The present on-farm research evaluated the occurrence of fatty liver syndrome and its predisposing risk factors for multiparous dairy cows from a commercial herd in Venezuela. Liver biopsy samples were collected at 35 days (d) prepartum (Holstein, ; Holstein × Carora crossbred, ) as well as 1 to 7 d (Holstein, ; Holstein × Carora crossbred, ) and 28 to 35 d (Holstein, ; Holstein × Carora crossbred, ) postpartum in order to analyse hepatic triacylglycerols (TAG, % wet basis) and glycogen concentrations. At postpartum, an occurrence of 72.0% for severe fatty liver along with 73.5% of subclinical ketosis (SCK) was found. The multiple regression model that best explained the association between milk production in the previous lactation (MYP) and TAG at first week postpartum was as follows: TAG, % = −11.2 + 3.16 (prepartum body condition) + 0.0009176 (MYP) (, ). Logistic regression indicated that Holstein × Carora crossbred cows tended to have 27% higher relative risk than Holstein to experience SCK, whereas prepartum liver TAG greater than 3% tended to be associated with a higher relative risk for SCK compared to cows with TAG ≤3%.

The aim of this study was to investigate whether season, lactation number, breed standard and milk production were risk factors relating to occurrences of gastrointestinal nematodes in dairy cows during the peripartum period. Eighty-four cows were randomly selected through proportional stratified sampling. In order to analyze the fecal egg per gram (EPG) count, the data were subjected to the Spearman test, Kruskal-Wallis test at 5% significance and linear regression. At the time of calving, the cows showed high EPG counts in relation to all variables analyzed. Among the animals studied, we observed that purebred Holstein cows at their first lactation and with high milk production showed high EPG counts (600) and comprised the group most at risk within the herd studied. In this group, the animals showed moderate EPG during the prepartum period (300) and a significant increase (p < 0.01) in EPG count from the time of calving (900), i.e. an increase of the order of 300%. Selection of animals for milk production in tropical countries should be based not only on productive potential, but also on adaptive features.

Identification of risk factors is important for the design of control programmes for mastitis in cows. Information about farms and management was collected at a farm visit. California Mastitis Test (CMT) was performed to assess sub-clinical mastitis, and cows, udder and milk were examined for clinical mastitis. A total of 347 lactating cows from 83 farms in the dry season (November - February) and 388 lactating cows from 89 farms in the wet season (June - October) were studied. The overall prevalence of mastitis was 19.9% and 44.8% in dry and wet seasons, respectively. The prevalence of mild mastitis was 17.3% and 40.7%, whereas that of moderate mastitis was 2.6% and 4.1% in dry and wet seasons, respectively. The prevalence of mastitis was higher (P<0.01) in wet than in dry season. On average, 18.7% quarters had mastitis during the wet season and 6.9% in the dry season. In the dry and wet seasons, respectively, 63.9% and 11.2% had completely dry floors, and the prevalence of mastitis was 22.6% and 30.0%. On the other hand, 88.8% and 36.1% of 83 farms had partly or completely wet and soiled floor and the prevalence of mastitis was 40.0% and 59.5% in the dry and wet seasons, respectively. Udder cleanliness, milk yield and peri-parturient diseases significantly (P<0.01) increased the risk of mastitis. The prevalence of mastitis is regarded as quite high. Negative energy balance (NEB) is a common pathological cause of ketosis. As the major organs of lipid metabolism, the liver and fat tissue take part in regulating lipid oxidative capacity and energy demands, which is also a key metabolic pathway that regulates NEB development during the perinatal period. Fibroblast Growth Factor 21 (FGF21) is a novel metabolic regulator involved in the control of fatty acid oxidation and lipid metabolism during a prolonged negative energy balance. Our study determined a correlation between serum FGF21 and β-hydroxybutyric acid (BHBA) levels in dairy cows with ketosis. We used sixty cows with low milk yield, abnormal glucose metabolism, and ketosis. Serum FGF21 and BHBA levels were measured using commercial kits. Serum FGF21 increased with increasing BHBA levels up to 1.6 mmol/L. At BHBA levels > 1.6 mmol/L, FGF21 decreased. Serum FGF21 levels were positively associated with BHBA levels, particularly in dairy cows with subclinical ketosis (r = 0.647, P < 0.01). At BHBA levels between 1.2 mmol/L and 1.6 mmol/L, FGF21 was more closely correlated with BHBA than with other metabolic parameters. At BHBA levels > 1.6 mmol/L, the association between FGF21 and BHBA was not significant. In conclusion, our results show that FGF21 was closely related with SK in cows. FGF21 may be a promising regulator in the prevention of subclinical ketosis.

The aim of the study was to evaluate the usefulness of the Precision Xceed® hand-held meter as an on-site method for determining blood ß-hydroxyboutyric acid (BHBA) and glucose concentrations, for the diagnosis of subclinical ketosis in dry and lactating dairy cows. A total of 163 clinically healthy Holstein cows (113 lactating, 8-50 days-in-milk; and 50 dry, 10-40 days pre-partum) from 5 farms located around Thessaloniki region, were blood-sampled once, from the jugular vein of each animal, 5 to 8 hours after the start of morning feeding. BHBA was determined in all 163 cows, whereas glucose only in 114 cows (50 dry and 64 lactating cows). These analyses were performed, for each cow, by both laboratory method (in serum) and Precision Xceed® meter (in whole blood, cowside). Using laboratory serum BHBA concentrations > 1.2 mmol/L as the cut-off point, 11/163 (6.7%) of the tested cows were considered as subclinically ketotic, whereas raising the cut-off to > 1.4 mmol/L, 9/163 (5.5%) cows had subclinical ketosis. All these cows (11 and 9, respectively) were lactating. None of the dry cows had subclinical ketosis at BHBA cut-off of > 1.4 mmol/L. One out of the 50 dry cows (2%) and 15/113 (13.3%) lactating cows sampled were classified as subclinically ketotic when the Precision Xceed® meter was set at BHBA concentrations > 1.2 mmol/L. Overall, mean BHBA and glucose concentrations were not statistically different (P>0.05) between the two methods. Significant positive correlations were found for BHBA (strong correlation: r=0.99; n=163; P<0.01) and glucose (moderate correlation: r=0.63; n=114; P<0.01) concentrations between Precision Xceed® and laboratory results. Precision Xceed® is less accurate for measuring glucose (glucometer) compared to BHBA (ketometer). The low percentage of false positive (<0.6%) and false negative (<4%) indicating that the Precision Xceed® meter is an accurate screening test and its results are highly reliable under field conditions. Precision Xceed® meter was highly sensitive (90.9%) and specific (96.05%) at cut off point of BHBA concentrations > 1.2 mmol/L and it had excellent test agreement for detection of subclinical ketosis when using a threshold of blood BHBA > 1.4 mmol/L.

SUMMARYA 3-year longitudinal study was conducted on a multi-site farrow-to-finish production system. For each of 18 cohorts at three finishing sites, 50 pigs were randomly selected. Faecal samples were collected every 2 weeks for 16 weeks. Salmonella was cultured from 453 (6·6%) of 6836 faecal samples. The pig-level incidence of Salmonella was 20·8% (187/899 pigs). Salmonella prevalence varied between cohorts and within pigs. The adjusted Salmonella prevalence decreased over the finishing period from 6·4% to 0·8%. Intermittent detection of Salmonella was found in more than 50% of pigs that were positive at more than one collection. The finding that the majority of pigs shed intermittently has implications for surveillance and research study design when determining Salmonella status. The variability in shedding over time, as well as between and within cohorts and pigs suggests that there may be time-variant risk factors for Salmonella shedding in swine.
