Two storage disorders, "pink white" and "pasty yolk" are known to develop in eggs from hens with cyclopropene fatty acids (e.g. malvalic and sterculic acid) in their diet. The pink white condition is related to increased diffusion processes in the egg during storage. The pasty yolk condition is related to an increase in the proportion of saturated to unsaturated fatty acids in the yolk. The change in texture becomes evident during storage at normal temperature, but can be induced quickly in any affected egg, even when new-laid, by low temperature. The present investigations were carried out to see if the development of these defects could be related to any structural differences detectable in eggs from hens fed a cyclopropene compound.

ABSTRACT There is considerable evidence correlating the production of increased proportions of membrane unsaturated fatty acids (UFAs) with bacterial growth at low temperatures or high pressures. In order to assess the importance of UFAs to microbial growth under these conditions, the effects of conditions altering UFA levels in the psychrotolerant piezophilic deep-sea bacterium Photobacterium profundum SS9 were investigated. The fatty acids produced byP. profundum SS9 grown at various temperatures and pressures were characterized, and differences in fatty acid composition as a function of phase growth, and between inner and outer membranes, were noted. P. profundum SS9 was found to exhibit enhanced proportions of both monounsaturated (MUFAs) and polyunsaturated (PUFAs) fatty acids when grown at a decreased temperature or elevated pressure. Treatment of cells with cerulenin inhibited MUFA but not PUFA synthesis and led to a decreased growth rate and yield at low temperature and high pressure. In addition, oleic acid-auxotrophic mutants were isolated. One of these mutants, strain EA3, was deficient in the production of MUFAs and was both low-temperature sensitive and high-pressure sensitive in the absence of exogenous 18:1 fatty acid. Another mutant, strain EA2, produced little MUFA but elevated levels of the PUFA species eicosapentaenoic acid (EPA; 20:5n-3). This mutant grew slowly but was not low-temperature sensitive or high-pressure sensitive. Finally, reverse genetics was employed to construct a mutant unable to produce EPA. This mutant, strain EA10, was also not low-temperature sensitive or high-pressure sensitive. The significance of these results to the understanding of the role of UFAs in growth under low-temperature or high-pressure conditions is discussed.

Chilling injury is especially prominent in postharvest bananas stored at low temperature below 13 °C. To elucidate better the relationship between cell membrane lipids and chilling injury, an untargeted lipidomics approach using ultra-performance liquid chromatography–mass spectrometry was conducted. Banana fruit were stored at 6 °C for 0 (control) and 4 days and then sampled for lipid analysis. After 4 days of storage, banana peel exhibited a marked chilling injury symptom. Furthermore, 45 lipid compounds, including glycerophospholipids, saccharolipids, and glycerolipids, were identified with significant changes in peel tissues of bananas stored for 4 days compared with the control fruit. In addition, higher ratio of digalactosyldiacylglycerol (DGDG) to monogalactosyldiacylglycerol (MGDG) and higher levels of phosphatidic acid (PA) and saturated fatty acids but lower levels of phosphatidylcholine (PC), phosphatidylethanolamine (PE), and unsaturated fatty acids were observed in banana fruit with chilling injury in contrast to the control fruit. Meanwhile, higher activities of phospholipase D (PLD) and lipoxygenase (LOX) were associated with significantly upregulated gene expressions of MaPLD1 and MaLOX2 and higher malondialdehyde (MDA) content in chilling injury-related bananas. In conclusion, our study indicated that membrane lipid degradation resulted from reduced PC and PE, but accumulated PA, while membrane lipid peroxidation resulted from the elevated saturation of fatty acids, resulting in membrane damage which subsequently accelerated the chilling injury occurrence of banana fruit during storage at low temperature.

Oxidation, synthesis, and desaturation of fatty acids were assessed in chick embryos and embryonic liver. No differences in the oxidation of palmitate-1-14C and oleate-1-14C by intact embryos and embryo-liver homogenates were found. De novo fatty acid synthesis and microsomal elongation of fatty acids were detected in embryo-liver homogenates, but the activities were low as compared with chick liver. The specific activities of the mitochondrial system of fatty acid elongation were similar in embryo and chick liver. Stimulation of the desaturation of stearic acid was achieved by the substitution of glucose for fatty acids in the culture medium and abolished by the addition of cyclopropene fatty acids to the medium. The hypothesis is advanced that in chick embryos, the rate of desaturation of fatty acids synthesized de novo is less than that of postembryonic liver, and that as a consequence, the liver of embryos cannot maintain the proportion of unsaturated to saturated fatty acids found in yolk.

Biodiesel has attracted considerable interest as an alternative biofuel due to its many advantages over conventional petroleum diesel such as inherent lubricity, low toxicity, renewable raw materials, biodegradability, superior flash point, and low carbon footprint. However, high production costs, poor low temperature operability, variability of fuel quality from different feedstocks, and low storage stability negatively impact more widespread adoption. In order to reduce production costs, inexpensive inedible oilseed alternatives are needed for biodiesel production. This study utilized inedible tamarind (Tamarind indica) seed oil as an alternative biodiesel feedstock, which contained linoleic (31.8%), oleic (17.1%), and lauric (12.0%) acids as the primary fatty acids. A simple and cost-effective high vacuum fractional distillation (HVFD) methodology was used to separate the oil into three fractions (F1, F2, and F3). Subsequent transesterification utilizing basic, acidic, and enzymatic catalysis produced biodiesel of consistent quality and overcame the problem of low temperature biodiesel performance. The most desirable biodiesel with regard to low temperature operability was produced from fractions F2 and F3, which were enriched in unsaturated fatty acids relative to tamarind seed oil. Other properties such as density and cetane number were within the limits specified in the American and European biodiesel standards.
