Comparative Electropherograms of Coregonus clupeaformis, Salvelinus namaycush, S. alpinus, S. malma, and S. fontinalis from the Family Salmonidae

Zone electropherograms of muscle myogens and blood proteins of members of the family Salmonidae revealed in addition to species specificity, marked similarities at the genus and family levels. In genera investigated, other than the Oncorhynchus, intraspecies polymorphisms were found. Two general hemoglobin patterns were uncovered from specimens of Coregonus clupeaformis from Great Slave Lake while their muscle myogen patterns were specifically uniform. Salvelinus namaycush from Lake Superior and Cayuga Lake were readily distinguishable by differences in their multiple hemoglobin patterns as well as by plasma proteins as revealed by polyacrylamide disc electrophoresis. Muscle myogen electropherograms of S. namaycush, S. alpinus, and S. malma showed striking similarities, the latter two being almost superimposable while the patterns of Salvelinus fontinalis were more closely allied to that of the genus Salmo. The value of muscle myogens in phylogenetic studies and in intraspecies protein variations as diagnostic characters in stock analyses is discussed.

The starch gel electropherogram of muscle myogens and blood hemoglobins of representatives from the families Petromyzontidae, Esocidae, Centrarchidae, and Percidae were found to be within the limits of species specificity with some exceptions. Evidence of polymorphism was found in the muscle myogen of Stizostedion vitreum of the family Percidae. Only one type of homozygous pattern, designated A, was found in specimens from Great Slave lake, while both the homozygotes A and B as well as the heterozygote AB were found in samples from Lake St. Clair. Further genetically interpretable polymorphisms were also found in the minor protein zones. These natural chemical markers are discussed from the standpoint of their usefulness in subpopulation analyses. The presence of protein zones of common electrophoretic mobilities in three species of the family Petromyzontidae indicated marked resemblances of the muscle myogens at the family level. Total plasma proteins of P. marinus showed vast changes during their parasitic and nonparasitic stages of development. The muscle myogens of some members of the family Centrarchidae also showed strong familial relationships. In hybrids of this group biosynthetic systems for muscle myogens and plasma proteins of both parents were present.

The muscle myogens of Catostomus catostomus exhibit polymorphism. Specimens from Great Slave Lake and Babine Lake were homozygous to pattern type A, whereas those from Lake Superior possessed patterns homozygous to A and B as well as the heterozygote AB. Those of C. commersoni, Moxostoma macrolepidotum, M. anisurum, and M. erythrurum were relatively uniform, showing only odd variants. The two specimens of Ictiobus bubalus examined displayed different patterns. Blood hemoglobin patterns of M. macrolepidotum existed in five, M. anisurum in two, and C. commersoni in three polymorphic forms. The number of specimens of I. bubalus, M. melanops, and Carpiodes cyprinus examined was insufficient to assess the existence of polymorphism. Muscle myogen electropherograms of the eight species investigated reflect overall family resemblances, distinctive generic groupings as well as phylogenetic relationships which compare favorably with those in existence. Intergradations might occur upon a more extensive coverage of this family and which might somewhat obscure this overall picture are discussed.

Polyacrylamide disc electrophoresis of plasma proteins and starch-gel electrophoresis of hemoglobins and muscle myogens of adult and ammocoete forms of three species of Great Lakes lamprey were carried out. Blood proteins were shown to undergo marked changes upon transformation of the ammocoete into the adult form. Muscle myogen did not undergo any change during transformation. The muscle myogen electropherograms of Ichthyomyzon unicuspis and Lampetra lamottei were found to be the same.

Understanding the behavior of nanoparticles upon contact with a physiological environment is of urgent need in order to improve their properties for a successful therapeutic application. Most commonly, the interaction of nanoparticles with plasma proteins are studied under in vitro conditions. However, this has been shown to not reflect the complex situation after in vivo administration. Therefore, here we focused on the investigation of magnetic nanoparticles with blood proteins under in vivo conditions. Importantly, we observed a radically different proteome in vivo in comparison to the in vitro situation underlining the significance of in vivo protein corona studies. Next to this, we found that the in vivo corona profile does not significantly change over time. To mimic the in vivo situation, we established an approach, which we termed “ex vivo” as it uses whole blood freshly prepared from an animal. Overall, we present a comprehensive analysis focusing on the interaction between nanoparticles and blood proteins under in vivo conditions and how to mimic this situation with our ex vivo approach. This knowledge is needed to characterize the true biological identity of nanoparticles.

We found no significant differences in slope or intercept for the regression of loge fecundity on loge fork length among samples of whitefish from four lakes near Yellowknife in the Northwest Territories. The equation describing the relationship between fecundity and fork length for these populations was:[Formula: see text]Five other populations for which length–fecundity relationships could be calculated had length exponents ranging from 3.20 to 4.38, suggesting a nonlinear relationship between weight and fecundity. Six of the nine populations as well as four others for which limited data were available all had similar relative fecundities. Fish from Buck Lake in Alberta and from Lake Erie had high relative fecundities while fish from Great Slave Lake had low relative fecundity.

Patterns of genetic variation among lake trout (Salvelinus namaycush) of eastern Lake Superior were examined using starch gel electrophoresis. We used 484 individuals sampled from three areas, representing three morphological types (leans, humpers, and siscowets). Of 50 loci examined, 44 were monomorphic in all groups sampled. Genetic variation occurs at six loci AAT-1,2, MDH-3,4, ME-1, and SOD-1. The average heterozygosity found (H = 0.015) is low relative to other salmonid species. A significant amount of heterogeneity exists among the 10 lake trout samples. These differences are due to variation within as well as between morphological types. The significance and management implications of these data are discussed.Key words: genetic variation, lake trout, Salvelinus namaycush, Lake Superior

The Mediterranean black coral fauna includes type species of four antipatharian genera belonging to four different families, therefore phylogenetic studies hold great potential for enhancing systematics within the order. The analysis of six Mediterranean antipatharian species by means of nuclear sequence data of internal transcribed spacer (ITS1 and ITS2) rDNA confirms the separation into different families, as was previously noted on a morphological basis, with a clear distinction of the family Leiopathidae, whose position is supported by a unique number of mesenteries and lack of spines on thicker ramifications. The position of a newly recorded black coral species for the Mediterranean basin belonging to the genus Phanopathes is discussed. Antipathes dichotoma, the type species of the genus Antipathes, on which the order Antipatharia was based, does not group with other members of the family Antipathidae. Supporting a recent finding based on mitochondrial markers, this suggests a critical need for revision of the families that will be impacted by reassignment of this nomenclaturally important taxon.

Catches per unit of effort (CPUE) with experimental gill nets, recruitment, growth, and condition were monitored between 1977 and 1992 to evaluate the impact of impoundment on the main fish species of La Grande 2, Opinaca, and Caniapiscau reservoirs and the Boyd–Sakami diversion. CPUE and recruitment of northern pike (Esox lucius) and lake whitefish (Coregonus clupeaformis) increased markedly at most stations after impoundment and decreased at the end of the series. The lake whitefish and cisco (Coregonus artedii) showed their most striking rise in CPUE at two bay stations of La Grande 2 and Opinaca reservoirs. CPUE and recruitment of the longnose sucker (Catostomus catostomus), white sucker (Catostomus commersoni), and lake trout (Salvelinus namaycush) (Caniapiscau) showed a general decrease following impoundment. CPUE for the walleye (Stizostedion vitreum) also decreased at several stations; however, the two most southerly stations in La Grande 2 reservoir and the Boyd–Sakami station showed high CPUE during the series. Concentration–redistribution phenomena explain part of the observed variations in CPUE. Correlation analyses showed that walleyes and white suckers were attracted to the warmer, more turbid stations, and that the high primary and secondary productivity of bay stations attracted the coregonines. Growth and condition of the main species increased during variable time intervals after impoundment and decreased at the end of the series.

Microniphargus leruthi Schellenberg, 1934 (Amphipoda: Niphargidae) was first described based on samples collected in Belgium and placed in a monotypic genus within the family Niphargidae. However, some details of its morphology as well as recent phylogenetic studies suggest that Microniphargus may be more closely related to Pseudoniphargus (Amphipoda: Pseudoniphargidae) than to Niphargus. Moreover, M. leruthi ranges over 1,469 km from Ireland to Germany, which is striking since only a few niphargids have confirmed ranges in excess of 200 km. To find out the phylogenetic position of M. leruthi and check whether it may be a complex of cryptic species, we collected material from Ireland, England and Belgium then sequenced fragments of the mitochondrial cytochrome c oxidase subunit 1 gene as well as of the nuclear 28S ribosomal gene. Phylogenetic analyses of both markers confirm that Microniphargus is closer to Pseudoniphargus than to Niphargus, leading us to reallocate Microniphargus to Pseudoniphargidae. We also identify three congruent mito-nuclear lineages present respectively in Ireland, in both Belgium and England, and in England only (with the latter found in sympatry at one location), suggesting that M. leruthi is a complex of at least three species with a putative centre of origin in England.
