Influence of early post partum ovulation on the re-establishment of pregnancy in multiparous and primiparous dairy cattle

To investigate the influence of early ovulation after calving on subsequent fertility, milk progesterone concentrations were monitored twice weekly in multiparous (n = 87) and primiparous (n = 60) Holstein/Friesian dairy cattle. Luteal activity before 21 days post partum was defined as ‘early’. In multiparous cows, calving-to-conception interval (CCI) was longer, services per cow were more numerous and conception rate to all inseminations was lower with early luteal activity (n = 41, 16·8±0·39 days post partum) than with later luteal activity (n = 46, 37·8±2·09 days post partum); multiparous cows with early luteal activity required more exogenous fertility treatments and were more likely to fail to conceive by 100 days post partum. In contrast, in primiparous cows, no significant relationships were detected between early (n = 32) or later (n = 28) luteal activity and the CCI, number of services per cow or conception rates to all inseminations. However, primiparous cows that ovulated before 21 days post partum had fewer normal progesterone profiles. Early ovulation after calving was associated with reduced pregnancy rates and prolonged CCIs in multiparous but not in primiparous dairy cows. Extra keyword: luteal function.

AbstractControlled breeding schemes for oestrous detection constitutes a proactive technical response that balances the infrastructural requirement for a profitable dairy operation and the demands for optimal animal performance. The present study compared (a) the reproductive response of a treatment based on a short vs longer-acting PGF2α analogue (tiaprost vs luprostiol), and (b) the reproductive response after a treatment of GnRH-PGF2α vs PGF2α alone for synchronizing dairy cows. Holstein-Friesian cows averaging 9000 kg milk/lactation and fed according to their requirements were used in the study. Cows were cyclic, at least 60 days post partum and were clinically sound before being considered for the experiments. In Experiment 1, animals were synchronised using an i.m. injection of either 15 mg of luprostiol or 0.75 mg of tiaprost, based on ultrasonic diagnosis of a corpus luteum. Animals were inseminated at observed oestrus. In Experiment 2, cows were synchronised, at random, by either an injection of 10pg ofbuserelin (day 0) followed by 0.75 mg of tiaprost at day 7 (GnRH-PGF2α) orjust 0.75 mg of tiaprost (PGF2α). For both treatments only cows with an ultrasonically detected corpus luteum were treated. Animals were inseminated at oestrus. At the time of treatment and again 3 days later, milk samples were collected and assayed for progesterone by RIA. Cows with progesterone concentrations >1 ng/ml were considered to have corpus luteum. Luteolysis was considered to have occurred when concentrations of progesterone were > 1 ng/ml at day 0 and <0.8 ng/ml at day 3. In Experiment 1, both analogues gave similar results in terms of induced luteolysis [luprostiol: 36/39 (92.3%) vs tiaprost: 36/41 (87.8%)], oestrous detection efficiency [luprostiol: 26/36 (72.2%) vs tiaprost: 30/36 (83.3%], oestrous distribution [day 2, 3 and 4, respectively: luprostiol: 26.9%, 50.0%, 19.2% vs tiaprost: 36.7%, 50.0%, 13.3%], and conception rates [luprostiol: 12/25 (48.0%) vs tiaprost: 14/28 (50.0%); P>0.05]. In Experiment 2, oestrous detection efficiency, interval to oestrus and conception rate were similar between treatments [97/149 (65.1%), 71.1 h, 43/95 (45.3%) for PGF2α vs 130/188 (69.1%), 68.2h, 65/126 (51.6%) for GnRH-PGF2α, respectively]. However the oestrous distribution was more concentrated in GnRH-PGF2α treated animals (P<0.01).

This trial evaluated the reproductive performance in an early routine oestrus induction programme using two different PGF2α preparations in dairy cattle. D-cloprostenol sodium (n = 192; Group A) or dinoprost (n = 187; Group B) was administered between days 35 and 42 post partum. Also, a group of non-treated cows (n = 135; Group C) was included as control. Pedometers were used to detect oestrus, and also secondary oestrous signs and vaginal mucus quality were assessed prior to artificial insemination (AI). When oestrus was not detected for 14 days after PGF2α administration, the treatment was repeated, up to a maximum of three times. There were no differences between the study groups in oestrus detection (A = 73.48%, B = 73.01%, C = 79.26%; P = 0.428), good mucus quality (A = 96.45%, B = 91.30%, C = 93.45%; P = 0.203) and the presence of mounting lesions (A = 98.58, B = 94.93%, C = 98.13; P = 0.414). First-service pregnancy rates were 19.78%, 15.64% and 32.03% in Groups A, B and C, respectively (P = 0.003). There were no inter-group differences for the interval from parturition to first AI. However, a significantly shorter interval from parturition to conception (92.17 days, 99.45 days, 118.93 days; P = 0.002) and significantly less services per conception (2.12, 2.18, 2.66; P = 0.003) were observed in Groups A and B in comparison with Group C. The use of PGF2α resulted in better fertility in a repetitive, routine postpartum programme, although no differences between Dcloprostenol and dinoprost were detected.

This paper presents results of investigations of gonadoreline application (14-16, 17-20 and 21-25 days post partum) influence, followed by prostaglanin F2 ? ten days later, on the reproductive parameters of dairy cows with difficult calving and endometritis. The study was carried out on a total of 92 Holstein-Friesian cows. Fifty of them treated with hormones, while 42 animals served as controls, receiving placebo injections. Analysis of the results in cows with postpartal endometrits revealed that mean anoestrus period, open days period and intervals between calving were significantly lower in cows that underwent hormonal treatment, when compared to the controls. The period from gonadoreline application, followed by prostaglandin injection, to conception was shorter by 44, 28 and 14 days respectively when compared to the control groups. The total conception rate from the first three artificial inseminations was higher in the experimental groups in comparison to the controls. .

Background: Milk production of cows is closely correlated with its reproductive efficiency. One of the several factors influencing the dairy efficiency is the decline in fertility of the cows/heifers. Use of hormonal protocol are often used to improve the fertility of the recipients. Several programs are available to inseminate dairy cows, with variation in the use of different types of hormones, time of use and range of duration period, as well as the time of AI. The objective of this study was to evaluate the efficiency of follicular wave emergence induction of estradiol benzoate in comparison to GnRH in a Heatsynch protocol and its influence on reproductive parameters.Materials, Methods & Results: In this study were used primiparous and multiparous Holstein cows (n = 325). Animals were divided randomly into two different treatments according with the hormone used to induce follicular wave emergence (D0). The GnRH group (n = 167), was given 25 mg, while in the EB group (n = 158) was used 2 mg. At same day, were inserted, a controlled internal drug-releasing containing 1.9 g of progesterone CIDR®. On day 7, CIDR was removed and cows were given 25 mg luteinizing hormone followed by an injection of 1 mg Inducer Ovulation at day 8. Cows were then observed for signs of estrus for at least two hours (twice daily at 7 am and 6 pm) for three days following CIDR® removal. Cows detected in estrus were inseminated 12 h later the estrus detection. Pregnancy status was determined by ultrasonography at 30 ± 5 d and 60 ± 5 d after AI. Ultrassonographic examinations with ultrasound, transrectal linear probe of 5 MHz, were performed for the diagnosis of pregnancy at 30 and 60 days after artificial insemination. Binomially distributed data, such as, estrous response, conception rate, pregnancy rate and pregnancy losses, were analyzed by Chi-square test. Pregnancy rates at 30-35 days and 60-65 days as well as pregnancy losses were considered in the model for the effect of parity (primiparous vs. multiparous) and the group of estrus behavior during the Heatsynch protocol. Conception rate was similar between GnRH (36.1%, 57/158) and EB (37.7%, 63/167) treatments. Cows showing estrus signs after CIDR removed were 71.9% in GnRH and 62% in EB group (P = 0.05). Embryonic losses were different (P = 0.03) according to groups of cows showing estrus signals or not.Discussion: Our results demonstrate that conception rates are not affected by using either EB or GnRH at the beginning of the Heatsynch protocol. Therefore, the EB can be an alternative choice to GnRH at Heatsynch beginning. Similar results were found by autor in dairy cows, once it was also observed that there is no difference in conception rate in two TAI protocols using either GnRH or EB. In Heatsynch protocols the increase in estrus detection are a key factor to increase fertility in lactating dairy cows. It can be explained once animals in regular estrous cycle have a palpable corpus luteum and circulating progesterone concentration higher than 1 ng/mL which leads than to enhance pregnancy ratios. The use of BE at the time of CIDR insertion in TAI protocols for dairy cattle proved to be an alternative to GnRH considering conception rates. Therefore, the use of BE at the time of CIDR insertion in a Heatsynch protocol in dairy cattle proved to be an alternative to GnRH which can be an important tool to increase the TAI utilization by farmers. Furthermore, in Heatsynch protocol cows showing estrus have pregnancy rates improved and lower pregnancy losses.

In early lactation dairy cows use stored body fat to provide an additional source of energy to support milk production and these reserves are replaced when energy requirements for milk synthesis are reduced. The extent to which body fat is mobilized depends on the nutritional state of the animal and its genetic potential. These changes are accompanied by substantial alterations in the intermediary metabolism of adipose tissue. The aim of this work was to investigate the timing and magnitude of changes in synthetic and lipolytic pathways in adipose tissue from dairy cows during a lactation cycle and at three levels of energy intake.Fifty four cows were fed grass silagead libitumand a concentrate ration at 3(L), 6(M) or 9(H) kg DM/day (Sutton et al 1992). An initial group of six cows was slaughtered at 2 to 4 dayspost partum(week 0) followed by two cows from each treatment at weeks 2, 5, 8, 11, 14, 19, 24 and 29.

Lactating crossbred Holstein-Friesian dairy cows (n = 331) were started on an Ovsynch regimen 68 ± 8.2 days after calving; 200 µg GnRH intramuscularly (i.m.) on Days 0 and 9, and 35 mg prostaglandin F2a i.m. on Day 7. Thirty-eight and 31 cows (11.5 and 9.4%, respectively) were in oestrus on Days 0 to 6 and 7 to 8, respectively, and inseminated, and the remainder were fixed-time inseminated (on Day 10). For these three groups, pregnancy rates (60-65 days after breeding) were 31.6, 38.7 and 34.0%, respectively (P = 0.82) and calving rates were 100, 100 and 89.9% (P = 0.23). In a preliminary trial, twelve lactating cows (45 to 60 days postpartum) with inactive ovaries were given 1500 IU eCG i.m.; 10 were in oestrus within 10 days after treatment (and inseminated) and eight of these were pregnant (30 days after breeding). The Ovsynch program resulted in acceptable reproductive performance in cyclic cows and eCG treatment has considerable promise for inducing oestrus in anoestrous cows.

Abstract The aim of the study was to find the relationship between some postpartum disorders and pregnancy losses (PL), and the influence of PL on further fertility of Holstein-Friesian dairy cows. The 1,056 pregnancies and 906 calving intervals (CI) of 378 cows during the nine-year observation period were analysed. The average milk yield during this period increased from 7,500 to 10,500 kg/305 d of lactation. All cows up to 4 weeks after calving were gynecologically examined and cases of genital tract disorders were recorded and treated. Artificial inseminations (AI) were made using commercial frozen/thawed semen. In cows with silent ovulation, oestrus was induced by injection of PGF2α or programmes using PGF2α and GnRH, or hCG were applied. Pregnancy was diagnosed with the use of ultrasonography or foetal membrane slip only from 30 and 42 d after service, respectively, and re-diagnosed between 70 and 90 d after AI. The cases of „early foetal loss” up to 90 d and „mid-to-late PL” between 91 and 260 d after AI were recorded. Endometritis was noted in 31.8%, ovarian cysts in 8.5%, whereas ovarian inactivity in 4.6% of cows. Early PL was observed in 5.2%, whereas mid-to-late PL in 6.8% of cows (total 12%). From all analysed factors, the influence of delayed or periodical lack of ovarian activity was significant with regard to early PL (P=0.029). The probability of early PL increased when besides the diagnosis of ovarian inactivity, AI was made following an induced oestrus (P=0.003). CI for cows, which did not lost pregnancy amounted 442.9 d. The early, mid-to-late and multiple PL extended the CI to 579.6, 661.1 and 657.7 d, respectively (P<0.05). Concurrently, CI for cows with “early foetal loss” was significantly shorter comparing to cows with “mid-to-late foetal loss” (579.6 d vs.661.1 d; P<0.05). On average 2.6 inseminations for one pregnancy were needed, and each PL increased the AI index. Significant differences were stated after early and multiple foetal losses (5.2 and 5.1 vs. 2.6; P<0.05).

AbstractIn order to investigate the rationale of manipulating post-ovulation progesterone (P4) concentrations, luteal activity was measured in 10 Holstein-Friesian cows treated with a progesterone-releasing device (CIDR, 1·9 g P4) inserted on the morning of day 2 post-oestrus for a period of 7 days and compared with 10 untreated control animals. Milk samples were collected daily during afternoon milking from 7 days pre- to 24 days post-oestrus and P4 concentrations were measured using enzyme-linked immuno-sorbent assay (ELISA). The milk P4 profiles were used to assess the effects of early post-oestrus P4 elevation on five intervals of the luteal phase. In the treated (T) and control (C) animals, the mean interval from oestrus to P4 rise (≥3 μg/l), was 2·38 (s.e. 0·18) and 4·90 (s.e. 0·28) days, respectively, (P < 0·01). The mean interval from oestrus to the attainment of peak P4 concentrations was not significantly different (P > 0·05) between the T (14·75 (s.e. 1-62) days) and С (14·30 (s.e. 0·70) days) animals, with cows in the T group showing a more variable (F7 9 = 4-30, P < 0·05) interval to the occurrence of the peak. The interval from peak P4 to corpus luteum (CL) regression (when P4 fell below ≥3 μg/l) was not significantly different between the T (4·13 (s.e. 1·30 days) and С (5·60 (s.e. 0·88) days) groups. Furthermore, CIDR insertion did not alter luteal phase length, number of days with P4 concentrations ≥3 μg/l (T = 16·50 (s.e. 0·80) v.C = 15·00 (s.e. 0·47) days) or the interval from oestrus to CL regression (T = 18·88 (s.e. 0·79) v.C = 19·90 (s.e. 0·41) days). We conclude that treatment with CIDR 2 days after oestrus successfully increased the availability of P4 to cows without significantly affecting luteal phase characteristics.
