease genes in response to exterior proteins is of great significance. In this review, the progress made in understanding the induction mechanisms of microbial ex-proteases in response to exterior proteins is summarized, with a focus on the inducer molecules, membrane sensors, and downstream pathways. Problems to be solved for better understanding of the induction mechanisms of microbial ex-proteases are also discussed.

Yeast extract (YE) is a sole organic nitrogen source in a commonly medium 3G for Streptomyces albulus to produce poly-ε-lysine (ε-PL). In this study, five YEs from five commercial suppliers were used to evaluate their effects on the growth of Streptomyces albulus 213 and ε-PL production. YE from bakers’ yeast with the highest total nitrogen content (TN), α-amino nitrogen content (AN) or the ratio of AN/TN produced the highest yield of ε-PL, while YE from brewers’ yeast with the highest RNA content got the most dry cell weight (DCW). However, there was little correlation between TN, AN or AN/TN in YE and the yield of ε-PL. There was also weak correlation between RNA in YE and DCW. When 60% baker YE was mixed with 40% brewer YE, the yield of ε-PL reached 1.89 g/l and increased by 16% and 37% than those of baker YE and brewer YE, respectively. Furthermore, five kinds of vitamins and nine kinds of amino acids were screened to supplement baker YE. When 6 mg/l biotin, 5mg/l pantothenic acid, 0.7 g/l L-glutamine and 0.5 g/l L-arginine were mixed with baker YE in M3G, ε-PL reached maximum production respondent to 3.05 g/l that was 61.4% higher than control.

Lysobacter antibioticus HS124 inhibited mycelial growth of Fusarium graminearum (74.66%) under the dual culture method. Microscopic investigation clearly showed that amendment with different concentrations (10%, 30%, and 50%) of HS124 bacterial culture filtrate on potato dextrose agar plates caused abnormal hyphal structures, including swelling and distortion. Its inhibition toward mycelial growth of F. graminearum was increased with increasing concentration of n-butanol crude extract of HS124. The highest inhibition (43.14%) was detected at a crude concentration of 10 mg/disc, whereas the lowest inhibition (21.57%) was observed at 2 mg/disc. Although mycelial growth of F. graminearum was promoted by volatile organic compounds (VOCs) produced by HS124 as compared with the control, these VOCs clearly decreased fungal pigmentation resulting in a reduction of fungal sporulation. Microscopic investigation revealed hyphal deformation of F. graminearum due to VOCs. These compounds also had a negative effect on spore germination of F. graminearum. In vivo evaluations demonstrated that HS124 inoculation of wheat plants reduced crown rot disease incidence by 73.70% as compared with the control. HS124 inoculation of wheat plants also promoted most of the growth characteristics compared with the control or fungicide-treated plants. Our results provide strong evidence that HS124 could control F. graminearum infections and promote growth of wheat plants as part of management strategies for crown rot disease.
