The present study was performed in order to evaluate, by the autoradiographic technique the sealing properties of three root canal sealers, combined with gutta - percha and silver points, in vitro. A total of one hundred and five extracted human, single root teeth were used and divided into five groups according to the filling combinations tested. The teeth were filled, using two techniques, of the single and multiple points. Autoradiographies, using S35, in all instances, were performed and the results obtained were compared with the respective dental radiograms The obtained results lead to the following conclusions: 1. Silver and gutta - percha points may be always combined with sealers. Otherwise the root canal is not adequately sealed. 2. A gutta - percha point, without sealer, produced a superior filling to silver point. 3. It seems possible to obtain a complete obturation of the rootcanal with the combination of gutta - percha and Grosman’s Sealer using the multiple - point technique. 4. It follows in effectivness, the combination of silver point and Grossman’s sealer.5. The combination of gutta - percha point with Zinc - oxide eugenol sealer and chloropercha are the least efficient. 6. Finally it must be stressed that since the conditions of the present study do not correspond exactly to those encountered in vivo, extrapolation of the results obtained, to man has to be performed with caution.

The aim of this ex vivo was to investigate if two radiopaque root canal sealers with different formulations would influence the radiographic perception of root canal fillings. The root canals of 48 extracted maxillary canines were prepared and randomly assigned to 3 groups of 16 specimens each. In each group, the root canals were filled by lateral condensation of gutta-percha and one of the tested sealers: Endométhasone, Sealer 26, or a non-radiopaque sealer. A through-and-through void was simulated in half of the specimens from each group (n=8). The buccolingual radiographic images obtained were randomly interpreted for voids existence by a radiologist and an endodontist. The differences in sensitivity and specificity between groups and examiners were compared using, respectively, Fisher's Exact and McNemar tests at 5% significance level. Both radiopaque sealers caused a significant decrease in sensitivity at the coronal part of fillings. The use of Endométhasone increased specificity values for both coronal and apical portions of the root canal fillings. In conclusion, the tested sealers influenced the radiographic perceptions of laterally condensed root canal fillings in a different way.

Objective. The objective of this study was to evaluate the efficacy of two retreatment NiTi systems (protaper universal retreatment files, R-Endo), when compared to manual technique in removing Gutta-percha obturated with two sealers. Study Design. Sixty extracted single-rooted premolars were instrumented with Protaper rotary files till F3. The specimens were divided into six groups. Groups 1, 2, 3 were obturated with Gutta-percha and zinc oxide eugenol and Groups 4, 5, 6 were obturated with Gutta-percha and AH-plus. The retreatment was carried out in groups 1 and 4 with H-files and GGdrills, groups 2 and 5 with R-endo retreatment files and groups 3 and 6 with Protaper retreatment files. The roots were sectioned and evaluated under optical stereomicroscope. Statistical analysis was performed with one-way ANOVA and Newman-Keul's test at . Results. The manual technique resulted in cleaner canal walls when compared with both rotary retreatment systems. Conclusion. NiTi rotary retreatment files can be used to remove the filling material quickly, but it should be followed by hand instruments to obtain better canal wall cleanliness.

The aim of this study was to evaluate ex vivo the sealing achieved at simulated lateral canals (SLC) and the quality of filling according to their position in the root canal after using the same filling technique. SLC were created at three levels in 55 teeth and divided into two groups depending on the root canal sealer used (1: BioRoot® RCS, 2: GuttaFlow® bioseal). They filled them with the continuous wave technique and submitted to a diaphanization technique. The samples were analyzed using a magnifying lens (20×), pictures were taken, which proceeded to linear measurement with the ImageJ® program and used a filling score system with five grades (0 to 4, 0 and 1 not acceptable, 2 to 4 acceptable); BioRoot® RCS has got a greater proportion than GuttaFlow® bioseal for SLC filled acceptably at 10 mm from the apex (p < 0.05). The highest proportion of SLC filled acceptably was found in the middle third (6 mm) (p < 0.05), followed by the apical third (3 mm) and the coronal third (10 mm). The difference between apical and coronal third could be significant; BioRoot® RCS has been better than GuttaFlow® bioseal for filling SLC in the coronal third of the teeth. Studies on the characteristics of these cements are missing to explain these differences.

Proper research design, appropriate evaluation and measurement methods, use of correct statistical tests, interpretation, and inference are the essentials without which any scientific research reporting is incomplete and does not serve its proposed purpose. In this communication, some common flaws in research designing, evaluation, analysis, and inference have been explained using a published article as a reference. The purpose of the paper is to help the scientific community recognize the significance of proper research designing and planning in order to achieve the results which are worthwhile.

Abstract Background The extrusion of overfilled materials that extend beyond the apical foramina into the periradicular tissue may serve as a reservoir for bacterial adhesion and further affect recovery from periapical diseases. The aim of this study was to evaluate the effects of serum proteins on Enterococcus faecalis adhesion and survival on the surface of a calcium hydroxide-based root canal sealer (Apexit Plus), an epoxy resin sealer (AH-Plus) and a bioceramic sealer (iRoot SP). Methods Apexit Plus, AH-Plus and iRoot SP were evenly coated on gutta-percha, using gutta-percha alone as the control. After root canal sealer setting, the number of E. faecalis adhering to the root canal sealers and gutta-percha was counted in fetal bovine serum (FBS) or tryptic soy broth supplemented with 1% glucose (TSBG) by viable cell plate counts. The morphology of 7-day-old E. faecalis biofilms in FSB and TSBG was observed by scanning electron microscopy (SEM). Furthermore, E. faecalis biofilms on the three root canal sealers were labeled with a LIVE/DEAD BacLight™ Bacterial Viability Kit, and the ratios of viable to dead cells were analyzed using laser scanning microscopy operative software (Zen software). Results In the assays, after 1 and 7 days, the number of E. faecalis adhering to the root canal sealers or gutta-percha in FBS were significantly lower than those in TSBG (P < 0.05). In FBS, E. faecalis adhesion to iRoot SP and gutta-percha was reduced to a greater extent than that adhered to Apexit Plus and AH-Plus. Few E. faecalis accumulated on iRoot SP in FBS, whereas many bacteria assembled on iRoot SP and formed biofilms in TSBG. The ratio of viable cells in the E. faecalis biofilm on iRoot SP was the lowest. Conclusions Calcium hydroxide-based root canal sealers, epoxy resin sealers and bioceramic sealers may provide a substrate for E. faecalis adhesion, and the bioceramic sealer in this study showed the least E. faecalis adhesion in the presence of serum proteins compared to the other two sealers.
