Conelet abortion and ATP levels in longleaf pine

During the past 5 years, an average of 87% of the 1st year strobili in a longleaf pine seed orchard in the Sand hills region of North Carolina have aborted development. Fresh weight, dry weight, ATP, and morphological characteristics were measured periodically after anthesis to determine some of the physiological changes associated with conelet abortion. In the early stages of conelet abortion, morphologically characterized by a slight browning in the pith area of the conelet, ATP levels almost doubled. As the conelet became more necrotic, ATP levels dropped. Sausage casing, which was used to shield strobili during the pollination period, was found to have a greenhouse effect on the conelets, hastening their development. Both the ATP burst and the beginning of conelet abortion was 1–2 weeks earlier for the encased conelets.

Morphological and physiological changes were investigated during flower growth and opening to extend the vase life of cut lotus flowers. When holding the flowers in distilled water, the edge of petals and the top edge of petals became black, and the stems were bent after 17 hours. Flowers fully opened and senesced after 25 hours. At full opening flower stage, there were decreases in fresh weight, and content of starch, auxin and zeatin, and increases in dry weight, and content of total sugar, anthocyanins and flavonols, ABA and gibberellin. Petal extract showed the presence two absorption peaks at 354 nm ( due to flavonols) and 535 nm (due to anthocyanins). Among the treatments, the combination of wilted flower (5 % fresh weight) and spraying of 2 mg/L NAA and 10 % coconut water (with 0,1 % Tween 20) gave a 2 days (43,22 hours) longer for cut lotus flowers than the control.

Conelet abortion in longleaf pine (Pinuspalustris L.) ranged from 85 to 100% in 4 of 5 years in a clonal seed orchard in southeastern North Carolina. Investigation of the cause of the malady was inconclusive, but freeze damage, seed bugs, and inadequate pollen supply were excluded as principal causes. Where combinations of balanced fertilizers and ammonium nitrate had been used in the orchard, abortion was variable by clone, with some clones showing more abortion under higher fertilizer regimes.

Various structural and physiological changes in squash cotyledons were followed as a function of germination time. Temperature, to which squash plants are very sensitive, was kept constant at 31 °C but light conditions were varied.In light-germinated plants (16 h light/day) the cotyledons changed from storage organs to greatly enlarged photosynthetic leaves, within 5 days. Incorporation of major amounts of thymidine-2-14C throughout the cotyledons occurred only during the enlargement phase.Dark-germinated plants, in which the cotyledons enlarged very little, produced a long, stout hypocotyl. No major incorporation of thymidine-2-14C into the cotyledons occurred, suggesting that little cell division occurs in the cotyledons of dark-germinated squash plants.Fresh- and dry-weight changes in the cotyledons were consistent with the changes in morphology of light- and dark-germinated plants. The cotyledons of plants which were dark-germinated for 6 days and then placed in 16 h light per day did not reach the fresh weight per cotyledon or dry weight per cotyledon level attained by cotyledons from plants germinated, from the beginning in 16 h light per day.

Barringtonia racemosa L. has many medicinal properties especially the fruit and leaf parts. The fruits are used to relief pain and inflammation, the leaves were proved to control high blood pressure whereas the roots barks are effective to treat chicken pox. Due to its medicinal importance for human health, it is essential to conserve this plant. A comparative study of different types of media was performed to study its effect on callus growth of endosperm explant from B. racemosa. Types of basal media studied including Murashige and Skoog (MS), Lloyd and McCown Woody Plant (WPM) and Gamborg (B5). The endosperm explant of B. racemosa were cultured on different basal MS, WPM and B5 media supplemented with 1.0 mg/L 2,4-D and 1.5 mg/L kinetin. The observation of callus growth and morphological characteristics of callus were done on weekly basis within 4 weeks. After 4 weeks of incubation period, the maximum fresh weight (0.300 ± 0.027g) and dry weight (0.025 ± 0.003g) were recorded from the explants cultured on MS medium followed by WPM and B5 media supplemented with 1.0 mg/L 2,4-D and 1.5 mg/L kinetin. The textures of callus produced from MS were nodular in shape and creamy colour. In conclusion, all the media studied had successfully induced the callus growth of B. racemosa with the present of 1.0 mg/L 2,4-D and 1.5 mg/L kinetin.

The present study was taken up to know the morphological and germination physiognomies of karonda (Carissa carandas) seeds influenced by seed storage period at ambient conditions (26 ⁰C temperature and 55 % RH). Seeds were subjected to store at 10, 20, 30, 40, 50 and 60 days at room temperature (26⁰C). Germination percentage and various morphological characteristics viz., vigour index, fresh weight of shoot and root, dry weight of shoot and root, root to shoot ratio were recorded. Among stored seeds 10 days old seeds recorded highest germina-tion percentage (57 %) and 60 days old seeds recorded lowest germination (20.33 %). At the end (90 days after sowing) 10 days old seeds again possessed higher fresh weight of root (0.300 g) and shoot (1.240 g), dry weight of root (0.103 g) and shoot (0.487 g), root to shoot ratio (0.212) and vigour index (1772.70 cm) among stored seeds. These physiological observations were quite similar with the freshly harvested seed which found maximum values for all the parameters owing to higher moisture content. Karonda seeds showed good viability upto10 days thereafter its value declined and it reached minimum after 60 days of storage.

ABSTRACT The objective of this work was to morphologically characterize 15 banana cultivars and assess the physical and chemical characteristics of their fruits at two maturation stages, unripe (pre-climacteric) and ripening. The plants were evaluated regarding their pseudostem height and diameter, petiole length, leaf blade length, width and length-to-width ratio. The cultivar Ouro had fruits with lower diameter, total length, market weight and fresh weight at both stages, and also firmer pulp when they were unripe. The cultivar Caru-Roxa had higher fresh fruit and pulp weights, and the cultivar Terrinha had the highest percentage of pulp dry weight percentage in unripe and ripe fruits. The cultivars Maçã and Ouro had higher pulp-to-peel ratio in unripe fruits. The ripe peels had lower fresh weight and thickness and higher dry weight percentage compared to unripe peels. The fruit peel of the cultivar Marmelo had the highest fresh weight at both stages. The cultivars Marmelo and Maçã had higher percentage of peel dry weight percentage at both stages. The unripe pulp had lower soluble solids. The titratable acidity in the pulp increased with ripening. The average plant height ranged from 2.25 to 6.15 m. The cultivars that had the largest pseudostem diameters had also the highest heights, except the Prata-Anã and Prata-Graúda. The cultivar and maturity stage influenced all the characteristics evaluated in fruits, except the total and market lengths, which did not vary with the ripening of fruits.

Putting out of using large areas of agricultural lands in the central region over the past years has led to worsening the prospects of their purposed use, although the problem of the relevance of their restoration still remains. For many years the unused land was exposed to both natural exogenous processes such as erosion, suffusion, etc. and biological and chemical changes, usually for the worse for agriculture. There are considered elements of monitoring aimed at assessing the prospects or lack of perspectives of rehabilitation of degraded lands. An energy approach to assessing the state of slopes and soils located within these slopes is presented. The main factors of natural and anthropogenic character in assessing the prospects for land restoration are their steepness, excess relative to local bases of erosion other morphological characteristics of slopes which in general is reduced to an assessment of the energy provision of slopes and soils. So the higher the energy capacity of slopes – they are less promising for development, for soils – there is a reverse picture – the higher their energy reserves, the more promising is their use. Approaches to zoning the territory for monitoring from larger taxons of natural and anthropogenic genesis to the sites of special surveillance within which the prospects for rehabilitation of the agricultural land are evaluated. The most important factor is the material expediency of such actions, i.e. before starting the restoration work it is necessary to assess the profitability or loss of the proposed event. In cases of the material expediency it is feasible as further actions to include energy assessments of slopes and soils; zoning of the object according to the steepness and oriented characteristics of soil washout; and the possibility of obtaining agronomic and meteorological data on a timely basis. The result of the work is a forecast assessment of the prospects for restoring degraded land for the intended purpose using modern databases and WEB-systems.

Cadmium (Cd) is a hazardous heavy metal, toxic to our ecosystem even at low concentrations. Cd stress negatively affects plant growth and development by triggering oxidative stress. Limited information is available on the role of iron (Fe) in ameliorating Cd stress tolerance in legumes. This study assessed the effect of Cd stress in two lentil (Lens culinaris Medik.) varieties differing in seed Fe concentration (L4717 (Fe-biofortified) and JL3) under controlled conditions. Six biochemical traits, five growth parameters, and Cd uptake were recorded at the seedling stage (21 days after sowing) in the studied genotypes grown under controlled conditions at two levels (100 μM and 200 μM) of cadmium chloride (CdCl2). The studied traits revealed significant genotype, treatment, and genotype × treatment interactions. Cd-induced oxidative damage led to the accumulation of hydrogen peroxide (H2O2) and malondialdehyde in both genotypes. JL3 accumulated 77.1% more H2O2 and 75% more lipid peroxidation products than L4717 at the high Cd level. Antioxidant enzyme activities increased in response to Cd stress, with significant genotype, treatment, and genotype × treatment interactions (p < 0.01). L4717 had remarkably higher catalase (40.5%), peroxidase (43.9%), superoxide dismutase (31.7%), and glutathione reductase (47.3%) activities than JL3 under high Cd conditions. In addition, L4717 sustained better growth in terms of fresh weight and dry weight than JL3 under stress. JL3 exhibited high Cd uptake (14.87 mg g−1 fresh weight) compared to L4717 (7.32 mg g−1 fresh weight). The study concluded that the Fe-biofortified lentil genotype L4717 exhibited Cd tolerance by inciting an efficient antioxidative response to Cd toxicity. Further studies are required to elucidate the possibility of seed Fe content as a surrogacy trait for Cd tolerance.

Abstract Introduction Colorectal carcinoma (CRC) is the most common gastrointestinal neoplasm in the world, accounting for 15% of cancer-related deaths. This condition is related to different molecular pathways, among them the recently described serrated pathway, whose characteristic entities, serrated lesions, have undergone important changes in their names and diagnostic criteria in the past thirty years. The multiplicity of denominations and criteria over the last years may be responsible for the low interobserver concordance (IOC) described in the literature. Objectives The present study aims to describe the evolution in classification of serrated lesions, based on the last three publications of the World Health Organization (WHO) and the reproducibility of these criteria by pathologists, based on the evaluation of the IOC. Methods A search was conducted in the PubMed, ResearchGate and Portal Capes databases, with the following terms: sessile serrated lesion; serrated lesions; serrated adenoma; interobserver concordance; and reproducibility. Articles published since 1990 were researched. Results and Discussion The classification of serrated lesions in the past thirty years showed different denominations and diagnostic criteria. The reproducibility and IOC of these criteria in the literature, based on the kappa coefficient, varied in most studies, from very poor to moderate. Conclusions Interobserver concordance and the reproducibility of microscopic criteria may represent a limitation for the diagnosis and appropriate management of these lesions. It is necessary to investigate diagnostic tools to improve the performance of the pathologist's evaluation, for better concordance, and, consequently, adequate diagnosis and treatment.

Abstract Background Orobanche crenata is an obligate root parasite belonging to Orbanchaceae. Broomrape causes great damage to the faba bean. Several attempts were applied for controlling parasitic weeds. So, the aim of this work is to study the application of Trichoderma spp. as well as three rhizobacteria species in comparison to herbicidal effect of Glyphosate (Glialka 48% WSC) for controlling broomrape infesting faba bean (Vicia faba). Materials and methods Three pot experiments were carried out in the greenhouse of the National Research Centre, Dokki, Giza, Egypt during two successive winter seasons. Trichoderma inocula were adjusted to 3.6 × 108 propagules/ml and the bacterium inocula were adjusted at 107–109 colony-forming unit (CFU)/ml. All treatments were applied, before 1 week of sowing, at rate of 50 ml per pot in experiments I and II, while 100 ml per pot in experiment III. Results Trichoderma spp. (T. harzianum, T. viride and T. vierns) as well as three rhizobacteria species (Pseudomonas fluorescens, Bacillus subtilis and Bacillus pumilus) enhanced the growth parameters in faba bean plants, i.e. shoot length, shoot fresh weight, shoot dry weight and leaf number in the first experiment when applied without O. crenata infection. In the second experiment, all bio-control could protect plants against O. crenata infection, where it had better juvenile number reduction, than glyphosate after 2 months of application. Both B. subtilis and B. pumilus had the highest reduction to juvenile fresh weight, while their effect was equal to herbicide for juvenile dry weight, respectively. The bio-control agents had high effects until the 4th month, but it was less than that of the herbicide. In experiment III, the bio-control agents could highly reduce the juvenile parameters after 2 months, as well as juvenile fresh weight and juvenile dry weight after 4 months, than the herbicide, respectively. The bio-control agents were effective until 6 months, but less than the herbicide effect. All bio-control treatments highly increased the plant growth parameters, than the herbicide. Conclusion The application of Trichoderma spp. as well as rhizobacteria species could play an important role in controlling broomrape in faba bean as a natural bioherbicide.
