Seed storage behaviour of 101 woody species from the tropical rainforest of southern China: a test of the seed-coat ratio–seed mass (SCR–SM) model for determination of desiccation sensitivity

The Xishuangbanna tropical rainforest in Yunnan Province is the greatest biodiversity hotspot in China. However, the biodiversity of this region is under threat, making seed conservation through seed and/or germplasm banking particularly urgent and crucial. Seed desiccation sensitivity limits the possibility of seed banking of 47% of tropical rainforest species. Thus, knowing if a species has desiccation-sensitive seeds is an important first step in seed banking; however, often resources are limited, making it difficult to determine storage behaviour for all the species in a region. Prediction of seed sensitivity using the SCR–SM model based on seed-coat ratio (SCR) and seed dry mass (SM) might be an alternative for determining desiccation sensitivity of seeds of each species. Here, seed-desiccation sensitivity of 101 woody species from the Xishuangbanna tropical forest were analysed using this model, and physiological determinations were made for a total of 25 species. Seed storage behaviour for 59 species was used for model validation, and storage behaviour of 88% of these species was successfully predicted. Seed storage behaviour of 83% of the 59 species was successfully predicted using the 1000-seed weigth–moisture content (TSW–MC) criteria, which include seeds with 1000-seed weight >500 g and seed moisture content at shedding of 30 –70%. The two predictive methods were subsequently used to predict seed desiccation sensitivity for another 42 species from Xishuangbanna whose storage behaviour was uncertain. Our results indicated that ~50% of the species in Xishuangbanna are likely to have desiccation-sensitive seeds.

Abstract Although the level of seed desiccation sensitivity (LSDS) may have an impact on plant species conservation, information is available for <10% of tropical angiosperms. A study was conducted to assess the LSDS of 28 tropical montane species in Sri Lanka. Seeds were extracted from freshly collected fruits. Initial weight was recorded, and thousand seed weight (TSW) was calculated. Seed moisture content (MC) was determined. LSDS was determined using seed desiccation experiments and predicted using the TSW–MC criterion. Seed storage behaviour was predicted using LSDS and storage data and using a model based on phylogenetic affiliation. The relationship between LSDS and seed dormancy, life form and forest strata was evaluated. Fresh seeds of only 12 species germinated to >80%. Although seeds of the other species had >80% viability, only 0–70% germinated due to dormancy. Seeds of five species had MC <15%, indicating desiccation tolerance (DT). Seeds of 12 species lost viability after desiccation, indicating desiccation sensitivity (DS). Seeds of Ardisia missionis, Psychotria gartneri and Psychotria nigra remained viable after desiccation, showing DT. Seeds of 17 species were DS and those of 11 species DT. The TSW of four species was >500 g. Thus, seeds of other species were predicted to be DT by the TSW–MC criterion. A relationship was identified between LSDS and the forest strata of the species. More canopy species produced DS than DT seeds. Since seeds of most of the studied species were DS, these species may be threatened due to prolonged droughts predicted for the region due to climate change.

Combinational traits of seed size and seed-coat hardness in Citrus garrawayi (F.M.Bailey) (syn. of Microcitrus garrowayi) were investigated as markers for estimation of seed morphological and physiological maturity. Seed size (length) and coat hardness correlated well with changes in seed coat and embryo morphological development, dry-weight accumulation, decreases in moisture content and a significant increase in germinability. Seed moisture content decreased from 82 ± 1% in immature seeds to 40 ± 1% at seed maturation. The outer integument of immature seeds consisted of thin-walled epidermal fibres from which outgrowths of emerging protrusions were observed. In comparison, mature seed coats were characterised by the thickening of the cell walls of the epidermal fibres from which arose numerous protrusions covered by an extensive mucilage layer. Immature seeds, with incomplete embryo and seed-coat histodiffereniation, had a low mean germination percentage of 4 ± 4%. Premature seeds, with a differentiated embryonic axis, were capable of much higher levels of germination (51 ± 10%) before the attainment of mass maturity. Mature seeds, with the most well differentiated embryonic axis and maximum mean dry weight, had the significantly highest level of germination (88 ± 3%).

The present study examined the effect of temperature (15, 20, 25, 30 and 20/30°C) on germination and the storage behaviour of freshly harvested mature seeds of Calamus palustris var. cochinchinensis. Seed desiccation tolerance and the effects of storage temperature (4 and 15°C), perlite water content (120, 180 and 240%) and seed moisture content (27.8, 38.2 and 49.2%) on viability were observed. Seeds had a higher germination at 25°C (88.3%) than at the other tested temperatures. Germination decreased as the seed moisture content decreased during desiccation. The germination of seeds stored at 15°C was higher than that of seeds stored at 4°C. Germination of seeds stored at 15 and 4°C was <65% and with extension of storage time, the germination decreased, indicating that neither temperature can be used for long-term conservation. For short-term storage, the seeds can be stored at 15°C with perlite with 180% water content in plastic bottles or at 15°C with 49.2% moisture content sealed inside aluminum foil bags.

AbstractPrechilling seeds of Sitka spruce (Picea sitchensis[Bong.] Carr.) at 4°C with 30% moisture content for 12–14 weeks (84–98 d) removed conditional dormancy (i.e. they were then able to germinate at 10°C). The non-dormant status was preserved after redrying to 6% moisture content. However, conditional dormancy was gradually reimposed during subsequent air-dry storage at 4°C and 6% seed moisture content in all five seed lots tested. Further investigations with one seed lot showed that reimposition was reversed by a second prechill treatment, but was reimposed again during subsequent air-dry storage. The trend of dormancy reimposition within seed lots over time was quantified by negative exponential relations between ability to germinate at 10°C and duration of air-dry storage. The progress of dormancy reimposition was influenced by seed storage moisture content and was most rapid at 4–10%. At higher moisture contents (15 and 20%) the rate of the reimposition of conditional dormancy was much reduced, while at moisture contents of 25 and 30% further loss in dormancy occurred. Thus it is clear that dormancy reimposition occurred during storage at low water potential rather than solely during desiccation from high to low water potential.

The Ellis-Roberts seed viability equation is used to predict seed survival after storage at specified temperatures and moisture contents. Seed priming, which can break dormancy and accelerate germination, can also reduce seed storage life. Because primed seeds were not used in developing the Ellis-Roberts equation, the reciprocal nature of specific seed moisture content (MC, fresh weight basis) and temperatures that applies to nonprimed lettuce (Lactuca sativa L.) seeds may not apply to primed seeds. To determine how priming affects lettuce seeds in relation to the viability equation, an experiment was conducted using two cultivars, ‘Big Ben’ and ‘Parris Island Cos’. Seeds primed in polyethylene glycol 8000 (–1.45 MPa, 24 h at 15 °C) and nonprimed seeds were first adjusted to 6% and 9% moisture contents and then stored at 48 and 38 °C for up to 30 days, respectively. These storage conditions (6% MC and 48 °C; 9% MC and 38 °C) were predicted by the viability equation to result in equal longevities. Subsequent viability assays at 20 °C revealed that nonprimed seeds in both storage environments exhibited similar losses in viability over time, thus validating the Ellis-Roberts equation and the use of these conditions to apply different but equal aging stress. Primed seeds of both cultivars deteriorated faster than nonprimed seeds as expected. However, primed seeds did exhibit different rates of deterioration between the storage environments. Primed seeds stored at 9% MC and 38 °C deteriorated faster than primed seeds stored at 6% MC and 48 °C. The rate of decline in probit viability percentage was three times greater in primed ‘Big Ben’ seeds stored at 9% MC and 38 °C than for those stored at 6% MC and 48 °C (–1.34 versus –0.26 probits per day, respectively). ‘Parris Island Cos’ seeds stored at 9% MC and 38 °C had twice the rate of deterioration that those stored at 6% MC and 48 °C (–1.19 and –0.49 probits per day, respectively). The results indicate that primed lettuce seeds were more sensitive to the adverse effects of higher seed MC than were nonprimed seeds during storage at elevated temperatures.

The research results of influence of various abiotic factors on seed quality of maize hybrids during long storage are given. The most effective of them are: temperature regime, seed moisture content and oxygen access. The storage methods (dry and cold, and controlled atmosphere storage) that have the greatest impact on the seed quality were established. It was found that the optimal moisture content of seed depends on the storage duration, if for 1–2 years – 10–11 %, 3 years and more – 7–8 %. In order to stabilize the moisture content, it is recommended to store the seeds in airtight containers, limiting the humidity access from the outside. During the long-term seed storage, particularly for the breeding purposes, the temperature range should advisably maintain within 8–10 °C in seed storage place. With that the laboratory germination of seeds increased by 3–5 %, field germination – by 6–8 %, growth rate by number of sprouts– by 5–9 % and by the weight of sprouts – by 2–5 g compared to the 18–20 °C temperature in a typical storage. Methods of pre-sowing improvement of seed quality were established – it is separation and chemical treatment (seed dressing). Separation is the sifting of the seed mass through sieves with round holes in order to divide from the smallest fraction, the content of which was 15–23 % depending on the seed uniformity of maize hybrids. It was found that in order to assess objectively the effectiveness of different chemicals and establish their suitability for presowing seed treatment, it is necessary to determine laboratory germination, especially the growth rate by the number of sprouts and weight of 100 sprouts. Premature chemical treatment of seed should be avoided. Key words: maize hybrids, storage methods, abiotic factors, seed quality, presowing treatment.

Prunus cerasoides D. Don the Himalayan wild cherry is one lesser known multipurpose tree species of Himalaya. The tree prefers to grow on sloping grounds between the altitudes of 1200-2400 m, on all types of soils and rocks. The tree is used as a medicinal plant in Himalayan region. The fruit is edible and the pulp is used to make a cherry brandy. The species has poor germination and seedling establishment in natural habitat. The over exploitation of seeds of the species coupled with relatively hard seed coat has adversely affects the germination of seeds in their natural habitat. The information about the seed maturity and technique of germination enhancement is scanty. The present study was conducted to assess the exact maturity time and optimum temperature for enhancement of germination in seed of P. cerasoides.  The fruit/seeds were collected from six sites covering the altitudinal range of 1350 – 1810 m during the period (2003-2004). The colour change of fruit from dark green to red was a useful indicator of seed maturity. Maximum germination coincided with 50.24 ± 0.19 % fruit and 30.11 ± 0.57 % seed moisture content. Negative correlation existed between germination and seed moisture content (r = 0.294; P< 0.01). Significantly higher germination occurred when seeds were placed above the paper at 25º C.

In the hot and dry conditions in which seeds of the tree legume Peltophorum pterocarpum develop and mature in Vietnam, seed moisture content declined rapidly on the mother plant from 87% at 42 d after flowering (DAF) to 15% at 70 DAF. Dry weight of the pods attained a maximum value at about 42 DAF, but seed mass maturity (i.e. the end of the seed-filling phase) occurred at about 62 DAF, at which time seed moisture content was about 45–48%. The onset of the ability of freshly collected seeds to germinate (in 63-d tests at 28–34°C) occurred at 42 DAF, i.e. about 20 d before mass maturity. Full germination (98%) was attained at 70 DAF, i.e. at about 8 d after mass maturity. Thereafter, germination of fresh seeds declined, due to the imposition of a hard seed coat. Tolerance of desiccation to 10% moisture content was first detected at 56 DAF and was complete within the seed population by 84 DAF, i.e. about 22 d after mass maturity. Hardseededness began to be induced when seeds were dried to about 15% moisture content and below, with a negative logarithmic relation between hardseededness and moisture content below this value.

Deterioration of bean seed quality in storage is a problem for consumers, seed producers, and growers. Research was conducted to evaluate imbibition and germination of dry bean seed cultivars/lines stored under conditions similar to those used by small farmers in the Dominican Republic. Eight dry bean cultivars and lines were grown at three locations in the Dominican Republic. Seed samples were stored in jute bags, plastic containers, and metallic cans under uncontrolled conditions in a small wooden building with slatted shelves. Sub-samples of seeds were taken at 2, 4 and 6 months to determine changes in moisture content, imbibition and germination. Mean cooking time values were determined for seed of four cultivars which had been stored in the three types of above mentioned containers for 6 months. Seed moisture content decreased from 11 to 7% during 2 to 6 months of storage for 'Venezuela 44' and 'ICA-Pijao', respectively. Seed of 'Constanza' had the lowest imbibition value. Imbibition value differed among cultivars for seed stored in different container types. Germination of "Venezuela 44' exceeded 90%, but was only 66% for 'José Beta'. At 6 months, the germination viability of seed stored in jute bags and plastic containers was 78% and 83%, respectively, whereas seed stored in metallic containers was 92% viable. Seed of 'Venezuela 44' had shorter cooking times than seed of the other cultivars. Cooking time of seed differed among container types. Results indicate that growing location, cultivars, storage containers and duration of seed storage can affect seed quality of beans in the Dominican Republic.
