By comparing the human and chimpanzee connectomes, van den Heuvel et al. demonstrate that connections unique to the human brain show greater involvement in schizophrenia pathology. Modifications in service of higher-order brain functions may have rendered the brain more vulnerable to dysfunction.

Abstract A comprehensive understanding of animal cognition requires the integration of studies on behavior, electrophysiology, neuroanatomy, development, and genomics. Although studies of comparative cognition are receiving increasing attention from organismal biologists, most current studies focus on the comparison of behaviors and anatomical structures to understand their adaptative values. However, to understand the most potentially complex cognitive program of the human brain a greater synthesis of a multitude of disciplines is needed. In this review, we start with extensive neuroanatomic comparisons between humans and other primates. One likely specialization of the human brain is the expansion of neocortex, especially in regions for high-order cognition (e.g., prefrontal cortex). We then discuss how such an expansion can be linked to heterochrony of the brain developmental program, resulting in a greater number of neurons and enhanced computational capacity. Furthermore, alteration of gene expression in the human brain has been associated with positive selection in DNA sequences of gene regulatory regions. These results not only imply that genes associated with brain development are a major factor in the evolution of cognition, but also that high-quality whole-genome sequencing and gene manipulation techniques are needed for an integrative and functional understanding of comparative cognition in non-model organisms.

Abstract Cognitive brain networks such as the default-mode network (DMN), frontoparietal network, and salience network, are key functional networks of the human brain. Here we show that the rapid evolutionary cortical expansion of cognitive networks in the human brain, and most pronounced the DMN, runs parallel with high expression of human-accelerated genes (HAR genes). Using comparative transcriptomics analysis, we present that HAR genes are differentially more expressed in higher-order cognitive networks in humans compared to chimpanzees and macaques and that genes with high expression in the DMN are involved in synapse and dendrite formation. Moreover, HAR and DMN genes show significant associations with individual variations in DMN functional activity, intelligence, sociability, and mental conditions such as schizophrenia and autism. Our results suggest that the expansion of higher-order functional networks subserving increasing cognitive properties has been an important locus of genetic changes in recent human brain evolution.

AbstractThe genetic basis and uniquely human character of schizophrenia has led to the notion of human brain evolution to have resulted in vulnerability to the disorder. We examined schizophrenia-related changes in brain connectivity in the context of evolutionary changes in human brain wiring by comparingin-vivoneuroimaging data from humans, chimpanzees and macaque monkeys. We find that evolutionary changes in human connectome organization overlap with the pattern of schizophrenia-related changes in brain connectivity, with connections evolutionary enhanced in the human brain showing significantly more involvement in schizophrenia pathology than connections shared between humans and non-human primates (effects shown in three independent patient-control datasets). Our findings suggest that the evolution of brain wiring in support of complex brain function in humans may have come at the cost of an increased vulnerability to brain dysfunction in disease.

Abstract J.T. Fraser used to emphasize the uniqueness of the human brain in its capacity for apprehending the various dimensions of “nootemporality” (Fraser 1982 and 1987). Indeed, our brain allows us to sense the flow of time, to measure delays, to remember past events or to predict future outcomes. In these achievements, the human brain reveals itself far superior to its animal counterpart. Women and men are the only beings, I believe, who are able to think about what they will do the next day. This is because such a thought implies three intellectual abilities that are proper to mankind: the capacity to take their own thoughts as objects of their thinking, the ability of mental time travels—to the past thanks to their episodic memory or to the future—and the possibility to project very far into the future, as a consequence of their enlarged and complexified forebrain. But there are severe limits to our timing abilities of which we are often unaware. Our sensibility to the passing time, like other of our intellectual abilities, is often competing with other brain functions, because they use at least in part the same neural networks. This is particularly the case regarding attention. The deeper the level of attention required, the looser is our perception of the flow of time. When we pay attention to something, when we fix our attention, then our inner sense of the flux of time freezes. This limitation should not sound too unfamiliar to the reader of J.T. Fraser who wrote in his book Time, Conflict, and Human Values (1999) about “time as a nested hierarchy of unresolvable conflicts.”
