Effects of intramammary infusion of Bifidobacterium breve on mastitis pathogens and somatic cell response in quarters from dairy cows with chronic subclinical mastitis

Abstract Subclinical mastitis in dairy cows is a big economic loss for farmers. The monitoring of subclinical mastitis is usually performed through Somatic Cell Count (SCC) in farm but there is the need of new diagnostic systems able to quickly identify cows affected by subclinical infections of the udder. The aim of this study was to evaluate the potential application of thermographic imaging compared to SCC and bacteriological culture for infection detection in cow affected by subclinical mastitis and possibly to discriminate between different pathogens. In this study we evaluated the udder health status of 98 Holstein Friesian dairy cows with high SCC in 4 farms. From each cow a sample of milk was collected from all the functional quarters and submitted to bacteriological culture, SCC and Mycoplasma spp. culture. A thermographic image was taken from each functional udder quarter and nipple. Pearson’s correlations and Analysis of Variance were performed in order to evaluate the different diagnostic techniques. The most frequent pathogen isolated was Staphylococcus aureus followed by Coagulase Negative Staphylococci (CNS), Streptococcus uberis, Streptococcus agalactiae and others. The Somatic Cell Score (SCS) was able to discriminate (p<0.05) cows positive for a pathogen from cows negative at the bacteriological culture except for cows with infection caused by CNS. Infrared thermography was correlated to SCS (p<0.05) but was not able to discriminate between positive and negative cows. Thermographic imaging seems to be promising in evaluating the inflammation status of cows affected by subclinical mastitis but seems to have a poor diagnostic value.

ABSTRACT Infections with Chlamydophila abortus and C. pecorum are highly prevalent in cattle and have been associated with bovine mastitis. A prospective cohort study was conducted with a herd of 140 Holstein dairy cows to investigate the influence of Chlamydophila infection on subclinical inflammation of the bovine mammary gland as characterized by somatic cell numbers in milk. PCR detection of C. abortus and low serum antibody levels against Chlamydophila spp. were significantly associated with subclinical mastitis. To examine the effect of the infection by response modification, immune perturbation was done by two subcutaneous administrations of an experimental vaccine preparation of inactivated C. abortus and C. pecorum elementary bodies. Vaccination against Chlamydophila highly significantly decreased milk somatic cell numbers, thus reducing bovine mastitis, and increased antibody levels against Chlamydophila but did not eliminate shedding of C. abortus in milk as detected by PCR. The protective effect peaked at 11 weeks after vaccination and lasted for a total of 14 weeks. Vaccination with the Chlamydophila vaccine, a mock vaccine, or a combination vaccine against bovine viral diseases highly significantly increased C. abortus shedding in milk for 1 week, presumably mediated by the vaccine adjuvant. In summary, this study shows an etiological involvement of the widespread Chlamydophila infections in bovine mastitis, a herd disease of critical importance for the dairy industry. Furthermore, this investigation shows the potential for temporary improvement of chlamydial disease by therapeutic vaccination. Chlamydophila vaccination of cattle might serve as a testing ground for vaccines against human chlamydial infections.

Subclinical mastitis, diagnosed by elevated somatic cell count (SCC) in milk, is an important monitoring parameter of dairy cows’ udder health, related to their productivity and welfare. The present retrospective study aims to evaluate the epidemiology of subclinical mastitis (SCM) among the 37 herds of the Holstein Association of Greece participating in the milk quality recording system “ΙΩ”, from the start of 2015 until the end of 2018. The herds’ inclusion criterion was the consistency of monthly SCC recording throughout at least one full year between 2015 and 2018, with a maximum interval of 61 days between two consecutive monthly SCC recordings. Twenty-six herds (8630 cows) in 2015, thirty herds (10763 cows) in 2016, thirty herds (10945 cows) in 2017 and twenty-six herds (9597 cows) in 2018 were included. The prevalence of SCM and chronic SCM, the incidence rate of new cases of SCM, as well as the average somatic cell score and bulk tank milk SCC were determined for each of the four years. The results indicate a progressive deterioration of udder health from the onset of the cow’s productive life until culling. A year-over-year increase in the number of cows with subclinical mastitis led to an overall SCM prevalence of 34.5%, chronic SCM prevalence of 26.9% and a bulk tank milk SCC of 463000 cells/mL, in 2018. The average somatic cell score, a base 2logarithm of individual cow’s SCC, was found persistently above the subclinical mastitis indicative cut-off in all four years, with a peak in 2018. At herd level, the incidence rate of new SCM cases was 12 new cases / 100 cows / month; the highest incidence rate was observed in the early lactation stage group (1-60 days-in-milk), in all four years, reaching a peak of 31 new cases / 100 cows / month, in 2018. In 2018, prevalence of heifers’ SCM and chronic SCM was23.4% and 16.9%, respectively. Despite the adequate average 305-days milk yield (9608 kg in 2018), the results were indicative of poor udder health status, pointed out by reduced duration of cows’ productive life (less than 3 lactations)and lower milk quality (elevated SCC). The severity and wide spreading of subclinical mastitis in Greek dairy herds highlights the necessity of a national mastitis control program, aiming to improve the productive efficacy, management decisions accuracy and quality of produced milk.

Background and Aim: Subclinical mastitis (SCM) is one of the most prevalent diseases of dairy cows, and somatic cell count (SCC) is widely used to determine SCM and milk quality. However, setting the threshold of SCC is very important. This study aimed to determine the cow-level threshold of SCC to differentiate milk of SCM affected cows from normal milk in Bangladesh. Materials and Methods: Sensitivity (Se) and specificity (Sp) along with other characteristics of different thresholds of SCC were determined considering the bacterial culture as the gold standard test. Three definitions of intramammary infection (IMI) were set based on the group of pathogens involved. Five categories of SCC thresholds were considered for analysis. Results: Se and Sp of thresholds of SCC greatly varied as definitions of IMI changed. Irrespective of SCC thresholds, Se increased when isolation of major pathogens included in IMI definitions. Se decreased when SCC thresholds increased (from 100 to 300 × 103 cells/mL) for all IMI definitions and ranged from 60.6% to 20.3%. The highest Se was found at low SCC threshold (100 × 103 cells/mL), which resulted in less false-negative outcome. On the other hand, Sp increased with the increment of SCC thresholds giving rise to a less false-positive outcome. Conclusion: The cow-level SCC threshold of 100 × 103 cells/mL was found appropriate to identify subclinical IMI of dairy cows.

The aim of this study was to determine the occurrence of pathogens in selected group of ewes and the relationship between somatic cell count (SCC) and the presence of pathogens. The experiment was carried out on a dairy farm, where predominantly breed was a Tsigai. Sampling was carried out in monthly intervals as part of the milk recording test day from February to July 2019. A total of 303 ewes were included in the survey, during the milk recording test day. The ewes with SCC ≥1000 × 103 cells.mL-1 were selected for further sampling at half udder level. Based on SCC the ewes were divided into five groups: <200 ×103; ≥200 <400 × 103; ≥400 <600 × 103; ≥600 <1000 × 103; ≥1000 × 103 cells.mL-1. The first group of SCC contained 33.9% of milk samples, the second 14.1% of samples, the third 5.7% of samples, the fourth 6.2% and the fifth 40.1% of samples. The most common pathogens were coagulase negative staphylococci (CNS). The most frequent CNS was Staphylococcus (S.) simulans (24.4%). S. aureus was identified in 5.3% of bacteriological positive samples. Almost 70% of ewes with bacteriological positive samples were repeated identified the presence of pathogens during tested period. SCC ≥500 × 103 cells.mL-1  were detected in 92.5% bacteriological positive milk samples. The presence of pathogens increased SCC in milk (p <0.001) as compared to samples free of pathogens. In conclusion, the SCC ≥500 × 103 cells.mL-1 could be important for detection of subclinical mastitis at half udder level in dairy ewes.

This study aimed to determine the presence or absence of subclinical mastitis cases with mastitis test IPB-1 and somatic cell count calculation of the Breed method for dairy milk farm in Tanggamus Regency.  The study was conducted in January 2020. Milk specimens were taken from farmers in Gisting Atas and Air Naningan district areas of Tanggamus Regency, Lampung Province.  Specimen testing was carried out at the Animal Production Laboratory, Department of Animal Husbandry, Faculty of Agriculture, University of Lampung.  Specimens of milk used were obtained from farms in the Gisting Atas region as many as 9 lactation dairy cows and 10 lactation dairy cows from farms in the Air Naningan region.  This research method was a survey by taking farm data through interviews, taking milk specimens for mastitis testing with the reagents of IPB-1, taking specimens to be used as preparations which are then tested to somatic cell count using the breed method.  The variables observed were the results of mastitis test and the results of somatic cell counts in dairy milk.  Data were processed in tabulation and histogram then explained descriptively.  The mastitis test results showed that 33% of cow nipples from Gisting Atas farms and 37% of nipples from Air Naningan farms positively had subclinical mastitis.  Calculation of the number of somatic cells showed that the average number of somatic cells was 2,341,538 cells / ml for the Gisting Atas area and as much as 821,176 cells / ml for the Air Naningan region.    Keywords: Subclinical mastitis, IPB-1 test, Breed method, Somatic cells, dairy milk
