Accelerated detection of lymphocytic choriomeningitis virus in diagnostic specimens

Lymphocytic choriomeningitis virus could be demonstrated earlier in mice inoculated with clinical specimens if the mice were sacrificed before they appeared sick for examination by the fluorescent antibody technique. In the procedure, the optimal route for inoculation was intracerebral and the best organ tested for staining was the brain.

The antigens of the Egypt virus have been detected by means of the fluorescent antibody technique in the neurons of the brain and spinal cord, in both sensory and motor areas, of infected mice. The antigens have also been observed in infected human epidermoid carcinoma cells in culture. In both cases the antigens occurred in the cytoplasm of the cells exclusively. In the early stages of infection the antigen was localized about the nucleus of the human cultured cells. In some cases the staining of the antigens produced a sponge-like effect, probably because of the numerous vacuoles of lipid material. The antigens were observed in the protoplasmic connections between cells and they probably served as a main route of spread of the agent in these human cell cultures. The number of cells initially infected and showing antigen was proportional to the amount of virus added, and titration with one substrain of cells proved much more sensitive than intracerebral titration in the mouse.

A total of 200 beta-hemolytic streptococci, isolated from clinical specimens submitted to our laboratory, were identified as group A versus non-A using the fluorescent antibody technique (FA), bacitracin susceptibility (BBL, Difco, and Raven disks), SeroSTAT, Streptex, Phadebact, and the API 20S system. Of the 122 group A isolates, all methods except SeroSTAT and Phadebact yielded 92–99% agreement when compared with the Lancefield precipitin test. Phadebact yielded an 84% agreement and SeroSTAT changed from 83 to 98% after trypsinization. Numerous false positives were obtained and only FA (91%) and API 20S (96%) yielded better than 90% agreement on non-A identification when compared with the Lancefield test. The most false positives were obtained (45%) using the SeroSTAT reagents. Considering accuracy, our data suggests the FA technique to be the method of choice for identifying group A streptococci.

Abstract Between December 2016 and April 2017, a spate of abortions occurred in a closed dairy herd from the central eastern region of Paraná, Brazil, in which 75 cows aborted. To identify its cause, organ fragments were collected from an aborted fetus for histopathology, and the blood samples from a stillborn, 4 aborted fetuses, and 9 farm dogs for indirect fluorescent antibody technique (IFAT). These tests found multifocal non-suppurative encephalitis, periportal hepatitis, and multifocal lymphoplasmacytic myocarditis, and detected anti-Neospora antibodies in all aborted fetuses, and in 5 of the 9 dogs. DNA of Neospora caninum was detected in the brain tissue of an aborted fetus. Blood samples of 340 cows and 146 heifers showed 33.5% and 30.8% seropositivity, respectively. In this closed herd, the parasite was probably introduced by infected domesticated or wild carnivores inhabiting the farm, through the infective oocysts present in their stool.

Fluorescent antibody technique by the monoclonal antibody method is very useful and helpful for the rapid quantification and in situ detection of the specific bacteria like nitrifiers in a mixed baxterial habitat such as a biofilm. In this study, twelve monoclonal antibodies against Nitrosomonas europaea (IFO14298) and sixteen against Nitrobacter winogradskyi (IFO14297) were raised from splenocytes of mice (BALB/c). It was found that these antibodies exhibited little cross reactivity against various kinds of heterotrophic bacteria. The direct cell count method using monoclonal antibodies could exactly detect and rapidly quantify N. europaea and N. winogradskyi. Moreover, the distribution of N. europaea and N. winogradskyi in a biofilm could be examined by in situ fluorescent antibody technique. It was shown that most of N. winogradskyi existed near the surface part and most of N. europaea existed at the inner part of the polyethylene glycol (PEG) gel pellet, which had entrapped activated sludge and used in a landfill leachate treatment reactor. It was suggested that this monoclonal antibody method was utilized for estimating and controlling the population of nitrifying bacteria as a quick and favorable tool.

Fluorescent labeled antibodies were used for the demonstration of uromucoid. This urine specific mucoprotein is demonstrably present only in the epithelial cells of the proximal segments of the normal human renal tubules and in the matrix of human kidney stones of all the common crystalline compositions.
