1. Blood pyruvate carboxylase (pyruvate-CO2 ligase (ADP-forming); EC 6.4.1.1; PC) activities in young chickens and turkeys given low-biotin diets supplemented with biotin at graded levels were studied in three experiments.2. In both species PC activity was related positively to the supplemental biotin level. The relationship was sigmoid and maximum activity was attained with supplemental levels above those required to give maximal growth response.3. Enzyme activity decreased between 2 and 4 weeks of age but remained almost constant thereafter.4. Activity in chicks was not affected by alterations in the fat or protein content of the diet.5. Changing poults from high to low and from low to high supplemental biotin levels resulted in reversals in the levels of enzyme activity.6. It is concluded that blood PC activity is a promising new criterion for assessing the biotin status of young chickens and turkeys.

The article performs new data on the relationship between the hydrolytic and transport enzyme activity at different poles of the enterocytes plasmolemma of cow's fetal large intestine with the content of individual fractions of polypeptides. An expressive direct dependence of enzyme activity dynamics on the apical and basolateral membranes of enterocytes containing low molecular weight proteins and an inverse relationship with the concentration of proteins with medium and large molecular weights has been proved. It was found that the alkaline activity of the phosphatase and γ-glutamyltransferase on the apical domain of enterocyte plasmolemma is directly related to the proteins content with molecular masses of 9.6–14.2 kD, 21 kD, 22.5 kD, 26 kD, 33 kD, 35 kD, 170–185 kD, and 205 kD (P ≤ 0.05–0.001). Gamma-glutamyltransferase activity is straightly related to protein quantity with molecular weights of 15.5 kDa and 39 kD (P ≤ 0.05). In contrast, alkaline phosphatase and GGT activity have inverse correlations with the content of polypeptides with molecular masses of 46 kD, 63 kD, and 250 kD in the apical membrane of enterocytes (P ≤ 0.01–0.001). The lactase activity in the cattle enterocytes apical membrane during the test period has significant direct correlations only with the amount of the polypeptide of polypeptides with molecular weights of 31 kD, 39 kD, and 100 kD (P ≤ 0.05–0.01) and inverse relationships containing proteins with molecular masses of 46 kD and 120 kD (P ≤ 0.05). A linear dependence of the different ATPase activity of the apical membrane of red blood cells containing proteins with molecular weights of 9.6–14.2 kD, 15.5 kD, 21 kD, 22.5 kD, 33 kD, 35 kD, 39 kD, and 205 kD (P ≤ 0.05–0.001) was observed. Alkaline phosphatase activity in the apical membrane of enterocytes is only directly related to the number of proteins with molecular weights of 17 kD and 24 kD (P ≤ 0.001) in this domain. It inversely depends on the content of proteins with molecular masses of 9.6–14.2 kD and 52 kD (P ≤ 0.001). G-glutamyltransferase activity is inversely related to protein content with molecular weights of 43 kD, 52 kD, 66 kD, 87 kD, and 100 kD and 155 kD (P ≤ 0.001). The Ca2+, Mg2+-ATPase of the basolateral membrane activity of enterocytes is directly related to the protein amount with molecular weights of 26 kD (P ≤ 0.01), Mg2+-ATPase and Mg2+-ATPase with protein content with the molecular value of 100 kD (P ≤ 0.05).

The relationship between dietary protein, essential amino acids (EM) and energy was examined in two experiments using male broiler chicks fed practical diets to eight weeks of age. All diets contained a balanced proportion of ten EAA which was maintained over various treatments. Growth of birds was closely related to the EAA content of the diet and poorly related to dietary protein content. When the dietary concentration of ten EAA was treated as a combined unit and expressed as proportion of diet, 7.6 per cent EAA in an 18 per cent protein starter diet and 5.8 per cent EAA in a 16 per cent protein finisher diet were adequate to support maximal growth of male birds to eight weeks of age. Feed intakes were reduced as energy concentrations increased in diets regardless of protein content of diets.

Six endomycorrhiza isolates from the Sonoran Desert of Mexico [Desert-14(18)1, 15(9)1, 15(15)1, Palo Fierro, Sonoran, and G. geosporum] were evaluated with a pure isolate of Glomus intraradices for their effect on the growth and gas exchange of Hibiscus rosa-sinensis L. cv. Leprechaun under low phosphorus fertility (11 mg P/L). Rooted cuttings of Hibiscus plants were inoculated with the seven mycorrhiza isolates and grown for 122 days. Gas exchange measurements were made on days 26, 88, and 122 after inoculation, and plants were harvested on day 123 for growth analysis. Plants inoculated with the seven isolates had 70% to 80% root colonization at harvest. Plants inoculated with G. intraradices had significantly higher leaf, shoot and root dry matter (DM), leaf DM/area (P ≤ 0.05) than those inoculated with any of the six isolates, and greater leaf area (LA) than Desert-15(9)1 and 15(15)1. Uninoculated plants had significantly lower leaf, shoot, root DM, leaf DM/area and LA (P ≤ 0.05) than the inoculated plants. There were no differences among the seven isolates in any of the gas exchange parameters measured [photosynthesis (A) stomatal conductance (gs), the ratio of intercellular to external CO2 (ci/ca), A to transpiration (E) ratio (A/E)]. The relationship between inoculated and uninoculated plants in these gas exchange parameters were variable on day 122 after inoculation.

AbstractThe need for qualitative research of deservingness perception is strongly emphasised in the literature. This article studies the perception of deservingness for a "Family 500 +"—cash benefit in Poland. For the first time, data from online forums was used in the studies of deservingness and welfare attitudes. It allowed to avoid numerous limitations associated with social surveys. The qualitative analysis showed how participants of Internet debates perceive the criteria of deservingness: control, attitude, reciprocity, identity, need, and what are the relations between the criteria. The impurity of all deservingness criteria was indicated and a new criterion “adequacy” was proposed. Moreover due to the fact that the study concerned a concrete, non-abstract family cash benefit addressed the relationship between the perceived deservingness of children and their parents was pointed out. The vast majority of posts on Internet forums referred to deservingness of parents, not children. This is particularly evident in relation to the criteria of control and reciprocity. Presenting the hypothesis of jealousy and scapegoat strategy, the article also shows the direction of future research on deservingness.

Abstract Background Due to the multitude industrial applications of ligninolytic enzymes, their demands are increasing. Partial purification and intensive characterization of contemporary highly acidic laccase enzyme produced by an Egyptian local isolate designated Alcaligenes faecalis NYSO were studied in the present investigation. Results Alcaligenes faecalis NYSO laccase has been partially purified and intensively biochemically characterized. It was noticed that 40–60% ammonium sulfate saturation showed maximum activity. A protein band with an apparent molecular mass of ~ 50 kDa related to NYSO laccase was identified through SDS-PAGE and zymography. The partially purified enzyme exhibited maximum activity at 55 °C and pH suboptimal (2.5–5.0). Remarkable activation for enzyme activity was recognized after 10-min exposure to temperatures (T) 50, 60, and 70 °C; time elongation caused inactivation, where ~ 50% of activity was lost after a 7-h exposure to 60 °C. Some metal ions Cu2+, Zn2+, Co2+, Ni2+, Mn2+, Cd2+, Cr2+, and Mg2+ caused strong stimulation for enzyme activity, but Fe2+ and Hg2+ reduced the activity. One millimolar of chelating agents [ethylenediamine tetraacetic acid (EDTA), sodium citrate, and sodium oxalate] caused strong activation for enzyme activity. Sodium dodecyl sulfate (SDS), cysteine-HCl, dithiothreitol (DTT), β-mercaptoethanol, thioglycolic acid, and sodium azide caused strong inhibition for NYSO laccase activity even at low concentration. One millimolar of urea, imidazole, kojic acid, phenylmethylsulfonyl fluoride (PMSF), H2O2, and Triton X-100 caused activation. The partially purified NYSO laccase had decolorization activity towards different dyes such as congo red, crystal violet, methylene blue, fast green, basic fuchsin, bromophenol blue, malachite green, bromocresol purple eriochrome black T, and Coomassie Brilliant Blue R-250 with various degree of degradation. Also, it had a vast range of substrate specificity including lignin, but with high affinity towards p-anisidine. Conclusion The promising properties of the newly studied laccase enzyme from Alcaligenes faecalis NYSO strain would support several industries such as textile, food, and paper and open the possibility for commercial use in water treatment. It will also open the door to new applications due to its ligninolytic properties in the near future.

An investigation was undertaken into the relationship between the enzyme activity of cells harvested from raw milk and time taken for age gelation (TAG) to occur in the milk after ultra-high-temperature processing. It was shown that there was no relationship between the TAG and the bacterial counts on milk agar at 30°C or 7°C nor was there a relationship between the counts and the level of enzyme activity of the harvested cells. There was, however, a significant correlation between the level of enzyme activity of the harvested cells and the TAG. When extra bovine leucocytes were added to raw milk before processing, the TAG was increased. This suggested that there was an inhibitory action of leucocytes in development of age gelation.

Abstract In 424 African-American and 75 white subjects, we found that the −5 (TPI 592 A→G), −8 (TPI 589 G→A), and −24 (TPI 573 T→G) variants in the triosephosphate isomerase (TPI) gene occurred frequently (41.0%) in the African-American subjects but did not occur in the whites. These data suggest that this set of polymorphisms may turn out to be one of the higher-incidence molecular markers of African lineage, a surprising finding because others had reported that these nucleotide substitutions were restricted to a small subset of African Americans who had been characterized as TPI-deficiency heterozygotes. Additionally, we investigated the relationship of these variants to TPI-enzyme activity. Although the variant substitutions (occurring in three haplotypes: −5 alone, −5 −8, and −5 −8 −24) were associated with moderate reduction in enzyme activity, severe-deficiency heterozygotes could not be identified with certainty, and none of the haplotypes were restricted to subjects with marked reduction of enzyme activity. Three subjects were homozygous for the −5 −8 haplotype, a finding inconsistent with the putative role of this haplotype as the cause of a null variant incompatible with life in homozygotes. Despite these findings, the possibility remains that the −5 −8 or −5 −8 −24 haplotypes may in some instances contribute to compound heterozygosity and clinical TPI deficiency. © 1998 by The American Society of Hematology.

In the whiting, no significant changes occur in the composition of the muscle substance, either throughout the year, or with increasing age, or during ripening of the gonads.The amount of fat in the liver is very low in immature fishes, increasing greatly with age, and reaching its maximum just after the commencement of the ripening of the gonads. It then falls off, reaching its lowest value amongst mature fishes in the “Spent” stages. Males are richer in liver fat than females.Immature fish have a decidedly higher protein content in the liver than have mature fish.
