1. The effect of methionine supplementation upon pancreatic trypsin, chymotrypsin, pancreatopeptidase E and amylase of chicks given raw and heated soya-bean diets (RSD and HSD) was studied before and after fasting.2. Statistical analysis of the ratios of amylase to pancreatopeptidase E, trypsin and chymotrypsin in each individual chick revealed that methionine supplementation consistently increased the ratio of amylolytic to proteolytic enzymes. This phenomenon was most marked in the fasted RSD group, less in the fed RSD and fasted HSD groups and least in the fed HSD group.3. The results are discussed in relation to methionine metabolism in chicks given RSD and HSD supplemented with trypsin inhibitors.

1. The action of raw and heated soya beans on pancreatic function was studied by measuring the amylase activity of the pancreases and intestinal contents of intact chickens and of chickens with ileostomies at fasting and after eating, before and after adaptation to heated and raw soya-bean diets, with a methionine supplement and without. 2. The concentration of amylase in the pancreases of 8-week-old chickens eating raw soya-bean diets was lower than that of those eating heated soya-bean diets. These chickens were deficient in methionine; supplementation of the raw soya-bean diet with methionine greatly increased the levels of amylase in the pancreases of such chickens; supplementation of the heated soya-bean diet with methionine increased the levels of amylase in pancreases to a smaller extent. 3. Methionine was less effective in increasing the amylase levels of the pancreases of 11-week-old chickens eating raw soya-bean diets, since at this age the deficiency of methionine was much less pronounced. 4. More amylase activity was found in intestinal contents of chickens eating raw soya-bean diets than in those eating heated soya-bean diets. 5. In vitro studies showed that fractions prepared from raw soya beans, all containing high levels of trypsin inhibitors, stabilized amylase activity despite the active proteolysis of other protein substrates in the same amylase-containing mixtures. 6. CaCl2 stabilized amylase activity in vitro in unactivated and activated pancreatic juice as such, and after additions of raw soya beans or their fractions before and after autoclaving. 7. The increased levels of amylase found in the intestinal contents of the chickens eating raw soya beans may represent greater secretion of amylase activity by the pancreas, stabilization of amylase activity by the raw soya bean, or both factors. Interpretation of measurements of amylase activity in the intestinal contents of chickens eating heated or raw soya beans is uncertain. 8. The bearing of this work on current views of the nutritional value of raw and heated soya beans is discussed.

1. Feeding on a raw soya-bean diet (RSD) increased the levels of trypsin, chymotrypsin and pancreatopeptidase E but decreased the level of amylase in the pancreas of chicks as compared to a heated soya-bean diet (HSD), while supplementation of HSD with soya-bean trypsin inhibitors increased the activity of all four enzymes. HSD + trypsin inhibitors caused significant enlargement of the pancreas but only a slight depression in growth rate.2. Fasting for 24 h of chicks previously given RSD and HSD increased the activity of all four enzymes but the increase was much greater in chicks previously given RSD than in those previously given HSD.3. Feeding RSD for 4 d to chicks previously adapted to HSD resulted in a dramatic inhibition in growth rate, a small increase in pancreas weight, and an increase in the activity of all proteolytic enzymes, while no change in the amylase was detectable.4. Trypsin, chymotrypsin and pancreatopeptidase E activities were assayed in the contents of the small intestine and caecum of chicks fed on RSD or HSD over a period of 35 d. Trypsin and chymotrypsin activities in the small intestine were lower in chicks fed on RSD while pancreatopeptidase E activity was almost equal or even higher in RSD-fed chicks, especially at the age of 35 d. Trypsin activity in the caecum of RSD-fed chicks was lower at all stages of the experiment, while the pancreatopeptidase E and chymotrypsin activities in the caecum of RSD-fed chicks exceeded the levels in the HSD group at the age of 21 and 35 d respectively. It would appear therefore that pancreatopeptidase E may play an important part in overcoming the inhibition of the proteolytic activity in the intestine of chicks fed on RSD.

<b><i>Objectives:</i></b> This study aimed to determine the oral morphine equivalents (OMEs) prescribed and refill rates following hysterectomy and hysteroscopy in the setting of opioid prescribing practice changes in 2 states. <b><i>Design:</i></b> This is a retrospective cohort analysis consisting of 2,916 patients undergoing hysterectomy or hysteroscopy between July 2016 and September 2019 at 2 affiliated academic hospitals in states that underwent legislative changes in opioid prescribing in 2018. <b><i>Methods:</i></b> Participants were identified using the Current Procedural Terminology procedure codes in Arizona and Florida. Hysterectomy was chosen as the most invasive gynecologic procedure, while hysteroscopy was chosen as the least invasive. Medical records were abstracted to find opioid prescriptions from 90 days before surgery to 30 days after discharge. Patients with opioid use between 90 and 7 days before surgery were excluded. Prescriptions were converted to OMEs and were calculated per quarter year. Statistical analysis included Wilcoxon rank sum <i>t</i> tests for OMEs and χ2 <i>t</i> tests for refill rates. Interrupted time-series analysis was used to determine significant change in OMEs before and after legislative change. Statistical analysis was performed using SAS version 9.4 (SAS Institute, Cary, NC, USA). <b><i>Results:</i></b> In Arizona, 1,067 hysterectomies were performed; 459 (43%) vaginal, 561 (52.6%) laparoscopic/robotic, and 47 (4.4%) abdominal. There were 530 hysteroscopies. Overall median OMEs decreased from 225 prior to July 2018 to 75 after July 2018 (<i>p</i> &#x3c; 0.0001). The opioid refill rate remained unchanged at 7.4% (<i>p</i> = 0.966). In Florida, there were 769 hysterectomies; 241 (31.3%) vaginal, 476 (61.9%) laparoscopic/robotic, and 52 (6.8%) abdominal. There were 549 hysteroscopies. Overall median OMEs decreased from 150 prior to July 2018 to 0 after July 2018 (<i>p</i> &#x3c; 0.0001). The opioid refill rate was similar (7.8% before July 2018 and 7.3% after July 2018; <i>p</i> = 0.739). <b><i>Limitations:</i></b> Limitations include involvement of a single hospital institution with a total of 10 fellowship-trained surgeons and biases inherent to retrospective study design. <b><i>Conclusions:</i></b> Legislative and provider-led changes coincided with decreases in opioid prescribing after 2018 in both states without increasing rates of refills and showed actual data reflected in the medical record. Gynecologists must actively participate in safe prescribing practices to decrease opioid dependence and misuse.

The content of inhibitors of proteolytic enzymes in bean flour, in puree from beans harvested in water and in puree from cereal beans welded in milk whey after soaking in water is determined. The use of whey as a liquid medium for cooking grain beans after soaking is substantiated. The technology of semi-finished product in the form of bean puree, providing for soaking beans in water, cooking in milk whey, coarse grinding of the bean brought to the ready, subsequent fine grinding and cooling is developed. It has been found that the activity of trypsin inhibitors in bean puree is much lower than in bean flour, which makes mashed potatoes a more preferred semi-finished product for the production of a wide range of culinary products.

<p>The advent of technology has improved the way statistics is taught and learned. It is claimed that the use of computer-based instructional tools can actively explore the meaning of statistical concepts among the students, as well as enhance their learning experiences. This study aimed to compare three methods of statistical analysis namely, the traditional technique (use of the calculator), Microsoft Excel and Statistical Package for Social Sciences (SPSS) software. This investigation utilized the experimental design, specifically the One-Group Pretest – Posttest Design. There were six education students who self-assessed their attitude before and after the introduction of the use of various computation techniques and performed the statistical analysis considering also the completion time required for each process. Results of the study revealed an increase in the level of attitude among the respondents form the pretest to the posttest. Also, the cognitive level regardless of the approach was very high. However, the t-test failed to establish a significant difference in the attitude among the respondents. On the other hand, there were significant differences in both the test scores and completion time of the respondents in the three methods in favor of SPSS.</p><p> </p><p><strong>Keywords: </strong>Information technology, statistics, traditional technique, Microsoft excel, SPSS, comparative analysis, experimental research design, Davao City, Philippines. </p>

From among a wide variety of protein purification techniques affinity chromatography has proved to be particularly effective for separation of proteolytic enzymes and their inhibitors. In this article, following a general description of affinity adsorbents used for purification of proteinases, we overview a simple separation procedure for some serine proteinases and their inhibitors by way of affinity chromatography in the presence of high NaCl concentration. It has been shown that some highly specific trypsin inhibitors exhibit also antichymotrypsin activity when high concentration of Na(+) but not K(+) or Li(+) ions are present in the reaction mixture. Taking advantage of this phenomenon the virgin forms of trypsin inhibitors from squash seeds, Kazal-type inhibitor from porcine pancreas and alpha(1)-proteinase inhibitor from human and sheep plasma, as an example, were separated using immobilized chymotrypsin or its inactive derivative methylchymotrypsin in the presence of 5 M NaCl.
