Excretion of a natural inhibitor of anthocyanin synthesis during germination of mung bean seedlings

A natural inhibitor of anthocyanin synthesis has been found in seeds of mung beans (Vigna radiata). The inhibitor might play an important role in the regulation of anthocyanin accumulation in seedlings. It is mainly localized in the seed coat and in the cotyledons and is excreted in the incubation medium soon after sowing. Removal of the inhibitor by rinsing the seedlings and transferring them to fresh medium results in higher anthocyanin accumulation as compared with seedlings left in the original incubation medium. Addition of the inhibitor 12 h after its removal does not affect anthocyanin accumulation when the seedlings have been grown in complete darkness. On the other hand, anthocyanin accumulation initiated by irradiation with red light is inhibited under these conditions. The inhibitor has been partially purified and some of its chemical characteristics were determined from its behaviour during purification.

Developmental patterns of amylase activity in mung bean (Vigna radiata) cotyledons were observed by histochemical methods. In axis-attached cotyledons, the development of amylase activity and the decompostion of starch grains occurred first in cells located farthest from vascular bundles, and 3 days after imbibition, starch granules appeared to remain intact only in two to three cell layers around the vascular bundles. In axis-detached cotyledons, on the other hand, no conspicuous change in amylolytic activity was detected. When cotyledons were allowed to imbibe water through the surface not covered by the seed coat, amylase activity developed, even if they were detached from the axis. However, the developmental patterns of the activity were quite different from those with attached cotyledons; high amylase activity was detected in the cell layers beneath the epidermis of the surface not covered by the seed coat. These histochemical observations are related to previous results of biochemical experiments. Key words: amylase development, cotyledons, germination, histochemistry, mung bean, Vigna radiata.

We have previously shown that light is essential for anthocyanin synthesis in detached petunia flowers. In this study, we examined the effect of light quality on anthocyanin accumulation and on the expression of the anthocyanin biosynthetic gene chalcone synthase (chs) in the attached petunia flowers. When plants were kept for 48 h in the dark,chsexpression could not be detected. Blue light and red light promoted the expression of the gene to similar levels, whereas the effect of green light was slightly smaller. Neither UV-A nor UV-B radiation were required for anthocyanin accumulation, or for the induction and maintenance ofchsexpression. The results of this study show that light is essential for anthocyanin synthesis in petunia flowers and that its active spectrum includes most of the visible part but not UV radiation.

ABSTRACT An extract of human urine, which was previously shown to stimulate aldosterone production by rat adrenal sections, was further purified. Evidence was obtained that its aldosterone-stimulating effect was due to the presence of ammonium ions. Addition of ammonium chloride and of urine extract to the incubation medium caused identical increases in aldosterone production in vitro. In addition to ammonium ions, rubidium and caesium ions also stimulated aldosterone production up to 250% that of control values without a significant effect on corticosterone production. Similar dose-response curves were obtained when increasing concentrations of potassium, ammonium, rubidium and caesium ions were tested. Aldosterone production was maximal at concentrations of 7 mval/1 and was significantly lower at higher concentrations. When ammonium chloride and ACTH were simultaneously added to the incubation medium, the production of aldosterone and of corticosterone was lower than with ACTH alone. On the other hand, the stimulating activity on aldosterone and corticosterone production by »TPN« (NADP) and glucose-6-phosphate was enhanced by the simultaneous addition of ammonium chloride.


For identification of varieties, it is desirable to use genetically predetermined traits that are decisive for the varietal identification of the distinguishing characteristics of the varieties production and new genotypes for selection. The presence a complete evaluation of the selection samples a particular crop is the basis for the formation of informative databases characteristics the varieties of standards used in plant examination to determine the level of expression a characteristic particular genotype according to the code of their manifestation. For the 20 traits we used to identify, we identified 6 least volatile, highly trait-stable traits over the years of study to identify. These are: “coloring of the pubescence of the main stem”, “the size of the lateral leaf”, “the intensity of green leaf color”, “flower color”, “the intensity of brown bean color”, “basic color of the seed coat”. The other 14 traits are highly variable and unstable among varieties, but may be significant under certain conditions. According to the results of the research we have determined the conformity of the collection varieties by code graduation. In the study, we selected varieties with corresponding codes of attributes of the reference varieties the UPOV technique. On the basic of the color the pubescence of the main stem, 36% varieties are with grey stem pubescence. Among the standards: Apache, Alaric, Talon according to the grey color of the pubescence, we identified varieties Zolotysta, Ozzie, Diona. The 2947 564/84, Stepnaya-90, Knyazhna. The lateral leaf size is a sign that affects the increase in green mass of plants and subsequently on productivity. Graduation of the sign small size of leaf had varieties: Zuma, Ksenya; middle – Heinong 37, Yuh-30; large – 2947 564/84, Vinni, DSS 2504. The following is a sign of the intensity of leaf plate, which is divided by gradation into light, moderate and dark. Among our samples with a light degree of color, we can distinguish Diona, Sribna Ruta, and Zuma. For varieties with moderate gradation of color are selected: Perlyna, 93/99, Knyazhna. To the dark color of the leaf plate we attributed the varieties: Stepnaya-90, Ozzie. Among the varieties we studied of the basics of flower color 37% varieties are with a white flower, and 63% with a purple. According to the varieties of standards: Chandor, Cresir, Toreador to the white color of the flower we attributed varieties Knyazhna, Ozzie, Zolotysta. With the purple color of the flower we have selected varieties: 93/99, Zhemchuzhna, Heinong 37. On the basis of the intensity of brown color bean, which characterizes the generative organs of the plant, is divided into weak, moderate and strong, but the other identifiers there are gradations: sandy, gray, light brown, brown and dark brown, so we decided what would be better identify by color gradation. To the sandy color of the beans we attributed varieties Perlyna, Yuh-30; gray – Sribna Ruta; light brown – Kobza, Knyazhna; brown – Ksenya; dark brown – Zhemchuzhna, Vinni. Important features that identify the description of varieties include basic color of the seed coat, which affects the taste of the seeds. In particular, the seeds are divided by color into yellow – Kobza, Yuh-30; yellow-green – Vinni, Luch Nadezhdy; green – Heorhina; light brown – 2947 564/84, brown – DSS 2504; dark brown – 2974 YS-24 and black color – not detected. Following the results of the previous collection of soybean varieties from the morphological sings of vegetative, generative organs of plants, it was possible to identify varieties with stable manifestation of standard identifying sings of soybean. Collection varieties with one standard characteristics: Heinong 37, Luch Nadezhdy, DSS 2504, 2974 YS-24; two signs: Zolotysta, Diona, Stepnaya-90, Zuma, Ksenya, Heorhina, Sribna Ruta, Perlyna, 93/99, Zhemchuzhna; three signs: Ozzie, 2947 564/84, Yuh-30, Vinni, Knyazhna.

Anthocyanins are vital components of plant secondary metabolites, and are also the most important coloring substances in wine. Teinturier cultivars are rich in anthocyanins. However, the differences in anthocyanin accumulation and profiles between teinturier and non-teinturier cultivars have not been reported. In this study, Yan 73 and Dunkelfelder were selected as the experimental materials, and three non-teinturier cultivars were used for comparison. LC-MS and qRT-PCR were used to determine the individual anthocyanin contents and the relative gene expression. The results show that the total anthocyanin content of the teinturier cultivars was considerably higher than that in non-teinturier cultivars, and the levels of individual anthocyanins increased gradually during ripening. Lower ratios of modified anthocyanins were found in the teinturier cultivars, which was not only due to the high expression level of VvUFGT and VvGST4, but also due to the relatively low expression of VvOMT in these cultivars. Cluster analysis of gene expression and anthocyanin accumulation showed that VvUFGT is related to anthocyanin accumulation, and that AM1 is related to the synthesis and transport of methylated anthocyanins. Our results will be useful for further clarifying the pathways of anthocyanin synthesis, modification, and transport in teinturier cultivars.

Studies on the use of silicate correctives in agriculture show that they have great potential to improve soil chemical characteristics, however, little information is available on the reactivity rates of their particle-size fractions. This study investigated whether the reactivity rates obtained experimentally could be considered in the calculation of ECC (effective calcium carbonate) for soil liming, promoting adequate development of alfalfa plants. Six treatments were evaluated in the experiment, consisting of two slag types applied in two rates. The experimental ECC was used to calculate one of the rates and the ECC determined in the laboratory was used to calculate the other. Rates of limestone and wollastonite were based on the ECC determined in laboratory. The rates of each soil acidity corretive were calculated to increase the base saturation to 80%. The treatments were applied to a Rhodic Hapludox and an Alfisol Ferrudalfs. The methods for ECC determination established for lime can be applied to steel slag. The application of slag corrected soil acidity with consequent accumulation of Ca, P, and Si in alfalfa, favoring DM production.

Plant derived products are not just the part of our food but also it contains many phytochemicals that possess many medicinal properties to tackle various ailments. Studies on the phytochemicals in the food sources might lead to the discovery of more safe and reliable drugs. So keeping it in mind, present study aims on exploring the potential antibacterial, antioxidant and anticancer potential of the seed coat extract of Vigna radiata. Seeds being very rich source of easily digestible proteins are part of vegetarian diet. Preliminary study on phytochemicals showed the significant differences in the composition of the seed coats and seeds without seed coat. Methanolic extract of seed coat showed good antibacterial activity against both Gram positive and negative bacteria with zones of inhibition ranging from 25-28.5 mm in diameter at concentration of 10 mg/ml. The extract also exhibited good antioxidant potential when tested with two different methods that are DPPH (2,2-diphényl-1-picrylhydrazyl) radical scavenging activity and FRAP (Ferric Reducing Antioxidant Power Assay) method showing activity of 80% and 77% respectively at the concentration of 100 µg/ml. For anticancer activity checked by SRB assay against the human lung cancer cell line HOP-62, the extract at the concentration of 80 µg/ml showed 65% cell growth inhibition activity. The work concludes that seeds must be consumed with seed coat which has medicinal values due to rich antioxidant, antibacterial activity and anticancer potential thereby it could act as raw source of drugs for treating various ailments.

The effect of some respiratory inhibitors on light-induced Paulownia tomentosa Steud. seed germination was studied. Millimolar solution of sodium azide was sufficient to completely prevent germination induced by a 5-min red light pulse. The inhibitory effect of azide was absent if seeds were rinsed before phytochrome activation by light. Sodium azide was effective only if present in the period of Pfr activity. The escape time from azide inhibition compared to the escape from far-red light action, was delayed for about 24 hours. When azide was applied after phytochrome activation, its effect depended on how long it was present in the incubation medium. The removal of azide allowed full restoration of germination by another red light pulse and the far-red escape time did not differ from the escape of untreated, i.e. water-imbibed seeds. Potassium cyanide alone did not produce any effect in light-stimulated germination of these seeds. However, it counteracted the inhibitory effect of azide in light-stimulated germination, if applied simultaneously at a concentration three times higher.

This study aimed at developing parameters of the Diagnosis and Recommendation Integrated System (DRIS) model for the assessment nutrient status for cotton grown in Benin.. Soil physical and chemical characteristics, leaves nutrient content (N, P, K, Ca, Mg) and seed-cotton yield were assessed on samples gathered from 150 farmers’ fields in 2018. Nutrient indices were computed using standard DRIS procedures. Results showed that phosphorus was in excess in the petiole and the whole leaves but in deficit in the limb. Potassium content was adequate according in the petiole and leaves but deficient in the limb. Ca content was limiting in the limb or the whole leaves and adequate in the petiole. Based on the diagnosis made in the petioles, Mg was deficient while adequate in the limbs and leaves. In the limb, the order of the macronutrients is as follows: K> P> N. On the other side, in the petiole and the whole leaves, the order of the macronutrients becomes: P> K> N. In the whole leaves and limb, the order of the secondary elements is as follows: Mg> Ca, whereas in the petiole the order of the secondary elements becomes: Ca> Mg.
