ABSTRACT Background Controversial findings have been reported in human and animal studies regarding the influence of n–6 (ω-6) to n–3 (ω-3) fatty acid ratios on obesity and health. Two confounding factors may be related to interactions with other dietary lipid components or sex-specific differences in fatty acid metabolism. Objective This study investigated main and interactive effects of total dietary lipid, ratio of n–6 to n–3 fatty acids, and sex on growth, adiposity, and reproductive health in wild-type zebrafish. Methods Male and female zebrafish (3 wk old) were fed 9 diets consisting of 3 ratios of n–6 to n–3 fatty acids (1.4:1, 5:1, and 9.5:1) varied within 3 total lipid amounts (80, 110, and 140 g/kg) for 16 wk. Data were then collected on growth, body composition (determined by chemical carcass analysis), and female reproductive success (n = 32 breeding events/diet over 4 wk). Main and interactive effects of dietary lipid and sex were evaluated with regression methods. Significant differences within each dietary lipid component were relative to the intercept/reference group (80 g/kg and 1.4:1 ratio). Results Dietary lipid and sex interacted in their effects on body weight (P = 0.015), total body length (P = 0.003), and total lipid mass (P = 0.029); thus, these analyses were stratified by sex. Female spawning success decreased as dietary total lipid and fatty acid ratio increased (P = 0.030 and P = 0.026, respectively). While total egg production was not associated with either dietary lipid component, females fed the 5:1 ratio produced higher proportions of viable embryos compared with the 1.4:1 ratio [median (95% CI): 0.915 (0.863, 0.956) vs 0.819 (0.716, 0.876); P < 0.001]. Conclusions Further characterization of dietary lipid requirements will help define healthy balances of dietary lipid, while the sex-specific responses to dietary lipid identified in this study may partially explain sex disparities in the development of obesity and its comorbidities.

This study studied the content and composition of the total lipid, lipid classes and fatty acids in 13 brown seaweed Sargassum species collected from Con Dao and Van Phong, Vietnam. The total lipid has a low content and varies among species from 0.10–1.70% of the fresh weight. From 13 species, seven lipid classes including polar lipid (Pol), free fatty acids (FFA), sterol (ST), hydrocarbon and wax (HW), triacylglycerol (TG), diacylglycerol (DG), and monoalkydiacylglycerol (MADG). Using the GC-FID technique, we have identified 29 fatty acids classified into 3 groups of saturated fatty acid, monounsaturated fatty acids, polyunsaturated fatty acids with an average content of 44.93%, 24.57% and 27.44%, respectively. Among those, many value fatty acids have been detected with high content such as C18:3n-3, C20:4n-6, 20:5n-3, and 22:6n-3. The lipid of 13 brown seaweed Sargassum species also fully contains omega-3,6,9 fatty acids with the content of 9.28%, 16.28% and 16.63%, respectively.

Fatty acid compositions were determined for total lipid (17.5% of the milk and > 95% triglycerides), 2-monoglyceride obtained by enzymatic hydrolysis of isolated triglyceride, and isolated phospholipid (~1% of total lipids). The total lipid fatty acids of the milk had a composition similar to fin whale depot fat but were enriched in hexadecanoic acid and polyunsaturated fatty acids at the expense of monoethylenic acids; correspondingly the iodine value of 136 (methyl esters) was higher than the normal range (105–120) of North Atlantic fin whale blubber oils. Over 80% of the fatty acids in the 2-position of the triglycerides were accounted for by relatively short chain fatty acids, especially hexadecanoic (54.6%), tetradecanoic (13.7%), and hexadecenoic (11.2%), so that the ester iodine value was only 48. The milk phospholipids had a fatty acid composition basically similar to that of liver phospholipids (methyl ester iodine value 120) with somewhat more polyunsaturated fatty acids and accordingly an iodine value of 144 for methyl esters.

ABSTRACT – HCTN 06The Sea urchin Tripneustes gratilla (Linnaeus, 1758) is a famous species of invertebrate member of the phylum Echinodermata of marine animals living in shallow tropical water. In Viet Nam, it is usually called by the name of yellow sea urchin. This species is mentioned much about its economic and commerce properties. Recently, many researchers have been paying attention to sea urchin because of its positive effect on human health. In terms of medicine and nutrition, the egg of yellow sea urchin was rich in Polyunsaturated Fatty Acids (PUFAs) and essential amino acids. In this research, we focused on determining the total lipid contents, the components of lipid classes and the compositions and contents of fatty acids, amino acids from the body and the egg of the yellow sea urchin T. gratilla (Linnaeus, 1758) collected in Hon Tam, Nha Trang, Khanh Hoa, Viet Nam in 2016. Although the yellow sea urchin T. gratilla has low percentage of total lipid in fresh weigh basis, they are full of essential lipid ingredients such as Wax and Hydrocabon (H + W), Triacylglycerol (TG), Monodiacylglycerol MDAG, Free Fatty Acid (FFA), sterol (ST), and Polar Lipid (PL). In both egg and body samples palmitic acid (16:0) was the most abundant fatty acid with the proportion of about 25%. In addition, arachidonic acid (20:4n-6) is dominent Omega-6 fatty acid in the Monounsaturated Fatty Acid group (MUFA). Furthermore, the research result also showed the very high percentages of omega-3 fatty acids in the egg and body of T. gratilla, with the total contents of 13.97% and 20.67% respectively, especially the presence of eicosapentaenoic fatty acid (C20:5n-3, EPA), a very valuable fatty acid. By the HPLC method, the research also found 17 amino acids in the egg and body of T. gratilla with the existence of 7 essential amino acids.Keywords: Tripneustes gratilla, lipid classes, yellow sea urchin, echinodermata, fatty acid content.

From a population of cells of Pseudomonas aeruginosa sensitive to 33 parts per million (ppm) of quaternary ammonium compound (QAC) and to 0.1 mg/ml of chloramphenicol (CAP), pure strains resistant to 750 ppm and 2.5 mg/ml of these antibacterial substances were isolated. Lipids from the sensitive and resistant cells grown under various conditions were extracted and characterized. Increased lipid biosynthesis was investigated as a possible factor in the resistance of this bacterium to CAP and QAC.Resistant cells grown in medium containing 207 ppm QAC at 25C averaged 77% more total lipid than sensitive cells. Resistant cells grown in the absence of QAC at 25C retained their resistance and averaged 27% more total lipid than sensitive cells. Cells grown at 32C in the presence of QAC contained 8% more total lipid than sensitive cells but less than cells grown at 25C under the same conditions. There was no increased lipid production by QAC-resistant cells grown at 37C in a Fermacell fermentor. Cells grown in CAP-containing medium (2.5 mg/ml) at 25C developed 28% more total lipid than sensitive cells grown in antibiotic-free medium.Gas–liquid chromatograms of the phospholipid, free fatty acid, and triglyceride fractions of the various cell types indicated that the same fatty acids were present. There was a 10% increase of the C18:1 fatty acid in both the phospholipid and free fatty acid fractions of the free lipids of cells grown in a Fermacell fermentor in the presence of CAP or QAC. However, there was no compensatory decrease in any single fatty acid, but rather a general decrease.Sensitive P. aeruginosa contained 14% free lipid and 3% to 6% bound lipid. The major phospholipid was phosphatidyl ethanolamine, which constituted 59% of the total phospholipid fraction of the sensitive cells. Analysis of fatty acids revealed that C16 amounted to 34%, C18:1 19%, and C16:1 and C18 each about 5% of the total present. These four fatty acids accounted for 63% of the free lipid present in the sensitive cells.

The leatherback turtle, Dermochelys coriacea (L.) studied was a male, weighing 916 kg, with a total dorsal length of 291 cm. It was beached on the Welsh coast, UK in September 1988 and is currently the largest leatherback ever recorded.Total lipid formed between 87.5 and 95.4% of the dry weight of representative samples of the blubber and 43.0% and 4.9% of the liver and pectoral muscle respectively. High levels of neutral lipid in the liver (79.0% of the total lipid) as well as the blubber (87.6–99.9% of the total lipid) suggest an important energy storage function for these tissues.Overall, with the notable exception of 22:lwll, fatty acids which are found in a putative jellyfish diet of Rhizostoma, Amelia, Cyanea and Chrysaora are also present in the leatherback liver and muscle, blubber and other fatty tissues. Fatty acid 22:lwll is present in the jellyfish samples, but is absent or at trace levels only in the leatherback tissues (0.1–0.3% of the total fatty acids).The polyunsaturated fatty acids of the w3 series 20:5w3, 22:5w3 and 22:6w3 are well represented in leatherback adipose tissues, muscle and liver as well as in the jellyfish examined. The leatherback and jellyfish lipids are therefore marine in character, but are also similar to terrestrial animal lipid in having a high proportion of fatty acids of the w6 series, principally arachidonic acid, 20:4w6. The significant levels of 20:4w6 in jellyfish total lipid (9.7–20.0% of the total fatty acids) and in the leatherback neutral lipid (1.0–10.9% of the total fatty acids) and phospholipid (0.6–15.5% of total fatty acids) fractions of all tissues sampled suggests that arachidonic acid assumes more importance in food chain relationships involving leatherbacks than in other marine food webs such as those involving fish.

Fractionation of total lipid extracted from Achlya sp. at various stages of its developmental cycle revealed that in spores total lipid was composed of 62% neutral lipid, 13% phospholipid, and 25% glycolipid. After germination, the proportion of neutral lipid rose slightly after 2 h then fell sharply to 10% after 8 h, whereupon it rose to 55% of total lipid after 30 h of growth, when sporulation was completed. Conversely, phospholipid rose to 77% of total lipid after 8 h, then declined to 40% after 30 h. Glycolipid was maintained at 10–20% of total lipid throughout the life cycle after spore germination. Quantitative determination of neutral lipid components by photoreflectometry showed that triglycerides accounted for 20% of neutral lipid in spores, and free fatty acids made up 50%. During growth, the absolute levels of both components fell precipitously on germination, remained at low levels throughout vegetative growth, and rose at the time of sporulation.The fatty acid composition of total lipid, phospholipid, neutral lipid, and free fatty acid fractions extracted from vegetative and sporulating Achlya cells was determined. The principal fatty acids present in all fractions at both stages of the life cycle were hexadecanoic and octadecanoic acids. Hydroxyhexadecanoic acid, eicosatetraenoic acid, and an unidentified long-chain acid were completely absent from the phospholipids of vegetative cells, although they were found in significant quantities in lipid fractions from other stages of growth.

This paper emphasizes those aspects of fatty acid research in fish that have relevance to the investigation of the functions of essential fatty acids in other species. Lipid requirements of fish came under investigation only in the 1960s. The most significant finding has been the requirement for n − 3 fatty acids. The dietary ratio of (n − 3):(n − 6) is critical if the essential requirement is met by C18 fatty acids because of competition between fatty acids for the enzymes involved in elongation and desaturation to produce the physiologically essential long-chain fatty acids. The fatty acid composition of fish lipids varies according to the fatty acid profile of the dietary lipid. The fatty acid composition of fish also responds to temperature changes in an adaptive mechanism for maintenance of membrane homeoviscosity and physiological function over a range of temperatures. The dietary intake of essential fatty acids by brood stock must be adequate for ova formation and for embryonic development, with the latter requirement being more critical for reproductive success. Absolute requirements of fish for essential fatty acids are difficult to define and may vary depending upon the dietary ratio of (n − 3) to (n − 6) fatty acids.Key words: essential fatty acids, nutritive requirements, fish.

In the uninfected hepatopancreas of L. truncatula 7·0–11·0% of the dry weight is lipid. Of the total lipid 60% is neutral lipid and 40% is phospholipid. Free fatty acid is the major neutral lipid component; triglycerides, diglycerides, monoglycerides, sterols and esterified sterols are also present. The phospholipids identified were phosphatidyl choline, phosphatidyl ethanolamine, lyso-phosphatidyl choline and sphingomyelin. The fatty acids were analysed by gas chromatography. The major fatty acid is C16 (palmitic) and 60% of the total fatty acids are saturated.In the hepatopancreas of L. truncatula infected with the rediae of F. hepatica, but with the rediae removed, 5·4–9·4% of the dry weight is lipid. Of this total lipid 73% is neutral lipid and 27% is phospholipid. All the fractions of neutral lipid, except the fatty acids are smaller than in the uninfected hepatopancreas. The fatty acids show an increase of 38%. The same phospholipids identified in the uninfected hepatopancreas are present, but all the fractions show a decrease in amount with the exception of the phosphatidyl choline fraction, which is present in approximately equal amounts in both the uninfected and the infected hepatopancreas. The major fatty acid is palmitic acid.

The features of polyunsaturated fatty acid-PUFA structures were corresponded to each separate functions  as adjusting the cellular physiology and gene expression. Therefore, lack of PUFA could lead to abnormalities in skin, kidney, neural networks, immune responses and inflammation; cardiovascular, endocrine, respiratory and reproductive systems. In fish oil, PUFA content were low, thus it was difficult to produce on a large scale. Therefore, the exploration of PUFA sources particularly as arachidonic acid-AA, eicosapentaenoic acid EPA, docosapentaenoic acid-DPA/DHA attracted many researches. Heterotrophic microalgae Thraustochytrids were capable of producing high amounts of DHA and PUFA composition varied. DHA can be synthesized by the metabolism of AA, EPA and DPA. The different types of PUFA reflected relationships in classification. Ten heterotrophic microalgae thraustochytrids isolated from mangrove Xuan Thuy, Nam Dinh contain fatty acid composition varied from C12 to C28. Especially, they had two important fatty acids of PUFA as EPA and DPA. Polyunsaturated fatty acids - PUFA content of ten thraustochytrid strains were from 28.95 to 49.62% total lipid. DPA compared to other PUFA were high for all thraustochytrid strains studied, accounting 20.22 to 39.35% TFA. Ten thraustochytrid strains had the highest growth with carbon source as glucose, total lipid reached 7 to 12.35 % dry weight biomass after 72 hours. Growth rate and lipid biosynthesis in organic nitrogen source were higher than in inorganic nitrogen sources. The best source of nitrogen for growth and lipid biosynthesis of ten thraustochytrid strains is yeast extract, total lipid were 8.57 to 18.87% dry weight biomass after 72 hours.

Fish production and fish processing waste have straight connection.  In India, the waste produced during the processing of fish is predicted to be approximate 3.6 million metric tonnes, 48 per cent of the total body weight of Indian and exotic major carps is thrown away as waste (non-edible ). The present research, it was conducted to compare the total lipid content (TLC) and fatty acid composition from the liver of captured and cultured fish, Labeo rohita  (Hamilton) having weight more than 500 gram during different months as well as to evaluate its nutritional quality. Maximum total lipid content (33.33±0.14%) was found in the liver of cultured fish in May month, while the minimum (15.26±0.24%) was in the liver of captured fish in the month of January. During the study, total lipid content was found to be considerably elevated in cultured than captured Fish, Labeo rohita  (Hamilton). The amount of three major groups of fatty acids namely polyunsaturated fatty acids, monounsaturated fatty acids and saturated fatty acids was found to be maximum in captured fish during the month of March 97.19±0.96%, 61.30±0.56% and 95.39±0.31% month of April respectively. Total n-6 polyunsaturated fatty acids were observed to be highest (21.95±8.05%) in the waste of liver in cultured fish during the month of March. Hence, it is concluded that the processing waste (liver) of the captured and cultured, Labeo rohita (Hamilton) is a prosperous resource of the essential fatty acids i.e.  PUFAs and total lipids. Further, it is observed that captured species are rich in fatty acid composition as compared to cultured species. Food industries can manufacture by-products from these high nutritional value contents of   waste for human utilization. EPA and DHA also reduce the risk of various life threatening diseases.
