AbstractApproximately 150 triatomine species are known to be infected with the Chagas parasite, Trypanosoma cruzi, but they differ in the risk they pose to human populations. The largest risk comes from species that have a domestic life cycle and these species have been targeted by indoor residual spraying campaigns, which have been successful in many locations. It is now important to consider residual transmission that may be linked to persistent populations of dominant vectors, or to secondary or minor vectors. The aim of this project was to define the geographical distributions of the community of triatomine species in Latin America. Presence-only data with over 12, 000 observations of triatomine vectors were extracted from a public database and target-group background data were generated to account for sampling bias in the presence data. Geostatistical regression was then applied to estimate species distributions and fine-scale distribution maps were generated for thirty triatomine vector species. The results for Panstrongylus geniculatus, P. megistus, Triatoma barberi, T. brasiliensis, and T. pseudomaculata are presented in detail and the model validation results for each of the 30 species are presented in full. The predictive maps for all species are made publicly available so that they can be used to assess the communities of vectors present within different regions of the endemic zone. The maps are presented alongside key indicators for the capacity of each species to transmit T. cruzi to humans. These indicators include infection prevalence, evidence for human blood meals, and colonisation or invasion of homes. A summary of these indicators shows that the majority of the 30 species mapped by this study have the potential to transmit T. cruzi to humans.Author summaryThe Pan American Health Organisation’s Strategy and Plan of Action for Chagas Disease Prevention, Control and Care highlights the importance of eliminating those triatomine vector species that colonise homes, and has had great success in many locations. Since indoor residual spraying campaigns have targeted these species, their importance relative to other vectors has diminished and their geographical distributions may also have changed. It is now vital to consider the full community of vector species, including previously dominant vectors as well as secondary or minor vector species, in order to target residual transmission to humans. Our aim was to define the geographical distributions of the most commonly reported triatomine species in Latin America. We extracted reports of triatomine vector species observed at specific locations from a public database and we used a geostatistical model to generate fine-scale predictive maps for thirty triatomine vector species. We present these maps alongside a summary of key indicators related to the capacity of each species to transmit the Chagas parasite to humans. We show that most of the 30 species that we have mapped pose a potential threat to human populations.

Abstract Background Vector control through long-lasting insecticidal nets (LLINs) and focal indoor residual spraying (IRS) is a major component of the Tanzania national malaria control strategy. In mainland Tanzania, IRS has been conducted annually around Lake Victoria basin since 2007. Due to pyrethroid resistance in malaria vectors, use of pyrethroids for IRS was phased out and from 2014 to 2017 pirimiphos-methyl (Actellic® 300CS) was sprayed in regions of Kagera, Geita, Mwanza, and Mara. Entomological surveillance was conducted in 10 sprayed and 4 unsprayed sites to determine the impact of IRS on entomological indices related to malaria transmission risk. Methods WHO cone bioassays were conducted monthly on interior house walls to determine residual efficacy of pirimiphos-methyl CS. Indoor CDC light traps with or without bottle rotator were hung next to protected sleepers indoors and also set outdoors (unbaited) as a proxy measure for indoor and outdoor biting rate and time of biting. Prokopack aspirators were used indoors to capture resting malaria vectors. A sub-sample of Anopheles was tested by PCR to determine species identity and ELISA for sporozoite rate. Results Annual IRS with Actellic® 300CS from 2015 to 2017 was effective on sprayed walls for a mean of 7 months in cone bioassay. PCR of 2016 and 2017 samples showed vector populations were predominantly Anopheles arabiensis (58.1%, n = 4,403 IRS sites, 58%, n = 2,441 unsprayed sites). There was a greater proportion of Anopheles funestus sensu stricto in unsprayed sites (20.4%, n = 858) than in sprayed sites (7.9%, n = 595) and fewer Anopheles parensis (2%, n = 85 unsprayed, 7.8%, n = 591 sprayed). Biting peaks of Anopheles gambiae sensu lato (s.l.) followed periods of rainfall occurring between October and April, but were generally lower in sprayed sites than unsprayed. In most sprayed sites, An. gambiae s.l. indoor densities increased between January and February, i.e., 10–12 months after IRS. The predominant species An. arabiensis had a sporozoite rate in 2017 of 2.0% (95% CI 1.4–2.9) in unsprayed sites compared to 0.8% (95% CI 0.5–1.3) in sprayed sites (p = 0.003). Sporozoite rates were also lower for An. funestus collected in sprayed sites. Conclusion This study contributes to the understanding of malaria vector species composition, behaviour and transmission risk following IRS around Lake Victoria and can be used to guide malaria vector control strategies in Tanzania.

The C. cerifera palm tree (carnaúba) is widely distributed in the Northeastem Brazil, including the State of Piauí. This investigation revealed that R. nasutus is the ortly triatomine species captured on that palm tree, in five different localities. 78% of palm trees were infested with triatomines, and 4.0% were infected with flagellates morphologically and biologically indistinguishable from Trypanosoma cruzi. Birds, rodents and marsupials were found as major blood meai sources for R. nasutus.

<p>Understanding water-energy-food interrelatedness and interdependencies (WEF Nexus) over environmental resources can result in improved water, energy, and food security by integrating management and governance across sectors and scales, reducing tradeoffs, and building synergies, overall promoting sustainability and a transition to green economy. One of the most relevant research areas on the Nexus is the development of indexes to assess the performance of the three sectors and their interlinkages. These tools are essential to understanding the Nexus concept and to determine areas for improvement, especially in developing countries. To evaluate the WEF Nexus for a developing region, namely Latin America and the Caribbean (LAC), two approaches are proposed to evaluate water, energy and food security at intra-country level. First, a composite diagram was developed for LAC countries and subregions that considers three key indicators per sector: availability, access, and sustainability of the sector’s resources. Second, an analysis was performed for selected countries using internal factors that represent the most important interrelationships that exist within the WEF Nexus. The results show that access to food in LAC is relatively low in comparison to other developing regions. Regarding renewable energy sources, with the exception of some countries, the region’s share (including hydroelectricity) in electricity production is low. Water resources represent the most vulnerable sector for food and energy development of the countries (water for food, and water for energy), as well as the need to implement green infrastructure for sustainable water production (food for water).</p>

AbstractEighteen strains of Trypanosoma cruzi isolated from two species of triatomines in the state of Paraná, Brazil, were characterized molecularly using three strategies: RAPD (randomly amplified polymorphic DNA) with four primers, analysis of the D7 polymorphic region of the 24Sα rDNA, and RFLP (restriction fragment length polymorphism) of region 5′ of the mitochondrial gene COII (cytochrome oxidase subunit 2). The phenogram constructed with the RAPD data showed that only three strains isolated from Panstrongylus megistus collected in the Municipality of Arapongas were grouped together in a sub-branch. None of the other 15 strains could be clustered according to triatomine species or geographical origin. The strains were grouped with the T. cruzi I reference sample, indicating closer association with the sylvatic transmission cycle of T. cruzi in the state of Paraná. However, analyses of the rDNA and COII gene polymorphisms revealed the presence of populations from both T. cruzi I and II major lineages. In half of the analyzed triatomines, we found parasites from both lineages coinfecting the same bugs. Of these, most (6/9) were isolated from Triatoma sordida, and 3/9 from Panstrongylus megistus. These results contribute to a better comprehension of the ecoepidemiology of Chagas’ disease in Paraná, and raise questions about the role of studies of polyclonal population dynamics for controlling the transmission of T. cruzi to humans in this region.

Abstract Background The general picture of human genetic variation has been vastly depicted in the last years, yet many populations remain broadly understudied. In this work, we analyze for the first time the Merchero population, a Spanish minority ethnic group that has been scarcely studied and historically persecuted. Mercheros have been roughly characterised by an itinerant history, common traditional occupations, and the usage of their own language. Results Here, we examine the demographic history and genetic scenario of Mercheros, by using genome-wide array data, whole mitochondrial sequences, and Y chromosome STR markers from 25 individuals. These samples have been complemented with a wide-range of present-day populations from Western Eurasia and North Africa. Our results show that the genetic diversity of Mercheros is explained within the context of the Iberian Peninsula, evidencing a modest signal of Roma admixture. In addition, Mercheros present low genetic isolation and intrapopulation heterogeneity. Conclusions This study represents the first genetic characterisation of the Merchero population, depicting their fine-scale ancestry components and genetic scenario within the Iberian Peninsula. Since ethnicity is not only influenced by genetic ancestry but also cultural factors, other studies from multiple disciplines are needed to further explore the Merchero population. As with Mercheros, there is a considerable gap of underrepresented populations and ethnic groups in publicly available genetic data. Thus, we encourage the consideration of more ethnically diverse population panels in human genetic studies, as an attempt to improve the representation of human populations and better reconstruct their fine-scale history.

SUMMARY It is estimated that about 10 million people are infected with Trypanosoma cruzi worldwide, mostly in Latin America and more than 25 million are at risk of acquiring this infection in endemic areas. Dogs are an important reservoir for this pathogen and thus, considered a risk factor for human populations. This report describes one case of Chagas disease in a dog from Cuiabá, Mato Grosso State, Brazil. The diagnosis was obtained by direct examination of trypomastigote forms in blood smears. Amastigotes forms were visualized in microscopy of the bone marrow, lymph nodes, kidneys, liver and brain. The T. cruzi (ZIII) infection was confirmed by Polymerase Chain Reaction, and sequencing. The animal presented multisystemic failure and died. Although acute Chagas disease in humans is not reported in Cuiabá, this is the first report of a canine case in this region. This case represents a warning, to health professionals and authorities, to the possibility of transmission of this zoonosis in Cuiabá.
