Associations between geographical origin and morphological characters in bread wheat (Triticum aestivum L.)

We report the morphological typing of 13,337 accessions of bread wheat. The individual characters assessed were: awnedness, awn colour, glume colour, presence/absence of glume hairs, spike type, spike density, spike branching, grain colour, stem type (hollow/solid) and presence/absence of ligules. We have associated these morphological data with accession provenance, to analyse global patterns of the traits. Whereas some of the traits (awnedness, awn colour, glume colour, presence/absence of hairs on glumes, stem type) are concentrated in distinct geographical regions, others (spike type, spike density, grain colour, presence/absence of ligules) appear to be randomly distributed. Spike branching was not observed. Possible reasons for the geographical distributions of the characters, in the light of their inheritance, are discussed.

Bread wheat is an important food crop of world and Pakistan. An experiment was conducted in winter wheat growing season to assess yield and yield related traits of newly evolved wheat genotypes. The 16 wheat genotypes includes 14 advanced lines viz., CIM-04-5, CIM-04-21, CIM-04-3, C7-98-11, 5-02, V2-10-12, CIM-03-2, C2-98- 6, 6-12, V3-10-9, C6-98-5, V3-10-32, C2-98-8, V2-10-21 and 2 local checks NIA Sunhari and Kiran 95 were tested. Experimental design was laid out in RCBD with 3 replicates. Mean square for genotypes showed high significantly differences for most of agro-morphological characters. Mean and range of all wheat genotypes for all the traits indicated a considerable variability between genotypes. Mean performance for the trait grain yield showed that newly developed genotypes C2-98-8, CIM-04-21, V3-10-32 and CIM-04-3 produced higher grain yield (3 to 3.25 kg plot-1) than both the contesting check varieties. High significantly and positively correlation of the plot yield to thousand grain weight (0.41**), biomass (0.41**) and harvest index (0.86***) with grain yield were found. It indicated that by improving these three traits, we can significantly improve grain yield. Selected genotypes and traits can be used in breeding program for wheat improvement.

A new species from southwest of Bojnord, NE of Iran is described and illustrated here as Dianthus pseudocrinitus (Caryophyllaceae). This species is morphologically similar to D. crinitus subsp. turcomanicus, but can be distinguished from the latter by bract number, width of lower and upper leaves, margin of thickness of outermost bracts, length of calyx, length and width of anther, branches of stem, type of sheath of lower leaf, and tip shape of petal fimbria. The new species is also similar to D. orientalis subsp. stenocalyx in terms of floral characters, but they differ by several non- overlapping morphological characters. Results obtained from the morphological data are consistent with those obtained from the molecular phylogenetic trees based on sequences of the two copies of DFR1 gene, confirming phylogenetic affinity of the new species to D. crinitus subsp. turcomanicus.

Aim: Quinoa is a popular source of protein, minerals and alternative to traditional grains. The objective of this study is to introduce the Quinoa in the semi-arid zone of Sindh province of Pakistan. Method: A variety of NARC-9 from the agricultural Punjab province was cultivated and subjected to analyze the growth, morphological characters of the varieties obtained, saponin, protein and the elemental composition viz. Cd, Cu, Fe, K, Na, Pb, and Zn. Result: The result demonstrated the optimum growth and no disease were found in the experimental area. At least three major varieties of quinoa were obtained. Seed morphological data of these three quinoa cultivars were collected. The average saponin levels were quite reasonable. Overall proteins band pattern revealed very high polymorphism in quinoa cultivars and the results were also in good agreement with earlier studies. Conclusion: All quinoa cultivars of Madinat al-Hikmah showed high concentrations of albumin than globulin concentrations (i.e. 48-52% and 24-27%, respectively) as compared to control seeds from market that had similar concentrations of the two fractions i.e. 35.58% and 37.68%, respectively. Likewise, low concentrations of prolamin 14-16% and glutelin 11-12% compared to control seeds 13% rank our crop much better quality than the imported one in the market. The trend of elemental accumulation was followed as K >Na >Fe >Zn >Cu >Pb >Cd, while for comparison it was Na >K >Zn >Fe >Cu >Pb >Cd >Pb for wheat grown under similar conditions. Traditional grains together make a major contribution to the total nutritional element intake of the average Pakistani citizen through diet, not only because of large amounts consumed, but also in part by suitable levels of their proteins and elemental up take for good health. Thus the successful cultivation of quinoa in the semi-arid zone of Sindh will certainly prove beneficial.

Abstract Polyploidy occurs prevalently and plays an important role during plant speciation and evolution. This phenomenon suggests polyploidy could develop novel features that enable them to adapt wider range of environmental conditions compared with diploid progenitors. Bread wheat (Triticum aestivum L., BBAADD) is a typical allohexaploid species and generally exhibits greater salt tolerance than its tetraploid wheat progenitor (BBAA). However, little is known about the underlying molecular basis and the regulatory pathway of this trait. Here, we show that the histone acetyltransferase TaHAG1 acts as a crucial regulator to strengthen salt tolerance of hexaploid wheat. Salinity-induced TaHAG1 expression was associated with tolerance variation in polyploidy wheat. Overexpression, silencing and CRISPR-mediated knockout of TaHAG1 validated the role of TaHAG1 in salinity tolerance of wheat. TaHAG1 contributed to salt tolerance by modulating ROS production and signal specificity. Moreover, TaHAG1 directly targeted a subset of genes that are responsible for hydrogen peroxide production, and enrichment of TaHAG1 triggered increased H3 acetylation and transcriptional upregulation of these loci under salt stress. In addition, we found the salinity-induced TaHAG1-mediated ROS production pathway is involved in salt tolerance difference of wheat accessions with varying ploidy. Our findings provide insight into the molecular mechanism of how an epigenetic regulatory factor facilitates adaptability of polyploidy wheat and highlights this epigenetic modulator as a strategy for salt tolerance breeding in bread wheat.

AbstractHypergravity—an evolutionarily novel environment has been exploited to comprehend the response of living organisms including plants in the context of extra-terrestrial applications. Recently, researchers have shown that hypergravity induces desired phenotypic variability in seedlings. In the present study, we tested the utility of hypergravity as a novel tool in inducing reliable phenotype/s for potential terrestrial crop improvement applications. To investigate, bread wheat seeds (UAS-375 genotype) were subjected to hypergravity treatment (10×g for 12, and 24 h), and evaluated for seedling vigor and plant growth parameters in both laboratory and greenhouse conditions. It was also attempted to elucidate the associated biochemical and hormonal changes at different stages of vegetative growth. Resultant data revealed that hypergravity treatment (10×g for 12 h) significantly enhanced root length, root volume, and root biomass in response to hypergravity. The robust seedling growth phenotype may be attributed to increased alpha-amylase and TDH enzyme activities observed in seeds treated with hypergravity. Elevated total chlorophyll content and Rubisco (55 kDa) protein expression across different stages of vegetative growth in response to hypergravity may impart physiological benefits to wheat growth. Further, hypergravity elicited robust endogenous phytohormones dynamics in root signifying altered phenotype/s. Collectively, this study for the first time describes the utility of hypergravity as a novel tool in inducing reliable root phenotype that could be potentially exploited for improving wheat varieties for better water usage management.

AbstractKristianstads Vattenrike Biosphere Reserve [KVBR] is a UNESCO designated area of Sweden possessing high biological value. Although several studies on tardigrades inhabiting Sweden have been performed, the KVBR area has been neglected. The current study investigates the tardigrade fauna of five areas of the biosphere reserve and includes 34 samples of different substrates analysed quantitatively and qualitatively. In total, 33 species of tardigrades were found in the samples, including 22 new records for the Skåne region, 15 new records for Sweden, and four species new to science. Mesobiotus emiliae sp. nov., Xerobiotus gretae sp. nov., Itaquascon magnussoni sp. nov., and Thulinius gustavi sp. nov. were described with an integrative approach (when possible) using morphological characters (light, electron scanning, and confocal laser scanning microscopies) and molecular markers (ITS2, 18S, 28S, cox1). A new protocol to increase morphological data was developed recovering mounted specimens within old slides for SEM analysis. Emended diagnoses for the genus Itaquascon and the transfer of Platicrista itaquasconoide to the genus Meplitumen are proposed. This study enriches the knowledge of the tardigrade biodiversity both within the KVBR and in Sweden and contributes to the rapidly increasing number of tardigrade species reported worldwide. The 33 species identified in the KVBR area represents 28% of all water bear species found in Sweden so far. The restricted study areas and limited number of samples collected suggests that the KVBR is very rich of tardigrades.

Abstract Hexaploid bread wheat (Triticum aestivum L. em. Thell) is one of the world's most important crop plants and displays a very low level of intraspecific polymorphism. We report the development of highly polymorphic microsatellite markers using procedures optimized for the large wheat genome. The isolation of microsatellite-containing clones from hypomethylated regions of the wheat genome increased the proportion of useful markers almost twofold. The majority (80%) of primer sets developed are genome-specific and detect only a single locus in one of the three genomes of bread wheat (A, B, or D). Only 20% of the markers detect more than one locus. A total of 279 loci amplified by 230 primer sets were placed onto a genetic framework map composed of RFLPs previously mapped in the reference population of the International Triticeae Mapping Initiative (ITMI) Opata 85 × W7984. Sixty-five microsatellites were mapped at a LOD >2.5, and 214 microsatellites were assigned to the most likely intervals. Ninety-three loci were mapped to the A genome, 115 to the B genome, and 71 to the D genome. The markers are randomly distributed along the linkage map, with clustering in several centromeric regions.
