A New Method of Blunt Cartilage Impact for a Model of Osteoarthritis

Impact injury to cartilage, a common consequence of joint trauma, is commonly thought to lead to degenerative osteoarthritis. Many pharmacologic agents have been developed to prevent or forestall this process. Development of a reliable model for the early stages of cartilage injury and osteoarthritis would enable the testing of novel treatments that could delay or even reverse the progression of this debilitating disease.

Abstract Ergothioneine is a natural and safe antioxidant playing an important role in anti-aging and the prevention of various diseases. Mushrooms are the main dietary source of ergothioneine. This study aims to report a kind of medicinal mushroom Panus conchatus with great potential for the bioproduction of ergothioneine. Molasses and soy peptone could promote cell growth of Panus conchatus and enhance the accumulation of ergothioneine. Meanwhile, three precursors of cysteine, histidine and methionine were added to the broth, and the highest ergothioneine concentration of 148.79mg/L was obtained when adding 0.4 g/L cysteine. Finally, the crude ergothioneine extract was purified and further evaluated. The ergothioneine from Panus conchatus showed higher antioxidant activity than vitamin C and glutathione with good stability at lower pH environment. This research would provide a new method for the bioproduction of ergothioneine and lay foundation for the in-depth study about the exploration of natural products from Panus conchatus.

Aseptic necrosis of the femoral head refers to the number of severe, rapidly progressing destructive-dystrophic hip joint lesions. In the overwhelming majority of cases, the disease develops in young and most active age group. In the absence of necessary treatment, the result of the disease is the early disability retirement with the need for radical reconstructive interventions on the joint. The use of modern diagnostic methods makes it possible to detect the beginning of the pathological process in the femoral head in the early stages and to conduct the most effective treatment of patients of this category. Aim - to improve the results of treatment of patients with early stages of aseptic necrosis of the femoral head due to the development and introduction into clinical practice of a new method of combined impaction autoplasty of the femoral head. Materials and methods. A new method of combined impaction autoplasty of the femoral head was used, including decompression, intraosseous resection and excision of the focus of osteonecrosis followed by combined autoplasty of the postresection defect (RF patent No. 2583577 dated 13.04.2016). Intraosseous resection of the focus of destruction of the femoral head was performed with the help of a specially developed device - milling cutters for bone tissue resection (RF patent for utility model No. 171951 dated 21.06.2017). Evaluation of the effectiveness of the treatment was carried out on the basis of the severity of the pain syndrome and functional capabilities of patients defined by the Harris scale (Harris Hip Score). Results. The analysis of the medium-term results of the conducted treatment showed that good results were obtained in 67.4% of cases in which the proposed method was used.

Asthma is a heterogeneous disease characterized by chronic airway inflammation that results in a wide spectrum of clinical manifestations. Patients with severe asthma represent a substantial share of consumption of healthcare resources and hospitalization. Moreover, these patients are at risk of increased morbidity and mortality. Recently, several phenotypes and endotypes of asthma have been identified. The identification of specific subtypes of asthma is fundamental for optimizing the clinical benefit of novel treatments. Although in most patients the disease can be controlled by some combination of pharmacologic agents, in some 5-10% of patients the disease remains uncontrolled. Several monoclonal antibodies (mAbs) targeting pathogenetic molecules (e.g., IgE, IL-5, IL- 5Rα, IL-4, IL-13, TSLP) are currently available or under development for the treatment of different forms of severe type 2 asthma. The identification of diagnostic and predictive biomarkers (e.g., IgE, blood eosinophil count, FeNO, periostin, etc.) has revolutioned the field of targeted therapy in severe asthma. Monoclonal antibodies targeting Th2-driven inflammation are generally safe in adult patients with moderate-to-severe asthma. The long-term safety of these biologics is a relevant issue that should be addressed. Unfortunately, little is known about non-type 2 asthma. Further studies are needed to identify biomarkers to guide targeted therapies of different forms of non-type 2 asthma.

A new method for sampling meat surfaces using abrasive discs has been tested against the spray gun technique already described by Clark (1965). Evenly inoculated beef and pork slices were sampled after 1, 48, and 120 h of incubation at 4 °C for beef and 1, 48, and 72 h at 4 °C for pork. The new method was slightly less efficient than the spray gun in recovering bacteria during the early stages of incubation but was about 600% more efficient after 72 and 120 h.
