The objective of this study was to compare the efficacy of different oocyte retrieval schemes over a period of 10 weeks. Fifteen multiparous Angus cows were randomly assigned (n=3/group) to the following groups: 1) OPU once/week (7-day interval), 2) OPU twice/week (3- and 4-day interval, alternately), 3) Dominant follicle removal (DFR)+OPU once/week. DFR followed by OPU 72h later, 4) DFR+FSH+OPU once/week. DFR followed 36h later by FSH (Folltropin, Bioniche, Belleville, Ontario, Canada) (120mg s.c. and 80mg i.m. administered simultaneously) followed by OPU 48h later, 5) FSH+OPU twice/week. FSH followed by OPU1 30h later and OPU2 96h after OPU1. The interval between OPU2 and next FSH was approximately 42h. The follicles were aspirated using an Aloka ultrasound scanner (Wallingford, CT, USA) and a 5-MHz probe. The COCs were selected based on morphology and matured in TCM-199, supplemented with 10% FCS, 0.01 unitsmL−1 bFSH, 0.01 unitsmL−1 bLH and antibiotics. Fertilization (Day 0) was carried out with TALP-FERT medium containing capacitation factors. Frozen semen from the same bull was used (1×106mL−1) throughout. After 18h the presumptive zygotes were cultured in SOF with 5% FCS (Holm P et al., 1999 Theriogenology 52, 683–700). The embryos were evaluated based on IETS guidelines (Grades 1 and 2 selected). The data were analyzed by chi-square test and ANOVA. In all parameters, the DFR followed by FSH and subsequent OPU once/week protocol gave the best results on a per-cow-per-week as well as total (3 cows over 10 weeks) basis. Though OPU was done only once/week, this group produced more total oocytes (303) than groups where OPU was done twice/week, either with FSH (286) or without FSH (229) and also produced more total embryos on Day 8 (71 blastocysts, 23.4% of oocytes cultured) than the latter two groups (64, 22.4% and 49, 21.4%, respectively). Among the nonstimulated groups, the OPU twice/week group had more total oocytes (236) than groups with OPU once/week, either without DFR (137) or with preceding DFR (160). However, a preceding DFR seemed to have a positive effect on oocyte quality as this group had a better embryo development rate (26.9%), producing more total embryos (42). In comparison, OPU twice/week produced total 49 embryos (21.4%) and OPU once/week produced 26 (19.4%). In conclusion, DFR coupled with single-shot FSH administration can be used effectively over a period of at least 10 consecutive weeks and can increase (P<0.05) the oocyte yield by two-fold and embryo production following IVF by two and half-fold, as compared to routine OPU-IVF done once a week. In commercial bovine in vitro fertilization (IVF) companies, there is a continuous need to improve results. Efforts to maximize in vitro embryo production have included modifications in the use of sperm separation gradients. The development of commercially available sperm centrifugation gradients represents a new possibility of increasing the number of viable sperm that can be obtained from low concentration (fresh or frozen, sexed or unsexed) semen samples in order to improve the efficiency of the IVF system to make embryo production as efficient as possible. The objective of this study was to compare two different separation gradients, as follows: Group 1: Percoll (Sigma, St. Louis, MO, USA), in 45% and 90% gradients; Group 2: EquiPure (Nidacon, Gathenburg, Sweden), in top and bottom layers. Before and after separation, sperm were evaluated at 200× magnification for total motility, and then stained to assess viability at 400× with fast-green/eosin stain (Sigma). Sperm separation was performed using frozen/thawed semen from one bull. Semen was separated by centrifugation at 200g for 30 min in both density gradients. Results obtained from Groups 1 and 2 were compared by chi-square test. Sperm separation with Percoll yielded lower numbers of sperm (average sperm concentration after separation of 92 × 106, vs. 159 × 106 sperm/mL for EquiPure; P < 0.05) but resulted in higher motility (60% vs. 39%, respectively; P < 0.05) of separated sperm. Rates of live sperm cells were not significantly different between groups (69.5% vs. 70%, respectively; P > 0.1). These results indicate that the commercial separation medium EquiPure may be associated with higher sperm concentration levels but with lowered sperm motility when compared to Percoll for bovine sperm separation. However, Equipure provided similar percentages of live sperm when compared to Percoll, which is currently used in our laboratory.

The incidence of perineum rupture at Limboto CommunityHealth Center is 46,7%. The purpose of this study was to analyze theeffectiveness of gymnastics pregnancy against rupture perineum inMaternity at Limboto Community Health Center. The method of researchwas used pre-experimental research with statistical group comparisondesign with using chi square test. The population in this study were allthirty pregnant trimester III which amounted 30 people. The sample inthis study were 15 case groups and 15 control groups. The variablepregnant exercise was independent variabel and the dependentvariable was incidence of perineal rupture. The result of this researchby using chi square statistic test was showed t arithmetic 8,571, thatmean the respondent who have high pregnancy exercise did notexperience rupture perineum. Results p - value 0.009 with degree ofsignificance 0.05 (p < 0.05). The conclusion there was a significant effectof pregnancy exercise effectiveness on the incidence of perineal rupturein maternal mothers at Limboto Community Health Center.

We previously reported that superstimulatory (SS) treatment-induced follicular wave synchronization after ovum pickup (OPU) was effective in enhancing the quality of obtained oocytes and blastocysts derived from in vitro maturation (IVM) and fertilization (IVF; Imai et al. 2010 Reprod. Fertil. Dev. 22, 296). The present study was designed to examine the efficiency of embryo production by 4 sessions of OPU-IVF using a series of the SS treatment-induced follicular wave synchronizations. For the SS protocols, 3 consecutive SS (3CSS) and 2 separated SS (2SSS) were used. In the 3CSS group, the first OPU was performed on random days of the oestrous cycle (Day 0) and all follicles larger than 2 mm in diameter were aspirated. On Day 5, follicles larger than 8 mm in diameter were aspirated and a CIDR (InterAg, Hamilton, New Zealand) was inserted. The cows then received 20 armour units of FSH (Kawasaki-Seiyaku, Kawasaki, Japan) in twice-daily decreasing doses by IM injection from Day 7 to 10. Cloprostenol (PGF; 0.75 mg, Fujita-Pharm, Tokyo, Japan) was administered on the morning of Day 9. The second OPU was performed 48 h after PGF administration on Day 11; the CIDR was removed from the cows just before OPU. After the second OPU, donors were treated consecutively with the SS protocol mentioned above for the third and fourth OPU sessions. In the 2SSS group, donors received 2 sets of the SS treatment mentioned above, with an interval of 11 days between the second and the third OPU session. Four OPU sessions were performed every 11 days on all cows. In this study, 8 Japanese Black cows were divided into the 3CSS and 2SSS groups, and the treatment for each group was reversed after a 65-day interval as crossover trials. After OPU, Grade 1 and 2 oocytes were used for IVM and IVF, and putative zygotes were cultured as described by (Imai et al. 2006 J. Reprod. Dev. 52, S19–S29 suppl.). A part of the zygotes were cultured in a micro-well system. Data were analysed by Student’s t-test and chi-square test. There were differences (P < 0.05) in the mean (±SD) number of follicles, collected oocytes, and cultured oocytes in the 3CSS (35.0 ± 8.6 and 24.4 ± 11.2, respectively) and 2SSS (30.8 ± 10.5 and 20.2 ± 9.0, respectively) groups. There were no differences in mean percentage of blastocyst formation and Grade 1 blastocyst rates between the 3CSS (38.5 and 55.8%, respectively) and 2SSS (34.8 and 54.8%, respectively) groups. However, the mean number of blastocysts produced per OPU session was significantly (P < 0.05) higher in the 3CSS group (8.1 ± 6.3) compared with the 2SSS group (5.8 ± 4.4). These results indicate that a series of 3 consecutive SS treatments had greater efficiency in producing OPU-IVF embryos. To evaluate the effect of pFSH dose on the in vivo embryo production of Dorper ewes in the semi-arid northeast of Brazil, 40 sheep females were distributed into two groups of 20 animals that received intravaginal CIDR for 14 days, and two days before device removal, they received one of the following treatments: in the FSH200 group, the ewes received 200 mg of pFSH; and in the FSH128 group, the ewes received a total of 128 mg in decreasing doses every 12 h. Beginning 12 h after the conclusion of the treatments, estrus detection was performed every four hours using two Dorper rams of proven fertility. The ewes were mated at estrus onset and 24 hours later. Seven days after intravaginal device removal, the superovulatory response was evaluated, and embryo collection was performed using the laparotomy method. The recovered flushings were subjected to embryo searches under a stereomicroscope and classified according to their qualities. Analyses of variance (ANOVAs) and LSD tests were used to compare the different parameters. The data expressed as percentages were analysed by chi-square test. The ovulation rate was higher in the FSH200 group, which had 16.3 ± 0.3 corpora lutea (CL), than in the FSH128 group, which had 11.3 ± 0.3 CL (P<0.05). However, higher fertilization rate (83.6% vs. 62.4%) and higher transferable (86.0% vs. 71.6%) and freezable (67.9% vs. 40.8%) embryo rates were observed in the FSH 128 group compared with the group that received 200 mg. Furthermore, no significant differences in the remaining parameters were observed between the experimental groups (P>0.05), demonstrating that pFSH dose reduction promoted a greater production of freezable and transferable embryos in Dorper ewes subjected to MOET.

<p class="0abstract">With the increasing relevance of mobile apps for companies, push notifications to address app users are also becoming more important. While the acceptance factors of push notifications have already been extensively researched, the effect of the different design elements on user interaction by opening the mobile app is still completely unexplored. Based on existing scientific findings from related fields, especially banner advertising and e-mail marketing, the authors first develop hypotheses on the effect of title, button and image on user interaction with push notifications. In several experiments the hypotheses are tested using the example of a mobile shopping app. The results are evaluated using Chi-square test and Cramer's V. While the use of a title seems to have a positive effect on interaction rates, the hypotheses on the positive effect of buttons and images on interaction rates have to be rejected.</p>

Artificial insemination, especially with the use of frozen semen, is one of the important tools for embryo transfer program in sheep. The present study investigated the effects of insemination times, breeds, and two inseminators on the fertility of ewes intrauterinally inseminated with frozen–thawed ram semen imported from New Zealand. At 8 sheep farms located in Hokkaido, Japan, during the breeding season (October to December) in 2005, a total of 64 mature (1- to 6-year old) Suffolk (32 heads) and Polled Dorset (32 heads) ewes were used. The ewes were treated with controlled intravaginal drug release (CIDR containing 0.3 g progesterone; Pharmacia &amp; Upjohn, Ltd., Auckland, New Zealand) for 12 days and an injection of 500 IU equine chorionic gonadotropin one day before CIDR removal. The fixed-time intrauterine inseminations (early: 43–46 h; late: 47–50 h) after CIDR removal were performed using the frozen–thawed semen from a Suffolk and Polled Dorset ram by two inseminators. The effects of breeds (Suffolk and Polled Dorset), fixed-time insemination times (early and late phases), and two inseminators on pregnancy (number of pregnant ewes/number of ewes inseminated, 60 days after insemination) and lambing (number of lambed ewes/number of ewes inseminated) rates were analyzed by chi-square test. The prolificacy was compared by Student&apos;s t-test, and differences were also analyzed by Tukey&apos;s omega procedure. The effect of the different farms on fertility was not examined due to the small numbers of ewes per farm. Pregnancy (60.0 and 72.4%, respectively) and lambing (60.0 and 71.4%, respectively) rates were not significantly different between Suffolk and Polled Dorset ewes. The inseminators also did not affect pregnancy (62.6 and 68.8%) and lambing (62.6 and 67.7%) rates. For the insemination times, the lambing rate tended to be higher (P ≤ 0.10) in the early insemination than in the late insemination (76.7% and 53.6%, respectively). The present results show acceptable fertility in ewes inseminated with Suffolk and Polled Dorset frozen semen imported from New Zealand. The early intrauterine insemination (43–46 h after CIDR removal) tended to result in higher fertility than the late insemination (47–50 h after CIDR removal). From 38 lambed ewes, 60 newborn lambs were produced, and this has provided new blood lines of Suffolk and Polled Dorset sheep in Japan.

This study examined the effects of X-rays on bovine oocytes used for in vitro embryo production. In recent years, airport security has become more stringent and the use of X-ray screening may be required for all items, which could be problematic when transporting oocytes. Ionizing radiation such as X-rays are high-energy particles that can penetrate into the nucleus of a cell and cause single and double-strand breaks in the DNA chain. Oocytes exposed to X-ray radiation may have DNA damage affecting maturation, fertilization, and embryonic development. Bovine ovaries obtained from a local abattoir were used to collect immature oocytes by follicular aspiration. The oocytes were divided into a control group and an experimental group then placed in maturation medium. The oocytes in the experimental group were placed in a hospital X-ray machine with a single exposure of 100 mA, 120 kVp for 0.3 s. This exposure challenged the oocytes with a comparable, calculated amount of mrems equivalent to a single screening from an airport X-ray machine (30 mrems). After X-ray exposure, the experimental group was cultured with the control at 37°C and 5% CO2. After 18 h, cryopreserved bovine sperm were thawed and processed using BoviPure (Nidacon International, Mölndal, Sweden) density gradient centrifugation and the oocytes were inseminated with 1 × 106 sperm in fertilization medium at 37°C and 5% CO2. After 24 h, both groups were transferred to 50-μL droplets of G1 medium (Vitrolife, Göteborg, Sweden) at 37°C and 6% CO2 and both were transferred into G2 medium after 72 h. Development of the oocytes was scored after an additional 72 h. Results were based on 2 criteria: the percent cleaved (presumptive fertilization) and the percent cleaved that developed to morula and blastocyst stages. The results indicated no significant difference in the percentages of oocytes cleaving after insemination for the control and experimental groups (P = 0.403, chi-square test). However, there was a significant increase (P = 0.037, chi-square test) in the percentages of morulae and blastocysts developing in the control (non-irradiated) v. experimental (X-rayed) groups. The outcome of this study cautions on the potential damage caused to bovine oocytes used for embryo production in vitro when exposed to X-ray electromagnetic radiation during routine air transport screening operations. ABSTRAKInseminasi Buatan dengan menggunakan semen cair digunakan untuk daerah yang sulit nitrogen cair dan mempunyai kualitas yang lebih baik dari pada semen beku. Tujuan penelitian ini adalah untuk mengetahui kualitas berbagai bahan pengencer dasar air kelapa penyimpanan dingin 2-5°C.Penelitian ini dilaksanakan di Loka Penelitian Sapi Potong Grati, Pasuruan. Semen yang digunakan berasal dari sapi madura sebanyak dua ekor, berumur 5 dan 3 tahun  dan berat badan yaitu 397 kg dan 360,5 kg. Sapi madura ditampung seminggu 2 kali dengan motilitas > 70% , sedangkan air kelapa yang digunakan adalah air kelapa hijau yang masih muda. Pengenceran semen cair dibagi menjadi 4 yaitu P0 (CEP-3 + 20% kuning telur) sebagai kontrol, P1 (air kelapa hijau +20% kuning telur), P2 (P1 + 0,4% putih telur + 1% fruktosa) dan P3 (P1 + 0,4% putih telur kuning telur +2% fruktosa). Data dianalisis menggunakan uji Pearson’s Chi Square dan Uji Deskriptif. Hasil penelitian menunjukkan motilitas spermatozoa sesuai standar SNI yaitu motilitas> 40% pada pengencer CEP-3 dapat disimpan selama hari ke-8 (40,50±6,43%) sedangkan pada pengencer dasar air kelapa hijau pada P1, P2 dan P3 tidak menunjukkan perbedaan yang nyata (P>0,05) selama disimpan 6 hari yaitu 40,50±10,12%, 38,00±4,22%, 40,00±8,50%. Abnormalitas dari semua perlakuan menunjukan nilai <20%. Viabilitas didapatkan nilai tertinggi pada perlakuan P0(89,58±2,16%) kemudian P1(89,39±3,79%), P2(88,62±4,59%) dan P3(87,93±4,41%).Kata kunci: CEP-3, semen cair, sapi madura, simpan dingin, air kelapa hijau ABSTRACTArtificial Insemination using liquid semen is performed for areas that where liquid nitrogen is difficult to find and havng better quality than frozen semen. Research was conducted at Laboratory of Reproduction of Grati Beef Cattle Research Station, Pasuruan.Semen that is used comes from two madura bulls aged 5 and 3 years with body weight is 397 kg and 360.5 kg. The semen was collected twice a week with motility> 70%, and the coconut water used is unripe green coconut water. The research treatments were P0 (CEP-3 + 20% egg yolk) as control, P1 (unripe green coconut water + 20% egg yolk), P2 (P1+ 1% fructose + 0.4% egg white) and P3 (P1+ 0.4% egg white + 2% fructose). Data were analyzed using Pearson's Chi Square test and Descriptive Test. The results showed that the motility of spermatozoa was within Indonesian National Standard (SNI) with more than 40% motility in the CEP-3 diluent and it can be stored until the 8th day (40.50 ± 6.43%). The basic diluents of green coconut water at P1, P2 and P3 was not significantly affected (P> 0.05) until 6 days storing with the motility number average are 40.50 ± 10.12%, 38.00 ± 4.22%, 40, 00 ± 8.50%. Compared with conventional semen, use of sexed semen decreases the conception rate (CR); therefore, it is important to address the factors that lead to sperm damage. Recently, a novel sexed semen technology was developed for improving dairy cattle fertility (Betthauser et al. 2016 J. Dairy Sci. 99 (E-Suppl. 1), 534). However, there are few reports about the fertility of this novel sexed semen in dairy cattle in Japan. The objective of this study was to compare the CR of traditional sorting semen (S1: flow cytometry/cell sorting) and the novel sexed semen (S2: SexcelTM, ABS Global Inc.) in Holstein heifers and cows in Japan. The CR for the first insemination was obtained from 391 Holstein cows and 148 heifers (10.7-17.8 months old) from 14 dairy farms in Hokkaido from June 2017 to April 2019. Semen used for AI was collected from six bulls and packaged into 0.25-mL straws and frozen. Cows inseminated with conventional frozen semen (S3: unsexed semen in 0.5-mL straws) were used as controls. Calving data were collected from 123 cows and 69 heifers that had calved between January 2018 and May 2019. The diagnosis of pregnancy was confirmed using ultrasonography between 30 and 45 days after insemination. The CR and sex ratio were analysed using chi-square test. The average parity number was 1.9±1.1, and average days open was 84.9±20.4 days. The average interval between calving and the first service was 86.2±20.3 days. The average milk yield (at 305 days) was 12 195±1595 kg. All of the animals were inseminated after the onset of standing heat or removal of the tail chalk of natural heats and oestrus and ovulation synchronisation programmes on the farm. Conception rates of heifers of the first service of S1 (n=54) and S2 (n=94) were 68.5 and 70.2%, respectively. Conception rates of cows of the first service of S1 (n=32), S2 (n=173), and S3 (n=186) were 56.3, 57.8, and 53.8%, respectively. There were no significant differences in CR between these groups (P&gt;0.4). The proportion of female calves produced using S1 (n=61) and S2 (n=57) was 91.8 and 93.0%, respectively, which was significantly more (P&lt;0.01) compared with the 50.0% produced using S3 (n=74). There were no significant differences between S1 and S2 (P&gt;0.8) in terms of the proportion of calves produced. The results of this study demonstrated that the traditional sorting and novel sexed semen (S1 and S2, respectively) did not differ in terms of the first-service CR and produced considerably more female calves than when the conventional semen (S3) was used. Furthermore, these findings indicate that the use of this novel sexed semen could achieve a sex ratio close to 90% female offspring.
