Bio-ink properties have been extensively studied for use in the three-dimensional (3D) bio-printing process for tissue engineering applications. In this study, we developed a method to synthesize bio-ink using hyaluronic acid (HA) and sodium alginate (SA) without employing the chemical crosslinking agents of HA to 30% (w/v). Furthermore, we evaluated the properties of the obtained bio-inks to gauge their suitability in bio-printing, primarily focusing on their viscosity, printability, and shrinkage properties. Furthermore, the bio-ink encapsulating the cells (NIH3T3 fibroblast cell line) was characterized using a live/dead assay and WST-1 to assess the biocompatibility. It was inferred from the results that the blended hydrogel was successfully printed for all groups with viscosities of 883 Pa∙s (HA, 0% w/v), 1211 Pa∙s (HA, 10% w/v), and 1525 Pa∙s, (HA, 30% w/v) at a 0.1 s−1 shear rate. Their structures exhibited no significant shrinkage after CaCl2 crosslinking and maintained their integrity during the culture periods. The relative proliferation rate of the encapsulated cells in the HA/SA blended bio-ink was 70% higher than the SA-only bio-ink after the fourth day. These results suggest that the 3D printable HA/SA hydrogel could be used as the bio-ink for tissue engineering applications.

In tissue engineering (TE), a porous scaffold structure of biodegradable material is required as a template to guide the proliferation, growth and development of cells appropriately in three dimensions. The scaffold must meet design requirements of appropriate porosity, pore size and interconnected structure to allow cell proliferation and adhesion. This paper presents a methodology for design and manufacture of TE scaffolds with varying porosity by employing open structure building units and Fused Deposition Modeling (FDM) rapid prototyping technique. A computer modeling approach for constructing and assembly of three-dimensional unit cell structure is presented to provide a solution of scaffolds design that can potentially meet the diverse requirements of TE applications. A parametric set of open polyhedral unit cells is used to assist the user in designing the required micro-architecture of the scaffold with required porosity and pore size and then the Boolean operation is used to create the scaffold of a given CAD model from the designed microstructure. The procedure is verified by fabrication of physical scaffolds using the commercial FDM system.

Pore size, external shape, and internal complexity of additively manufactured porous titanium scaffolds are three primary determinants of cell viability and structural strength of scaffolds in bone tissue engineering. To obtain an optimal design with the combination of all three determinants, four scaffolds each with a unique topology (external geometry and internal structure) were designed and varied the pore sizes of each scaffold 3 times. For each topology, scaffolds with pore sizes of 300, 400, and 500 µm were designed. All designed scaffolds were additively manufactured in material Ti6Al4V by the direct metal laser melting machine. Compression test was conducted on the scaffolds to assure meeting minimum compressive strength of human bone. The effects of pore size and topology on the cell viability of the scaffolds were analyzed. The 12 scaffolds were ultrasonically cleaned and seeded with NIH3T3 cells. Each scaffold was seeded with 1 million cells. After 32 days of culturing, the cells were fixed for their three-dimensional architecture preservation and to obtain scanning electron microscope images.

Three-dimensional (3D) printing technologies have become an attractive manufacturing process to fabricate scaffolds in tissue engineering. Recent research has focused on the fabrication of alginate complex shaped structures that closely mimic biological organs or tissues. Alginates can be effectively manufactured into porous three-dimensional networks for tissue engineering applications. However, the structure, mechanical properties, and shape fidelity of 3D-printed alginate hydrogels used for preparing tissue-engineered scaffolds is difficult to control. In this work, the use of alginate/gelatin hydrogels reinforced with TiO2 and β-tricalcium phosphate was studied to tailor the mechanical properties of 3D-printed hydrogels. The hydrogels reinforced with TiO2 and β-TCP showed enhanced mechanical properties up to 20 MPa of elastic modulus. Furthermore, the pores of the crosslinked printed structures were measured with an average pore size of 200 μm. Additionally, it was found that as more layers of the design were printed, there was an increase of the line width of the bottom layers due to its viscous deformation. Shrinkage of the design when the hydrogel is crosslinked and freeze dried was also measured and found to be up to 27% from the printed design. Overall, the proposed approach enabled fabrication of 3D-printed alginate scaffolds with adequate physical properties for tissue engineering applications.

Being one of the three elements of tissue engineering, three-dimensional porous structure scaffold plays an important role in tissue engineering. As it not only prvovide cells for the life, but also serves as a template to guide tissue regeneration and control of organizational structure and other functions. In this study, hyaluronic acid and gelatin are successfully cross-linked by 1-ethyl- (3-dimethylaminopropyl) -carbodiimide hydrochloride (EDC) , and compound β-TCP microspheres to prepare porous hydrogel scaffolds. The microspheres were analyzed by X-ray diffraction (XRD). The scaffolds were characterized by scanning electron microscopy (SEM) and Fourier transform infrared spectroscopy (FTIR). At the same time, the compressive strength, swelling ratio, degradation of the scaffold were tested. To assess the in vitro cell compatibility of the scaffolds, mouse L929 fibroblasts were seeded onto scaffolds for cell morphology and cell viability studies. The results showed that the pore size of the porous scaffold can be adjusted by changing the ratio of gelatin to hyaluronic acid (HA), increasing the proportion of hyaluronic acid in a certain range, pore size will be significantly increased. With the increase of the proportion of hyaluronic acid in the scaffold, the swelling ratio and the degradation rate also increased. The compressive strength of the scaffold increased with the increase of the proportion of gelatin. The appropriate ratio of β-TCP can promote cell growth and proliferation.

Three-dimensional (3D) printing, also known as additive manufacturing, was developed originally for engineering applications. Since its early advancements, there has been a relentless development in enthusiasm for this innovation in biomedical research. It allows for the fabrication of structures with both complex geometries and heterogeneous material properties. Tissue engineering using 3D bio-printers can overcome the limitations of traditional tissue engineering methods. It can match the complexity and cellular microenvironment of human organs and tissues, which drives much of the interest in this technique. However, most of the preliminary evaluations of 3Dprinted tissues and organ engineering, including cardiac tissue, relies extensively on the lessons learned from traditional tissue engineering. In many early examples, the final printed structures were found to be no better than tissues developed using traditional tissue engineering methods. This highlights the fact that 3D bio-printing of human tissue is still very much in its infancy and more work needs to be done to realise its full potential. This can be achieved through interdisciplinary collaboration between engineers, biomaterial scientists and molecular cell biologists. This review highlights current advancements and future prospects for 3D bio-printing in engineering ex vivo cardiac tissue and associated vasculature, such as coronary arteries. In this context, the role of biomaterials for hydrogel matrices and choice of cells are discussed. 3D bio-printing has the potential to advance current research significantly and support the development of novel therapeutics which can improve the therapeutic outcomes of patients suffering fatal cardiovascular pathologies.
