AbstractThe grape berry moth Eupoecilia ambiguella (Lepidoptera: Tortricidae) is causing significant damage to grape berries, however, little is known on population genetics of this lepidopteran pest insect, hindered so far by the lack of suitable molecular markers. Here we report on the development of ten microsatellite markers of which six were used to characterise 21 E. ambiguella populations obtained from two viticultural regions in Germany and Italy. Moths were sampled during two subsequent generations (flights) in the same vineyard as well as in vineyards surrounded by different landscape types. German and Italian populations were genetically differentiated and a significant isolation by distance was evident. No significant divergence was observed among the populations from first or second flight moths, however, inbreeding was higher in first than in second flight populations. Moreover, inbreeding was influenced by habitat composition and complexity of landscape around vineyards, being positively associated with the percentage of area covered by grapevine. Population genetics of E. ambiguella could thus be affected by the presence of alternative host plants in viticultural landscapes, which is important in the light of both insecticide resistance management and sustainable pest management.

Erythrocytes treated with polysaccharide from Past. tularensis are specifically agglutinated by the sera of persons recovered from tularemia or vaccinated against it, and by the sera of animals immunized with living cultures. The serum hemagglutination titer is correlated with the amount of antibody as measured by the quantitative precipitin method with polysaccharide, but not with the bacterial agglutination titer. The meaning of the results is discussed.

There is an increasing appreciation of the importance of transgenerational effects on offspring fitness, including in relation to immune function and disease resistance. Here, we assess the impact of parental rearing density on offspring resistance to viral challenge in an insect species expressing density-dependent prophylaxis (DDP); i.e. the adaptive increase in resistance or tolerance to pathogen infection in response to crowding. We quantified survival rates in larvae of the cotton leafworm ( Spodoptera littoralis ) from either gregarious- or solitary-reared parents following challenge with the baculovirus S. littoralis nucleopolyhedrovirus. Larvae from both the parental and offspring generations exhibited DDP, with gregarious-reared larvae having higher survival rates post-challenge than solitary-reared larvae. Within each of these categories, however, survival following infection was lower in those larvae from gregarious-reared parents than those from solitary-reared, consistent with a transgenerational cost of DDP immune upregulation. This observation demonstrates that crowding influences lepidopteran disease resistance over multiple generations, with potential implications for the dynamics of host–pathogen interactions.

Abstract The fall armyworm, Spodoptera frugiperda, is a lepidopteran pest that feeds in large numbers on the leaves, stems and reproductive parts of more than 350 plant species, causing major damage to economically important cultivated grasses such as maize, rice, sorghum, sugarcane and wheat but also other vegetable crops and cotton. Native to the Americas, it has been repeatedly intercepted at quarantine in Europe and was first reported from Africa in 2016 where it caused significant damage to maize crops. In 2018, S. frugiperda was first reported from the Indian subcontinent (Ganiger et al., 2018; Sharanabasappa Kalleshwaraswamy et al., 2018). It has since invaded Bangladesh, Thailand, Myanmar, China and Sri Lanka (IPPC, 2018b, 2019; FAO, 2019c). The ideal climatic conditions for fall armyworm present in many parts of Africa and Asia, and the abundance of suitable host plants suggests the pest can produce several generations in a single season, and is likely to lead to the pest becoming endemic.

Objective: The current investigation involves the purification, characterization of the lectin from the leaves of Pimenta dioica (L.) Merr. (Myrtaceae) a medicinal plant, and its application in bacterial typing.Methods: A lectin was purified from the leaves by cation exchange chromatography. SDS PAGE revealed the molecular weight of the purified lectin. Biochemical characterization was carried out by performing various tests. Hemagglutination inhibition was conducted to detect the sugar specificity. Additionally, bacterial agglutination was performed to predict whether the purified lectin was able to agglutinate the bacterial strains.Results: SDS PAGE analysis revealed the lectin to be a tetramer in the range of 43-66 kDa. The purified lectin agglutinated human, avian, and mouse erythrocytes, and was inhibited by 125 mmol of mannose and xylose. The lectin was stable at 0-60 ° C for 30 min and was unaffected by either 2-Mercaptoethanol (2-ME) or Dithiothreitol (DTT) (50-250µM). A pH of 6.0–8.0 was found optimum for its activity and was nearly independent of metal ions. The purified lectin contained about 20% carbohydrate as estimated by Anthrone method. Purified lectin agglutinated the Gram-negative Escherichia coli and Proteus vulgaris.Conclusion: The isolated lectin was found to possess significant hemagglutinating activity. Due to its ability to agglutinate Gram negative bacteria such as Escherichia coli and Proteus vulgaris, it could be used for bacterial typing and for the design of bacterial filters.
