Growth of psychrophilic microorganisms at subzero temperatures

In an attempt to find a suitable medium for determining the ability of psychrophilic bacteria and yeasts to grow at subzero temperatures, six antifreeze compounds were added individually and in combination to Trypticase soy broth and agar. The media were tested for their ability to support good growth of psychrophilic bacteria at 20 °C and to resist freezing at subzero temperatures. In general, media which supported good growth at 20 °C froze at −7 °C, whereas media which did not freeze at −7 °C did not support good growth at 20 °C. In supercooled liquid media, however, three species of Bacillus grew at −2 °C, one at −4.5 °C, and five at −5 to −7 °C. Generation times were approximately 4 days at −2 °C, 7 days at −4.5 °C, and 9 to 11 days at −5 to −7 °C. On supercooled growth-supporting agar media, two Gram-positive cocci, one Gram-negative rod, and four species of yeast grew at −2 °C. The latter also grew at −4.5 °C. Three species of Bacillus grew at −7 °C, and two at −10 °C.

Four isolates of C. sativus were able to use a number of carbon sources to varying degrees with little evidence of a differential growth response by the isolates. With most carbon sources the response was different when growth on liquid media was compared with that on agar media. Dextrin and L-sorbose were exceptions as dextrin supported good growth with both types and L-sorbose poor with both. Lactose supported the best mycelial growth on liquid media and dextrin the best radial growth and sporulation on agar media when compared with sucrose.

Context: Aspiration Pneumonia is a common complication in patients with altered consciousness, if general supportive care is not taken properly. There is no national study on microbial aspect of aspiration pneumonia in hospitalized patients with altered consciousness. This study was done to see the microbial aspect of aspiration pneumonia in patients with altered consciousness admitted in a tertiary level hospital in Bangladesh. Methods: A prospective observational study was done on 50 adult patients of aspiration pneumonia with altered consciousness admitted in the medicine department of medicine, Dhaka Medical College Hospital, Dhaka, between January and December 2010. Aspiration pneumonia was confirmed by clinical examination and laboratory investigations. Hematologic measurements, chest x-ray, blood gas analysis, estimation of blood urea, serum creatinine and random blood sugar were done in all patients. Sputum for gram staining and culture & sensitivity as well as blood culture & sensitivity were done in patients who were diagnosed as aspiration pneumonia clinically and radiologically. Validated sputum was cultured in blood agar, chocolate agar and McConkey’s agar media. Blood culture was done in trypticase soya broth, blood agar, chocolate agar and McConkey’s agar media. Results: The mean age was 38.9±17.3 years. 35 (70%) patients were male and 15 (30%) patients were female. Blood culture was found positive in 30.8% patients who developed aspiration pneumonia among the study patients. In sputum for Gram staining, only Gram positive Cocci was present in 46.15%, only Gram negative Bacilli was present in 38.46%, mixed Gram positive cocci and Gram negative bacilli was present in 7.7% and Gram negative Coccobacilli was found in 7.7% patients who developed aspiration pneumonia among the study patients. In growth positive cases (n=11) sputum culture revealed 45.45% streptococcus pneumoniae, 36.36% Klebsiella pneumoniae, 9% Pseudomonas species and 9% Haemophylus influenzae. Introduction: Pneumonia is a worldwide, serious threat to health and an enormous socio-economic burden for health care system. According to recent WHO data, each year 3-4 million patients die from pneumonia. The clinical presentations and bacterial agents responsible for community acquired pneumonia (CAP) varies according to geography and culture.Methods: A cross sectional observational study conducted among the 53 consecutive patients with a clinical diagnosis of CAP in admitted patient in the department of Medicine, DMCH, during January 2010 to December 2010. Hematological measurements (TC of WBC, Hb%, ESR, platelet count), blood culture, chest X-ray P/A view, sputum for Gram staining and culture sensitivity, sputum for AFB, blood urea and random blood sugar were done in all cases. ELISA for IgM antibody of Mycoplasma pneumoniae and Chlamydia pneumoniae were done in sputum culture negative cases.Results: The mean (±SD) age was 38.9±17.3 years and Male female ratio was 3:1. Fever, chest pain and productive cough were the most common clinical features. The mean (±SD) respiratory rate was 23.0±2.8 /minute . COPD and DM were found in 17.0% and 5.7% of patients respectively . Blood culture was found positive in only 1.9% of the study patients. Gram positive Cocci 62.26%, Gram negative Bacilli 9.43%, mixed Gram positive cocci and Gram negative bacilli 11.32% and Gram negative Cocco Bacilli 1.9% were observed and in 15.03 % cases, no bacteria could be seen. Sputum culture revealed 53.8% streptococcus pneumoniae, 26.9% Klebsiella pneumonia as predominant organism. Mycoplasma pneumoniae and Chlamydia pneumoniae were found in 7.4% and 3.7% respectively by serological test. For Streptococcus pneumoniae, sensitive antibiotics were Amoxyclav and Levofloxacin. For Gram negative bacilli and coccobacilli, more sensitive antibiotics were Meropenem, Ceftriaxone, and Clarithromycin. The best sensitive drug were found meropenem. The mean (±SD) duration of hospital stay was 5.0±1.7 days with ranging from 3 to 10 days.Conclusion: Region based bacteroiological diagnosis of Cap is important for selecting the best and sensitive drugs for complete cure.J Bangladesh Coll Phys Surg 2016; 34(3): 128-134

Abstract BackgroudTo compare the performance of the new flow cytometer UF-5000 with UF-1000i (Sysmex, Kobe, Japan) and Gram stain in predicting the bacterial patterns in urine samples MethodsWomen with symptoms suggestive of urinary tract infection were enrolled. Mid-stream urine sample was collected for gram staining, urine analysis and urine culture. Bacterial patterns were classified though UF1000i (none, cocci bacteria or rods/mixed growth), UF-5000 (none, cocci, rods or mixed growth) and Gram stain. Results Among the 102 samples, there were 10 gram-positive cocci, 2 gram-positive bacilli, 66 gram-negative rods, and 24 mixed growth. The sensitivity/specificity of the UF-1000i was 81.8/91.1% for gram-negative rods and 23.5/96.9% for cocci/mixed. The sensitivity/specificity of the UF-5000 was 80.0/88.2% for gram negative rods and 70.0/86.5% for gram-positive cocci.ConclusionsThe UF-5000 demonstrated the good sensitivity and specificity for Gram-negative bacilli bacteria and demonstrated an improved sensitivity for detecting Gram-positive cocci.

The purpose of the study is to improve the selective supplement for selective media with the purpose to produce enterobacteria. Tasks of the study are to identify the sensitivity of strains obtained of enterobacteria in regard to an-tibiotics; develop a new selective supplement with antibiotics to the nutrient medium Drigalski Lactose Agar. Media should have a content that in the best way possible ensures the growth and reproduction of microorganisms of cer-tain species or family. Intensive biotechnology development and Microbiology allows today to develop new nutrient media and modify the already existing content of media. The object of the study was a new selective additive with antibiotics to the nutrient medium Drigalski Lactose Agar. 253 isolates of bacteria produced from the intestinal mi-crobiotope of different animal species have been the Material for research. The study was conducted in the period from 2010 to 2017. Carbenicillin 30±2.3 from the group of carboxypenicillins and piperacillin 37±2.5 from the group of ureidopenicillins, kanamycin 24±1.5, amikacin 26±1.7 and gentamicin 25±0,8, cefepime 38±3.2 from the group of IV generation cephalosporins, tetracycline 28±1.6, doxycycline 34±2.3 and chloramphenicol 31±2.5, nalidixic acid 37±2.8, trimethoprim 35±3,4 demonstrated the greatest antimicrobial activity against all cultures of enterobac-teria that has been achieved. The high resistance of enterobacteria was shown to benzylpenicillin from the group of natural penicillins, to streptomycin, cephalotine from the group of cephalosporins of the first generation, to polymyx-in B, to ofloxacin (tarivid) and metronidazole. Antibacterial drugs effective against the accompanying gram-positive and gram-negative microflora were considered as the samples of the selective components. Vancomycin from the group of glycopeptides, linezolid from the group of oxazolidinones, and telithromycin from the group of ketolides were chosen. Antibiotics vancomycin and telithromycin in a dose of 0.008 g/dm3, linezolid 0.004 g/dm3 were cho-sen as the selective additive to Drigalski Lactose Agar medium.

In 1998, plants of periwinkle (Catharanthus roseus L.) showing small leaves, short internodes, and dieback symptoms were observed in a garden at the Instituto Agronomico do Parana (IAPAR), Londrina, PR, Brazil. Stems of these plants were cut into short sections and the sap extracted from the tissue by squeezing with pliers. The sap was blotted onto a glass slide and examined for the presence of bacteria by light microscopy (×400). Microscopy observations revealed the presence of a large number of slender, rod-shaped bacterial cells. The bacteria present in the stems of periwinkle were isolated on buffered cysteine-yeast extract (BCYE) and periwinkle wilt (PW) agar media. Stems were disinfected in 70% alcohol and cut into short sections, and the sap extracted as described above. The sap was blotted directly onto the media and the plates were incubated at 28°C. Typical colonies of Xylella fastidiosa were observed 10 days after isolation on both media. Indirect immunofluorescence tests with antibody specific to X. fastidiosa and anti-IgG conjugated with tetrametylrhodamine isothiocyanate (TRITC) were carried out with xylem sap of periwinkle stem and the isolated bacteria. In both cases, immunofluorescence tests were positive for X. fastidiosa. These results confirm that periwinkle plants were infected with X. fastidiosa. This is the first report of the association of X. fastidiosa with periwinkle plants in Brazil. However, the symptoms observed for the X. fastidiosa-infected periwinkle plants differed from those described previously in the U.S. (1): those symptoms consisted of marginal chlorosis and occasional vein clearing of leaves and wilting of the plants. <span>Pollution of water environment with heavy metals is becoming one of the most severe environmental and human health hazards. Lead (Pb) is a major pollutant and highly toxic to human, animals, plants, and microbes. </span><span lang="IN">Toxic metals are difficult to remove from the environment, since they cannot be chemically or biologically degraded and are ultimately indestructible. Biological approaches based on metal-resistant microorganisms have received a great deal of attention as alternative remediation processes. </span><span>This study aim to isolat</span><span lang="IN">e</span><span> and characterize Pb resistant of heterotrophic bacteria in Cilalay Lake, </span><span lang="IN">West Java, </span><span>Indonesia. The most abundant type of resistant </span><span lang="IN">bacteria </span><span>to lead was Gram negative more than Gram positive. Present study was conducted to determine the microbiological status of water from dispensers in different roadside hotel and restaurants of Dhaka city. Samples were collected from seven hotel and restaurants. Aerobic heterotrophic bacterial count ranged between 1.5 × 10 and 8.8 × 103 cfu/ml. Enteric and related bacterial abundance in MacConkey, SS and Cetrimide agar media ranged from 0 to 4.9 × 106, 0 to 2.1 × 105 and 0 to 1.2 × 106cfu/ml, respectively. In total, 28 bacterial isolates were obtained during the study period. Among them, 15 were heterotrophic isolates and 13 were enteric and related bacteria. Among 15 aerobic heterotrophic isolates, 11 were gram positive and five were gram negative. Out of 11 gram positive isolates, 7 belonged to the genus Bacillus viz. B. circulans, B. subtilis, B. stearothermophilus, B. brevis and B. cereus and one to coccus viz. Micrococcus roseus. The other gram positive species were Kurtia gibsonii, Auriobacterium liguefaciens and Curtobacterium luteum. Four gram negative isolates were Neisseria elongate sub. spp. glycolytica, Plesiomonas shigelloides, Pseudomonas fluorescens biovar 1, Pseudomonas aeruginosa. All 13 enteric and related isolates were gram negative, short rod; and non-spore formers and belonged to the genera Escherichia, Klebsiella, Shigella and Pseudomonas. Introduction. Cypress (Cupressus sempervirens L.) belongs to the family Cupressaceae. It is evergreen, and grows in Mediterranean region. The Cypress leaves and young branches are utilized to produce the essential oil. Cypress oil contain a number of components, in it α-pinene, Δ3-carene, α-terpinyl acetate, cedrol, α-terpinolene, β-myrcene, limonene, α-terpineolene, terpinen-4-ol, β-pinene, δ-cadinene and sabinene. The oil is used in therapy different diseases. It to have antimicrobial activity. Aim. The aim of the date was evaluation the susceptibility of anaerobic bacteria to Cypress oil. Material and methods. The anaerobic bacteria were isolated from patients. The 62 microorganisms, in it 36 strains of Gram-negative rods, 14 Gram-positive cocci and 12 Gram-positive rods, and 7 reference strains were tested. Susceptibility (MIC) was determined by means of plate dilution technique in Brucella agar supplemented with 5% defibrynated sheep blood, menadione and hemin. The Cypress oil was dissolved in DMSO and distilled water to obtain final following concentrations: 2.5, 5.0, 7.5, 10.0, 15.0 and 20.0 mg/ml. Inoculum containing 106 CFU per 1 ml was seeded with Steers replicator upon the agar with oil or without the oil (strains growth control). The agar plates was incubated in anaerobic condition in anaerobic jar in 37°C for 48 hrs. The MIC was interpreted as the lowest concentration of Cypress oil inhibiting the growth of tested bacteria. Results. The results indicated that from among Gram-negative rods the most susceptible to Cypress oil was the strains from genus Tannerella forsythia (MIC < 2.5-5.0 mg/ml), Bacteroides uniformis (MIC = 5.0 mg/ml), Bacteroides vulgatus and Porphyromonas asaccharolytica (MIC 5.0-7.5 mg/ml) and Porphyromonas levii (MIC = 7.5 mg/ml). The strains from genera Fusobacterium and of Bacteroides fragilis were the susceptible to 2.5-≥ 20.0 mg/ml. The Cypress oil was least active towards Prevotella and Parabacteroides strains (MIC ≥ 20.0 mg/ml).The tested Gram-positive cocci were more susceptible. The growth of the strains were inhibited by concentrations in ranges ≤ 2.5-7.5 mg/ml. The oil was minor active towards Gram-positive rods (MIC ≤ 2.5-20.0 mg/ml). Among the strains the genus of Actinomyces odontolyticus (MIC = 5.0 mg/ml) and Actinomyces viscosus (MIC ≤ 2.5-7.5 mg/ml) were the most susceptible. The growth of rods of Bifidobacterium breve was inhibited by concentrations 10.0 mg/ml. The data indicates that the Gram-negative rods were the less susceptible than Gram-positive bacteria to cypress oil. Conclusions. Among Gram-negative rods the most susceptible were the strains Tannerella forsythia, Bacteroides uniformis, Bacteroides vulgatus, Porphyromonas asaccharolytica and Porphyromonas levii. The oil was more active against Gram-positive cocci. Gram-positive anaerobic bacteria demonstrate the more susceptible to Cypress oil then Gram-positive rods.

Background: Surgical site infection is one of the commonest complications after surgical intervention. Source of wound infections may be exogenous or endogenous. Wound swabs culture is the most frequently used method for confirming the diagnosis of surgical site infection. A regular bacteriological review is necessary to identify the causative agents and their antibiotic susceptibility pattern.Objective: The purpose of the present study was to find out the aerobic bacteria from wound swabs culture and their antibiotic susceptibility pattern.Methodology: A total of 175 wound swabs sample were collected aseptically from surgical site infection during the study period. Standard bacteriological methods were used for isolation and identification of organisms and their susceptibility pattern.Results: A total of 175 wound swab samples were collected from surgery departments. Out of 175 samples 102 (58.28%) were positive by culture. Among the isolated organisms the gram negative bacilli were predominant 67 (65.68%) than the gram positive cocci 35 (34.32%).The most common isolated bacteria were Escherichia.coli 30(29.41%) followed by Staphylococcus aureus 28 (27.45%) Pseudomonous areuginosa 20(29.85%), Acinetobacter spp 08(11.94%), Coaggulase negative Staphylococcus (CONS) 07(6.86%) Klebsiella pneumoniae 05(4.90%), Proteus spp 03(4.45%), and Citobacter spp 01(1.49%). Among the total 175 samples 114(65.14%) were collected from male patients and 61(34.86%) were from female patients. All the isolated gram negative bacilli showed resistant to all antibiotics and all isolated gram positive cocci were resistant to all antibiotics except Linezolid. Among the isolated Staphylococcus aureus 8 (24.85%) were MRSA and one (3.57%) were VRSA. The most effective antibiotics were Amikacin, Meropenem, Imipenem and Tazobactam in this study.Conclusion: Surgical site infections remain the commonest post operative complications after surgery and one of the commonest encountered hospital acquired infections. Therefore, periodic review needs be done to find out the causative agents and their antibiotic susceptibility pattern for better treatment and managementBangladesh Journal of Infectious Disease 2015;2(2):28-32
