Recovery of pasture seed ingested by ruminants. 2. Digestion of seed in sacco and in vitro

Seeds of the grasses Brachiaria decumbens (signal grass) and Axonopus afinis (carpet grass), and the legumes Neonotonia wightii cv. Tinaroo, Trifolium semipilosum cv. Safari, Stylosanthes hamata cv. Verano and S. scabra cv. Seca were suspended in nylon bags in the rumen ofcattle (in sacco) and also subjected to in vitro digestion techniques. Legume seeds were evaluated in 3 categories: seed as supplied (mixture of hard and soft), 100% soft and 100% hard. Seeds were either placed in the rumen of cattle (using nylon bags) for 24, 48 or 96 h or subjected to in vitro digestion (in pepsin, and in rumen liquor or cellulase either with or without subsequent digestion in pepsin). Other seed of the same seed lots had been previously fed to penned cattle, sheep and goats and the recovery in faeces had been measured. Soft legume seed were destroyed by the digestion treatments whereas hard seeds were largely resistant to digestion. Average effects of digestion in vitro on viability were similar to average effects of digestion in nylon bags, but there were large differences between different treatments and between seed lots. The percentage of hard seed in the seed sample was the best guide to the resistance of legume seed to digestion.

Two new isosynthetic strains of birdsfoot trefoil (Lotus corniculatus L.), containing low or high concentrations of tannin, were tested using in vitro and in sacco digestibility techniques. The high tannin (HT) strain contained an average of five times more tannin on a dry matter basis, than the low tannin (LT) strain. After 24-h in vitro digestion, by a rumen inoculum, there was less (P < 0.01) dry matter disappearance (45.7 vs. 54.4%) from the HT strain. Also, the development of gas pressure was lower (P < 0.01) and production of volatile fatty acids less (P < 0.05) with the HT strain than with the LT strain. Average dry matter loss from four replicated nylon bags was also lower (P < 0.01) for the HT strain than for the LT strain after 8, 12, and 24 h of incubation in rumen. These data indicate that a LT strain of birdsfoot trefoil might improve the forage digestibility. Key words: Birdsfoot trefoil, tannin, digestibility

This study examined the effects of different species of ruminants and pasture plants on the passage of ingested seed. Penned cattle, sheep and goats were fed a basal diet of 80% lucerne hay and 20% milled wheat. After 14 days on the basal diet a mixture of known numbers of pasture seeds (Brachiaria decumbens, Axonopus afinis, Neonotonia wightii cv. Tinaroo, Trgfolium semipilosum cv. Safari, Stylosanthes hamata cv. Verano and S. scabra cv. Seca) was fed on 1 day. Faeces were collected for 6 days after feeding the seed while animals remained on the basal diet. Seed samples of each species were tested for germination and hardseededness (or dormancy) both prior to feeding and on seed washed from a subsample of the faeces. Other subsamples were kept moist and emerging seedlings were removed over a 104 week period. Cattle digested less seed than did sheep and goats, but the germination characteristics of the recovered seed were similar for the different animals. The mean recovery of viable seeds ingested was 42, 10 and 19% for cattle, sheep and goats, and 39, 39, 23, 21, 12 and 8% for Safari, Tinaroo, Seca, Axonopus, Brachiaria and Verano. Most of the seeds excreted were recovered on the second and third days following feeding. For sheep and goats, the percentage recovery of ingested legume seed as emerging seedlings was similar to recovery calculated from washing out seed followed by germination testing. However, there was a lower recovery, as seedlings, for the seeds ingested by cattle. This was partially due to the larger number of hard seeds remaining in cattle faeces after the 104 week germination period.

Three digestibility experiments were conducted to evaluate the potential of determination of apparent ileal digestibilities (AID) of crude protein (CP) and amino acids (AA) by the mobile nylon bag technique (MNBT) using 21 feedstuffs and three mixed diets. In two conventional digestibility experiments (Exp. 1 and 2), AID were determined using in total 10 barrows (BW 35 kg) fitted with simple T-cannulas at the terminal ileum. For the MNBT studies, four pigs were fitted with a simple T-cannula at the proximal duodenum and a Post-Valve T-Caecum (PVTC) cannula at the terminal ileum. The MNBT studies included the feedstuffs (n = 10) from Exp. 1 and 2 as well as 14 further feedstuffs and mixed diets in which AID coefficients had been determined in previous trials. For each feedstuff 60 nylon bags were used. In vitro digestion of the bags was carried out in pepsin-HCl solution with 450 IU pepsin/l at pH 2.0 and 37&deg;C for 4 h. In the 28-day in vivo experiment, 15 nylon bags per pig and day were inserted through the duodenal cannula and collected through the PVTC cannula after passage through the small intestine. Coefficients of AID were calculated based on the disappearance of CP and AA from the nylon bags during the in vitro and in vivo phase. In comparison with AID determined by the conventional method, AID of CP was on average 2.4% lower, whereas AID of lysine was on average 8.5% higher when determined by the MNBT. There was no significant (P &gt; 0.05) correlation between AID coefficients of CP and AA determined by the conventional method and the MNBT, when all feedstuffs were taken into account. However, in cereals (n = 11), the correlation between AID coefficients determined by both methods was significant (P &lt; 0.05) for CP (r = 0.61) and some AA (r ranging between 0.62 and 0.72). In conclusion, the potential of the MNBT to determine AID of CP and AA is rather limited. Differences in coefficients of AID of CP and AA were attributed to several factors such as diffusion of sample particles or endogenous protein through the nylon bags as well as to the presence of anti-nutritional factors (e.g. in legume seeds and oilseed meals).

Efficient lipid digestion in formula-fed infants is required to ensure the availability of fatty acids for normal organ development. Previous studies suggest that the efficiency of lipid digestion may depend on whether lipids are emulsified with soy lecithin or fractions derived from bovine milk. This study, therefore, aimed to determine whether emulsification with bovine milk-derived emulsifiers or soy lecithin (SL) influenced lipid digestion in vitro and in vivo. Lipid digestibility was determined in vitro in oil-in-water emulsions using four different milk-derived emulsifiers or SL, and the ultrastructural appearance of the emulsions was assessed using electron microscopy. Subsequently, selected emulsions were added to a base diet and fed to preterm neonatal piglets. Initially, preterm pigs equipped with an ileostomy were fed experimental formulas for seven days and stoma output was collected quantitatively. Next, lipid absorption kinetics was studied in preterm pigs given pure emulsions. Finally, complete formulas with different emulsions were fed for four days, and the post-bolus plasma triglyceride level was determined. Milk-derived emulsifiers (containing protein and phospholipids from milk fat globule membranes and extracellular vesicles) showed increased effects on fat digestion compared to SL in an in vitro digestion model. Further, milk-derived emulsifiers significantly increased the digestion of triglyceride in the preterm piglet model compared with SL. Ultra-structural images indicated a more regular and smooth surface of fat droplets emulsified with milk-derived emulsifiers relative to SL. We conclude that, relative to SL, milk-derived emulsifiers lead to a different surface ultrastructure on the lipid droplets, and increase lipid digestion.
