Porcine zonae pellucidae immunisation of tammar wallabies (Macropus eugenii): fertility and immune responses

This study looked at the feasibility of targeting the zona pellucida for a contraceptive vaccine as a possible alternative method of control for overabundant macropods. Tammar wallabies, as a model for other macropods, were immunized with porcine zonae pellucidae (PZP) and were found to achieve significant concentrations of antibody to PZP in sera and reproductive tract fluids. Wallabies immunized with PZP exhibited lower ovarian weight with reduced numbers of antral follicles when compared with control animals. Wallabies were placed in a natural mating trial followed by an artificial insemination trial. None of the PZP-immunized wallabies produced offspring in the natural mating trial compared with 67% of control animals. To further assess fertility, a sub-sample of the wallabies were superovulated and artificially inseminated. This resulted in all control wallabies producing fertilized ova and all PZP-immunized wallabies failing to ovulate. These results suggest that immunocontraception based on targeting antigens of the zona pellucida may be an effective strategy for fertility reduction in macropods.

Introduced marsupial brushtail possums (Trichosurus vulpecula) are a major pest in New Zealand because of their impacts on conservation values and agricultural production. Immunologically-based fertility control (immunocontraception) offers an effective and humane alternative approach to possum management. The zona pellucida (ZP) is an extracellular coat around all mammalian eggs and an attractive target for the development of immunocontraceptive vaccines. Antibodies against ZP are ovary-specific and act by preventing sperm from binding and penetrating the ova and/or by disrupting the development of follicles or early embryo. The aim of these studies was to test the efficacy of possum-derived ZP antigens for their ability to elicit sustained immune responses and cause infertility, and to assess a range of options for development of a bait-delivered contraceptive vaccine. Alloimmunisation with possum ZP2 and ZP3 proteins showed that self-ZP antigens elicited strong humoral immune responses and reduced the fertility of female possums by 72-80%. Several potentially possum-specific immunocontraceptive peptides have been identified by linear epitope mapping and amino acid alignment and are being tested for their ability to reduce fertility. Recent trials have demonstrated that possums mount immune responses against ZP antigens delivered in transgenic plants and bacterial ghosts. Research on antigen and specific peptide identification, non-target effects and delivery systems is ongoing. Ovaries from a marsupial, the tammar wallaby (Macropus eugenii), were grafted into a eutherian recipient at known stages of development to ascertain whether normal development would occur. Xenografted ovaries from pouch young < 20 days old, before the onset of meiosis, retained few germ cells and developed tubule-like structures reminiscent of seminiferous cords. Ovaries from 50-day-old pouch young, which contain primordial follicles, developed into antral follicles and corpora lutea within the eutherian host, and produced hormones that stimulated the reproductive tract of the host. The timing of onset of antrum formation and the progress of follicle development were advanced relative to the timing of events in ovaries in situ. Frozen-thawed ovaries from 50-day-old donors developed into preantral follicles, but at a reduced rate and number. This finding shows that gonads of a marsupial species can develop as xenografts in a eutherian, forming large antral follicles. Accelerated follicular development in xenografts provides a potentially valuable model for studying the factors that control follicle development. Assisted reproduction of endangered marsupials may also be feasible using follicles from pouch young grown as xenografts in a eutherian host.

The distribution of spermatozoa and seminal plug in the reproductive tract and the timing of ovulation were examined at various times in a naturally mated monovular macropodid marsupial, namely the tammar wallaby (Macropus eugenii). After the first post partum (p.p.) mating, 28 females were isolated and their reproductive tracts dissected at 0.5, 6, 18, 36 and 40 h post coitum (p.c.). Each tract was ligated into 13 major anatomical sections and spermatozoa and eggs were recovered by flushing. Mating was possibly delayed by handling and occurred 21.7 ± 2.5 h p.p. in these animals. Copulation lasted 7.8 ± 0.7 min. Within 0.5 h after a single mating, the tract contained 25.8 ± 10.2 × 106 spermatozoa and 21.6 ± 8.8 g of seminal plug, 96% and 70% of which was lost within 6 h p.c. respectively. Spermatozoa reached the uterus, isthmus and ampulla of the oviduct on the side of the developing follicle within 0.5, 6 and 18 h p.c., respectively, and a uterine population of 26.1 ± 12.103 spermatozoa was maintained for over 40 h. Sperm numbers were reduced at the cervix (up to 57-fold) and uterotubule junction (eight-fold) and only one in approximately 7500 ejaculated spermatozoa (3.4 ± 0.9 × 103) reached the oviduct on the follicle side. Differential transport of spermatozoa was not observed. Although the numbers of spermatozoa were reduced in the parturient uterus, they were highly variable and were not significantly different to those in the non-parturient uterus. Ovulation and recovery of sperm-covered eggs from the isthmus occurred 36–41 h p.c. (49–72 h p.p.). In contrast with the polyovular dasyurid and didelphid marsupials, the tammar wallaby ejaculates large numbers of spermatozoa, but transport is relatively inefficient and sperm storage in the tract before ovulation is limited.

Despite many efforts, the control of reproduction in feral cat populations is still a problem in urban regions around the world. Immunocontraception is a promising approach; thus the present study examined the suitability of the widely used pig zona pellucida proteins (pZP) for contraception in feral domestic cats. Purified zona pellucida proteins obtained from pig and cat ovaries were used to produce highly specific antisera in rabbits. Antibodies against pZP raised in rabbits or lions were not effective inhibitors of either in vitro sperm binding (cat spermatozoa to cat oocytes) or in vitro fertilization in cats, whereas antibodies against feline zona pellucida proteins (fZP) raised in rabbits showed a dose-dependent inhibition of in vitro fertilization. Immunoelectrophoresis, ELISA and immunohistology of ovaries confirmed these results, showing crossreactivity of anti-fZP sera to fZP and to a lesser extent to pZP, but no interaction of anti-pZP sera with fZP. It is concluded that cat and pig zonae pellucidae express a very small number of shared antigenic determinants, making the use of pZP vaccine in cats questionable. A contraceptive vaccine based on feline zona pellucida determinants will be a better choice for the control of reproduction in feral cats if immunogenity can be achieved.

Synchronized estrus and ovulation were induced during the anestrous season (April–May 1974) in 373 ewes of three synthetic (one sire and two dam) strains and two unselected (Suffolk and Finnish Landrace) purebred strains by treatment with 30 mg fluorogestone acetate (FGA) impregnated in polyurethane intravaginal sponges for 12 days. Following sponge removal each ewe received 500 IU pregnant mares’ serum gonadotrophin (PMSG) IM. Of these, 167 were bred by artificial insemination (AI) at 48 and 60 h post sponge removal with 0.2 ml raw unextended semen collected by electroejaculation (EE). Five days after AI, ewes were exposed to a follow up ram for 16 days for subsequent mating if a second estrus occurred. The remaining 206 were exposed to rams for a period of 22 days for natural mating. Blood samples were collected from 69 ewes, 9, 19 and 27 days post sponge removal and analyzed for progesterone to ascertain if corpora lutea were formed and whether the ewes recycled. The age of ram by mating method interaction significantly affected both fertility and fecundity mainly because some of the younger rams lacked libido and experience for natural mating. There were no significant differences in prolificacy due to any of the main effects tested. Among the 69 ewes examined for progesterone levels, 93% had formed corpora lutea after hormone treatment and 16% recycled. Only 16 of the 255 ewes that did not conceive to the synchronized estrus lambed to the subsequent estrus.

The viscous seminal plasma (SP) is currently a major impediment to the handling of ejaculate and the development of some biotechnologies in South American camelids. The vas deferens-collected spermatozoa of alpacas is a useful technique to avoid this problem. On the other hand, SP contains a large protein component that has been implicated in the function of spermatozoa within the female reproductive tract. In this sense, the low fertility achieved using transcervical insemination with frozen-thawed spermatozoa in alpacas could be improved by adding SP. This study aimed to evaluate the effect of the whole SP on some in vitro parameters of alpaca spermatozoa after the freezing-thawing-process and the fertility after artificial insemination. It would contribute to a better understanding of the interaction between thawed sperm cells and SP. Spermatozoa were obtained by surgically diverted vas deferens. The samples were diluted with a Tris-based extender, packaged in straws, and frozen. At thawing, each straw was divided into two post-thawing conditions: with the addition of 10% of PBS (control) or with 10% SP (treatment). The sperm cells were evaluated using dynamic parameters, sperm cell morphology, and morphometry. Fertility was assessed by an artificial insemination trial. All in vitro parameters were analyzed by ANOVA. A heterogeneity test was scheduled for the fertility trial. After the freezing-thawing process, motility and plasma membrane functionality was improved when SP was added. No differences were found for post-thaw viability between the control and treatment samples. The percentage of normal cells was higher with SP at post-thawing, and a decrease of the presence of bent tailed spermatozoa with a droplet in the SP group was observed. The length of the head spermatozoa was 3.4% higher in the samples with PBS compared to those in which SP was added. Females pregnant at day 25 post-insemination were 0/12 (with SP inside the straw) and 1/10 (without SP inside the straw). In conclusion, the presence of 10% SP at post-thawing improves sperm cells' motility, functionality, and morphology, indicating that it would be beneficial to improve the frozen-thawed alpaca's physiology spermatozoa. More fertility trials must be developed to increase this knowledge.
