Ovary of the southern hairy-nosed wombat (Lasiorhinus latifrons): its divergent structural organisation

The organisation of the ovarian interstitial tissue in the southern hairy-nosed wombat Lasiorhinus latifrons was investigated. Unlike in most other marsupials, the outer cortical region of the ovary contains abundant luteinised interstitial tissue that largely occurs in discrete lobules, many of which contain a localised area of non-cellular, highly eosinophilic and periodic acid-Schiff-positive material. The findings suggest that the latter arises from the zona pellucida that surrounded the oocyte in growing follicles and that the luteinised interstitial tissue thus developed from transformed theca interna of degenerated atretic follicles. It is hypothesised that this tissue synthesises and secretes progestogens, which may result in the long, and variable, oestrous cycle length that has been found to occur in this species.

The Bidder's organ and ovary of the Brazilian toad Bufo ictericus were studied by light microscopy, using hematoxylin-eosin (HE) and periodic acid Schiff (PAS) staining. The expression and distribution of carbohydrate moieties was analyzed by lectin histochemistry, using 8 lectins with different carbohydrate specificities: Ulex europaeus (UEA I), Lens culinaris (LCA), Erythrina cristagalli (ECA), Arachis hypogaea (PNA), Ricinus communis (RCA I), Aleuria aurantia (AAA), Triticum vulgaris (WGA), and Glycine maximum (SBA). The results showed that the Bidderian zona pellucida presented alpha-mannose, alpha-L-fucose, beta-D-galactose, N-acetyl-D-glucosamine, and alpha/beta-N-acetyl-galactosamine residues. The Bidderian follicular cells showed the presence of beta-D-galactose and N-acetyl-D-glucosamine. In the extracellular matrix, alpha-mannose and alpha/beta-N-acetyl-galactosamine residues were detected. The ovarian zona pellucida showed alpha-L-fucose, N-acetyl-D-glucosamine, alpha/beta-N-acetyl-galactosamine residues, and alpha-mannose and N-acetyl-D-glucosamine residues were detected in the follicular cells. Thus, the zona pellucida in both organs contains N-acetyl-D-glucosamine, and alpha/beta-N-acetyl-galactosamine residues. alpha-L-fucose residues were detected in the zona pellucida of both organs, using different lectins. Considering that beta-D-galactose residue was absent from ovary but present in the Bidder's organ, this sugar residue may play an important role in follicle development, blocking the Bidderian follicles and preventing further development of the Bidder's organ into a functional ovary.

Transforming growth factor-β (TGF-β) is a potent fibrotic factor responsible for the synthesis of extracellular matrix (ECM) and is implicated as the major determinant in pathogenesis of chronic fibroses, including kidney. The novel small compound SMP-534 reduced ECM production induced by TGF-β in fibroblast cells. SMP-534 inhibited TGF-β-induced p38 mitogen-activated protein kinase (p38) activation but did not inhibit epidermal growth factor (EGF)-induced extracellular signal-related kinase (ERK) activation. We also found that oral administration of SMP-534 dose dependently lowered hydroxyproline contents in the cortical region of the kidney in rat anti-Thy-1 nephritis models. In periodic acid-Schiff staining of kidney sections, ECM accumulation was reduced by SMP-534 treatment. These data indicate that SMP-534 has potential in therapy for fibrotic diseases, including nephropathy.

AbstractSpinocerebellar ataxia type 34 (SCA34) is an autosomal dominant inherited ataxia due to mutations in ELOVL4, which encodes one of the very long-chain fatty acid elongases. SCA38, another spinocerebellar ataxia, is caused by mutations in ELOVL5, a gene encoding another elongase. However, there have been no previous studies describing the neuropathology of either SCA34 or 38. This report describes the neuropathological findings of an 83-year-old man with SCA34 carrying a pathological ELOVL4 mutation (NM_022726, c.736T>G, p.W246G). Macroscopic findings include atrophies in the pontine base, cerebellum, and cerebral cortices. Microscopically, marked neuronal and pontocerebellar fiber loss was observed in the pontine base. In addition, in the pontine base, accumulation of CD68-positive macrophages laden with periodic acid-Schiff (PAS)-positive material was observed. Many vacuolar lesions were found in the white matter of the cerebral hemispheres and, to a lesser extent, in the brainstem and spinal cord white matter. Immunohistological examination and ultrastructural observations with an electron microscope suggest that these vacuolar lesions are remnants of degenerated oligodendrocytes. Electron microscopy also revealed myelin sheath destruction. Unexpectedly, aggregation of the four-repeat tau was observed in a spatial pattern reminiscent of progressive supranuclear palsy. The tau lesions included glial fibrillary tangles resembling tuft-shaped astrocytes and neurofibrillary tangles and pretangles. This is the first report to illustrate that a heterozygous missense mutation in ELOVL4 leads to neuronal loss accompanied by macrophages laden with PAS-positive material in the pontine base and oligodendroglial degeneration leading to widespread vacuoles in the white matter in SCA34.

The amoebocytes, which form the most abundant type of blood-cell in Rhodnius, contain rounded, oval, or rod-shaped inclusions which stain by the periodic acid/Schiff method. These are believed to be neutral mucopolysaccharides. The amoebocytes apply themselves to the basal membranes, which likewise are PAS-positive, and appear to contribute to these membranes by discharging their contents. They insinuate themselves into the developing muscles and give rise to the sheaths of connective tissue by which the muscle fibres are surrounded. And they collect around deposits of injected Indian ink, producing similar sheets of PAS-positive material, presumably mucopolysaccharide. The chitinous endocuticle is PAS-negative (except in a few special regions such as the neck and the conjunctival membranes of the limbs). It becomes positive during digestion by the moulting fluid. The tracheae react similarly. Other PAS-positive structures which are not produced by the amoebocytes are the striated border of the mid-gut, the basement membrane of the gut, the perilemma around the ganglia and nerves, and the ground substance within the ganglia.

The study has been conducted on the testes of 10 adult Large White Yorkshire pig (1.5-2) years of age. The testis was surrounded by a capsule made up of dense irregular connective tissue comprising three layers viz., outer visceral layer of tunica vaginalis, middle tunica albugenia and inner tunica vasculosa. The connective tissue trabeculae were extended from the capsule and divided the parenchyma of the testis into number of lobules and consisted of collagen, elastic and reticular fibers along with rete testis and blood vessels. The Periodic Acid–Schiff shows strong affinity in basement membrane of seminiferous tubules and tunica albuginea with a moderate activity in Leydig cells. The Alcian blue shows strong affinity in basement membrane of seminiferous tubule and weak activity was observed in tunica albuginea and interstitial tissue. The different histomorphometrical parameters with regard to thickness of testicular capsule, diameter and height of seminiferous tubules were found to be higher in the left testicle.
