Long-term use of green manure legume and chemical fertiliser affect soil bacterial community structures but not the rate of soil nitrate decrease when excess carbon and nitrogen are applied

Legumes add not only nitrogen (N), but also carbon (C) to soils, so their effects on the soil microbial community may be different from those of chemical fertiliser. Soil microbes often compete with plants for N when excess C is applied due to their increased N immobilisation potentials and denitrification. In the present study we evaluated the effects of the 9-year use of a green manure legume (hairy vetch; Vicia villosa) in a greenhouse tomato system on soil microbial community structures as well as on the decrease of nitrate when rice straw was incorporated into the soil. Soil microbial community structures and their diversity were altered by the long-term use of legumes. The ratios of Acidobacteria, Gemmatimonadetes and Proteobacteria increased in the hairy vetch soils. The rates of decrease in nitrate were similar in soils with a history of chemical fertiliser and hairy vetch, following the addition of rice straw. In addition, during incubation with added rice straw, the difference between the two soil microbial community structures became less clear within 2 weeks. Thus, we conclude that even though growing a green manure legume changed soil bacterial community structures, this did not result in relatively faster loss of available N for plants when rice straw was added to the soils.

Long-term organic farming aims to reduce synthetic fertilizer and pesticide use in order to sustainably produce and improve soil quality. To do this, there is a need for more information about the soil microbial community, which plays a key role in a sustainable agriculture. In this paper, we assessed the long-term effects of two organic and one conventional cropping systems on the soil microbial community structure using high-throughput sequencing analysis, as well as the link between these communities and the changes in the soil properties and crop yield. The results showed that the crop yield was similar among the three cropping systems. The microbial community changed according to cropping system. Organic cultivation with manure compost and compost tea (Org_C) showed a change in the bacterial community associated with an improved soil carbon and nutrient content. A linear discriminant analysis effect size showed different bacteria and fungi as key microorganisms for each of the three different cropping systems, for conventional systems (Conv), different microorganisms such as Nesterenkonia, Galbibacter, Gramella, Limnobacter, Pseudoalteromonas, Pantoe, and Sporobolomyces were associated with pesticides, while for Org_C and organic cultivation with manure (Org_M), other types of microorganisms were associated with organic amendments with different functions, which, in some cases, reduce soil borne pathogens. However, further investigations such as functional approaches or network analyses are need to better understand the mechanisms behind this behavior.

Rice straw is a byproduct of agricultural production and an important agricultural resource. However, rice straw has not yet been effectively used, and incorrect treatment methods (such as burning in the field) can cause serious damage to the environment. Studies have shown that straw returning is beneficial to soil, but there have been few studies focused on the effect of the amount of short-term straw returned on the soil microbial community. This study evaluates 0%, 50%, 75%, and 100% rice straw returned to the field on whether returning different amounts of straw in the short term would affect the diversity and composition of the soil microbial community and the correlation between bacteria and fungi. The results show that the amount of straw returned to the field is the main factor that triggers the changes in the abundance and composition of the microbial community in the paddy soil. A small amount of added straw (≤ 50% straw added) mainly affects the composition of the bacterial community, while a larger amount of added straw (> 50% straw added) mainly affects the composition of the fungal community. Returning a large amount of straw increases the microbial abundance related to carbon and iron cycles in the paddy soil, thus promoting the carbon and iron cycle processes to a certain extent. In addition, network analysis shows that returning a large amount of straw also increases the complexity of the microbial network, which may encourage more microbes to be niche-sharing and comprehensively improve the ecological environment of paddy soil. This study may provide some useful guidance for rice straw returning in northeast China.
