The addition of the antithyroid agents, thiocyanate and propylthiouracil, to a basal diet low in the sulphur-containing amino acids and in α-tocopherol ameliorated its necrogenic effect, but did not prevent the development of acute massive liver necrosis when this ration was fed to young rats. The necrogenic effect of a basal diet supplemented with desiccated thyroid, an agent that greatly potentiates the necrogenicity of the ration, was completely abolished by the addition of cystine or α-tocopherol. These substances did not overcome the well known toxic effects of excessive amounts of thyroid tissue.

A paper chromatographic study of the amino acids and peptides in the blood of rats with dietary liver necrosis has been made.The relative levels of alanine, methionine, tyrosine, glutamic acid, threonine, citrulline, glutamine, oxidized glutathione, and gamma aminoisobutyric acid were increased. The level of reduced glutathione was slightly reduced and levels of valine, taurine, and proline remained unchanged. Beta-alanine, which could not be detected in the blood of normal rats, was found in the case of those with acute liver damage.Although arginine was present in the blood of normal animals and in those on a necrogenic diet prior to the development of the liver lesion, it was not detectable in those animals with acute hepatic necrosis. The possible implications of this finding have been discussed.

Abstract Two experiments were conducted to evaluate the digestible energy (DE), metabolizable energy (ME), and standardized ileal digestibility (SID) of amino acids (AA) in dried yeast (DY) and soybean meal (SBM) fed to pigs. In Exp. 1, 30 barrows with an initial body weight (BW) of 20.7 ± 1.01 were assigned to 5 diets in a randomized complete block design with period and BW as blocking factors. A basal diet was prepared to contain corn, canola meal, and soybean oil as energy-contributing ingredients. Four additional diets were prepared by adding 5 or 10 g/kg DY or SBM at the expense of energy-contributing ingredients in the basal diet to estimate the DE and ME in test ingredients by regression analysis. On a dry matter basis, estimated DE and ME in DY were 4,022 and 3,352 kcal/kg, respectively, and those in SBM were 3,876 and 3,601kcal/kg, respectively. There was no difference in estimated DE or ME between DY and SBM. In Exp. 2, 21 barrows (initial BW = 20.0 ± 1.31 kg) surgically fitted with T-cannulas at the distal ileum were assigned to 3 diets in a randomized complete block design with BW as a blocking factor. Two diets were prepared to contain DY or SBM as the sole source of nitrogen, and a nitrogen-free diet was prepared to determine the basal ileal endogenous losses of AA. The SID of AA, except for Gly and Pro, in SBM were greater (P < 0.05) than in DY. The SID of indispensable AA in DY ranged from 64.7% for Thr to 86.1% for Arg, whereas those in SBM ranged from 84.8% for Thr to 92.3% for Arg. In conclusion, energy values in DY was comparable with SBM, but the SID of most AA in DY were less than in SBM.

This study is an attempt to evaluate the hepatoprotective activity ofTabernaemontana divaricataagainst DEN and Fe NTA induced liver necrosis in rats. Ethanolic extract of the whole plant ofTabernaemontana divaricataat doses of 200 and 400 mg/kg body weight and 5-fluorouracil (standard drug) was orally administered to male Wistar Albino rats once daily for 24 weeks, simultaneously treated with the carcinogen DEN and Fe NTA. In simultaneously treated animals, the plant extract significantly decreased the levels of uric acid, bilirubin, AST, ALT, and ALP in serum and increased the levels of liver marker enzymes in liver. Treatment with the extracts resulted in a significant increase in the levels of antioxidants accompanied by a marked reduction in the levels of malondialdehyde when compared to DEN and Fe NTA treated group. When compared with 200 mg/kg bw rats, 400 mg/kg bw rats and 5-fluorouracil treated rats showed better results in all the parameters. The histopathological studies confirmed the protective effects of extract against DEN and Fe NTA induced liver necrosis. Thus, it could be concluded that the use ofTabernaemontana divaricataextract in the treatment of carcinogen induced hepatic necrosis.

SummaryThe effects of the form in which amino acids are presented to the abomasum on the milk production of dairy cows receiving a basal diet of grass silage and a barley-based supplement were examined in two experiments. Effects of abomasal infusions of sodium caseinate were compared with the effects of corresponding levels of either an enzymic hydrolysate of casein (Expt 1) or a corresponding mixture of free amino acids (FAA; Expt 2). In Expt 1, although the yield of protein in milk increased progressively with each level of infusion, the yields of protein were greater for the caseinate than for the hydrolysate. Again, in Expt 2, for milk protein yield, sodium caseinate was superior to FAA at the lower level of infusion. In both experiments, the hydrolysate and FAA treatments were associated with higher concentrations of fat in the milk. There were indications of differences in the pattern of secretion of glucagon between the caseinate and FAA treatments. It is concluded that the differences between treatments relate either to the kinetics of absorption of amino acid residues or to the action of bioactive peptides released during digestion of casein.

The aim of this study was to evaluate the effects of aflatoxin B1 (AFB1) and monensin (MONS) interaction on the liver and intestinal histological changes in poultry, and the influence of Pichia kudriavzevii RC001. One-day-old commercial line (Ross 308) broilers (n=120) were individually weighed and randomly assigned to 8 treatments (15 broilers/treatment, 5 broilers per cage and 3 replicates/treatment). The experimental diets were: Group 1: basal diet (BD); Group 2: BD + MONS (50 mg/kg); Group 3: BD + P. kudriavzevii RC001 (1 g/kg); Group 4: BD + AFB1 (100 μg/kg); Group 5: BD + MONS + P. kudriavzevii RC001; Group 6: BD + AFB1 + P. kudriavzevii RC001; Group 7: BD + AFB1 + MONS + P. kudriavzevii RC001; Group 8: BD + AFB1 + MONS. When MONS was added, the typical AFB1 macroscopic and microscopic alterations were intensified. The P. kudriavzevii RC001 cytotoxicity and genotoxicity assays with Vero cells and with broiler chicken’s erythrocytes, demonstrated that P. kudriavzevii RC001 neither were non-cytotoxic nor genotoxic. When MONS was added in the presence of P. kudriavzevii RC001, the toxic effect of AFB1 on liver was not prevented. When P. kudriavzevii was present alone, the same prevention of the pathological damage was observed in the intestine of poultry fed with AFB1. The smallest apparent absorption area was obtained when AFB1 and MONS were added in the feed (P<0.05). AFB1 and MONS interaction demonstrated important toxic effects. Although P. kudriavzevii was effective in ameliorating the adverse effects of AFB1 alone on liver pathology and gut morphology, it was not able to diminish the toxic effects of AFB1 in presence of MONS. It suggests that P. kudriavzevii could be used as feed additive or counteracting the toxic effects of AFB1 in poultry production in the absence of MONS.

To determine the role of nonsulfur vs. sulfur amino acids in regulation of cysteine metabolism, rats were fed a basal diet or diets supplemented with a mixture of nonsulfur amino acids (AA), sulfur amino acids (SAA), or both for 3 wk. Hepatic cysteine-sulfinate decarboxylase (CSDC), cysteine dioxygenase (CDO), and γ-glutamylcysteine synthetase (GCS) activity, concentration, and mRNA abundance were measured. Supplementation with AA alone had no effect on any of these measures. Supplementation of the basal diet with SAA, with or without AA, resulted in a higher CDO concentration (32–45 times basal), a lower CSDC mRNA level (49–64% of basal), and a lower GCS-heavy subunit mRNA level (70–76%). The presence of excess SAA and AA together resulted in an additional type of regulation: a lower specific activity of all three enzymes was observed in rats fed diets with an excess of AA and SAA. Both SAA and AA played a role in regulation of these three enzymes of cysteine metabolism, but SAA had the dominant effects, and effects of AA were not observed in the absence of SAA.

AbstractCa2+-sensing receptor (CaSR) represents a potential therapeutic target for inflammatory bowel diseases and strongly prefers aromatic amino acid ligands. We investigated the regulatory effects of dietary supplementation with aromatic amino acids – tryptophan, phenylalanine and tyrosine (TPT) – on the CaSR signalling pathway and intestinal inflammatory response. The in vivo study was conducted with weanling piglets using a 2 × 2 factorial arrangement in a randomised complete block design. Piglets were fed a basal diet or a basal diet supplemented with TPT and with or without inflammatory challenge. The in vitro study was performed in porcine intestinal epithelial cell line to investigate the effects of TPT on inflammatory response using NPS-2143 to inhibit CaSR. Dietary supplementation of TPT alleviated histopathological injury and decreased myeloperoxidase activity in intestine challenged with lipopolysaccharide. Dietary supplementation of TPT decreased serum concentration of pro-inflammatory cytokines (IL-1β, IL-6, IL-8, IL-12, granulocyte-macrophage colony-stimulating factor, TNF-α), as well as the mRNA abundances of pro-inflammatory cytokines in intestine but enhanced anti-inflammatory cytokines IL-4 and transforming growth factor-β mRNA levels compared with pigs fed control diet and infected by lipopolysaccharide. Supplementation of TPT increased CaSR and phospholipase Cβ2 protein levels, but decreased inhibitor of NF-κB kinase α/β and inhibitor of NF-κB (IκB) protein levels in the lipopolysaccharide-challenged piglets. When the CaSR signalling pathway was blocked by NPS-2143, supplementation of TPT decreased the CaSR protein level, but enhanced phosphorylated NF-κB and IκB levels in IPEC-J2 cells. To conclude, supplementation of aromatic amino acids alleviated intestinal inflammation as mediated through the CaSR signalling pathway.

A histologic examination of spontaneous cases of sawdust livers in cattle indicated that the focal liver necrosis was an expression of vitamin E-selenium deficiency. The condition was reproduced in Hereford steers by feeding a diet rich in polyunsaturated fatty acids and poor in protein, vitamin E. and selenium. Lesions also occurred in the kidney, heart, skeletal muscled and pylorus. Addition of dictary protein or injection of selenium partially prevented the condition. Cellular anoxia with formation of hyalinc bodies in the liver and kidney was considered to be the common denominator of the degenerative changes. Due to the relatively mild tissue changes, plasma GOT and OCT determinations were found to be of no diagnostic value.
