The increase of a skin malignant melanoma (melanoma malignum) incidence in the world has been observed in recent years. The tumour, especially in advanced stadium with metastases, is highly resistant to conventional treatment. One of the strategies is to modulate melanogenesis using chemical compounds. In this study, the processes of differentiation and melanogenesis induced by dimethylsulfoxide (DMSO) in human melanoma cells (A-375) were investigated. Natural melanin isolated from A-375 melanoma cell line treated with 0.3% DMSO was analyzed by pyrolysis-gas chromatography-mass spectrometry (Py-GC/MS) method. The products derived from pheomelanin have not been stated in the pyrolytic profile of analyzed melanin. Within all products derived from eumelanins, 1,2-benzenediol has been predominated. It has been shown that in the melanoma cells stimulated with 0.3% and 1% DMSO, the increase of transcriptional activity of the tyrosinase gene took place. It was accompanied by the rise of tyrosinase activity and an accumulation of melanin in the cells. The better knowledge about the structure of melanins can contribute to establish the uniform criteria of malignant melanoma morbidity risk.

Human malignant melanoma is notoriously resistant to pharmacological modulation. We describe here for the first time that the synthetic retinoid CD437 has a strong dose-dependent antiproliferative effect on human melanoma cells (IC50: 5 x 10(-6) M) via the induction of programmed cell death, as judged by analysis of cell morphology, electron microscopical features, and DNA fragmentation. Programmed cell death was preceded by a strong activation of the AP-1 complex in CD437-treated cells as demonstrated by gel retardation and chloramphenicol transferase (CAT) assays. Northern blot analysis showed a time-dependent increase in the expression of c-fos and c-jun encoding components of AP-1, whereas bcl-2 and p53 mRNA levels remained constant. CD437 also exhibited a strong growth inhibitory effect on MeWo melanoma cells in a xenograft model. In tissue sections of CD437-treated MeWo tumors from these animals, apoptotic melanoma cells and c-fos overexpressing cells were colocalized by TdT-mediated deoxyuridine triphosphate-digoxigenin nick end labeling (TUNEL) staining and in situ hybridization. Taken together, this report identifies CD437 as a retinoid that activates and upregulates the transcription factor AP-1, leading eventually to programmed cell death of exposed human melanoma cells in vitro and in vivo. Further studies are needed to evaluate whether synthetic retinoids such as CD437 represent a new class of retinoids, which may open up new ways to a more effective therapy of malignant melanoma.

AbstractA soluble melanin pigment produced by Streptomyces sp. ZL-24 was purified and named StrSM. The elemental analysis of StrSM showed it consists of carbon, hydrogen, and oxygen. The spectrum analysis, including ultraviolet–visible absorption spectrum, Fourier-transform infrared spectrum, and pyrolysis–gas chromatography–mass spectrometry, indicated that StrSM might be pyomelanin. High performance liquid chromatography and liquid chromatography–mass spectra analysis of intermediate metabolite showed the presence of homogentisic acid (HGA). Moreover, the enzyme 4-hydroxyphenylpyruvate dioxygenase, involved in HGA biosynthesis, showed high activity during melanin production. Subsequently, a tyrosinase gene (melC2) and hydroxyphenylpyruvate dioxygenase gene double mutant demonstrated StrSM is pyomelanin. In vitro bioactivity assay showed that StrSM had excellent protective capability against SH-SY5Y cell oxidative injury. To our knowledge, the results firstly provide comprehensive data on Streptomyces pyomelanin identification and a promising candidate compound to treat oxidative injury of neurocytes.

Human malignant melanoma is the most aggressive type of skin cancer with high metastatic ability. Despite several traditional therapies, the mortality rate remains high. Lactobacillus plantarum (L. plantarum), a species of lactic acid bacteria (LAB), is being studied for human health, including cancer treatment. However, few studies have elucidated the relationship between L. plantarum extract and human malignant melanoma. To investigate the effects of L. plantarum on human melanoma cells, A375 human melanoma cells were used and treated with L. plantarum L-14 extract. After the treatment, viability, migration ability, molecular changes of migration- and apoptosis-related genes, and the location of cytochrome c was evaluated. The L-14 extract inhibited the viability, migration of A375 cells as well as reduced expression of migration-related genes. In addition, it was confirmed that the L-14 extract induced intrinsic apoptosis in A375 cells. This study demonstrated that the L-14 extract exerted anticancer effects on A375 cells. Therefore, these data suggest that the L-14 extract is worth studying for the development of melanoma drugs using LAB.

Abstract Agarwood grows wildly in the nature and its leaves may be utilized as raw materials for agarwood tea, especially from Aquilaria species. The study was conducted to determine the chemical compounds in the young and mature leaves of wild gaharu (Aquilaria malaccensis Lamk). Chemical compounds were detected using pyrolysis gas chromatography mass spectrometry (py-GCMS) while antioxidant capacity of the leaf extracts was assayed using 2,2-diphenyl-1-picryl-hydrazyl-hydrate (DPPH) free radical method. The results obtained a total of 30 chemical compounds in agarwood leaves extract with different composition between young and mature leaves. The agarwood leaves extracts displayed strong antioxidative capacity with a main compound namely octadecanoid acid or stearic acid.
