Evaluation of the efficacy of sequential or continuous administration of probiotics and phytogenics in broiler diets

Three dietary combinations of probiotics and phytogenics administered sequentially or continuously were assessed for their effects on broiler performance, nutrient digestibility, caecal microbiota composition, volatile fatty acid (VFA) pattern and plasma total antioxidant capacity (TAC). One-day-old Cobb male broilers (n = 525) were allocated to five experimental treatments for 42 days. Each treatment had three replicates of 35 broilers each. Depending on the time-frame of sequential or continuous addition in the basal diet (BD) of the probiotics (108 CFU/kg BD) and phytogenics (125 mg/kg BD), experimental treatments were: control (no additions, Days 1–42); PE1, probiotic Days 1–14 and phytogenic Days 15–42; PE2, probiotic Days 1–28 and phytogenic Days 29–42; PE3, probiotic and phytogenic continuously (Days 1–42); and A (2.5 mg avilamycin/kg diet, Days 1–42). There was no loss of probiotic viability upon mixing with the phytogenic for up to 7 days. Overall bodyweight gain was higher (P = 0.025) in the control and PE1 than PE2 and PE3, whereas Treatment A was intermediate and not different from the rest. Overall feed intake in the control was the highest (P = 0.003). Treatments PE1, PE2 and PE3 had better (P = 0.004) overall feed conversion ratio (FCR) than the control. The FCR in PE1 and PE3 did not differ from A, which had the best FCR. Mortality did not differ among treatments. Nutrient digestibility and caecal microbiota composition did not differ among treatments. However, differences (P ≤ 0.05) in caecal VFA pattern were noted, with the control having the lowest acetate molar ratio as well as the highest sum of valeric, hexanoic and heptanoic acids. Plasma TAC in the control was lower than in PE3, PE1 and PE2 at Day 14 (P < 0.001) and PE1 (P = 0.003) at Day 42. The concept of sequential or throughout probiotic and phytogenic administration showed promising results regarding feed efficiency and plasma TAC.

The study was conducted to evaluate the feed intake, digestibility, bodyweight change and carcass characteristics of Farta lambs fed Brachiaria (now: Urochloa) hybrid Mulato II and natural pasture hays in various proportions as a basal diet supplemented with a local concentrate mixture (CM). Twenty-five yearling male Farta lambs with a mean initial body weight of 19.6±0.29 kg (mean±s.e.) were used in feeding (90 days) and digestibility (7 days) trials. The lambs were randomly allocated to the following 5 dietary treatments on the basis of stratified body weight: 100% natural pasture hay (NPH) (T1); 75% NPH+25% Urochloa hybrid Mulato II hay (MH) (T2); 50% NPH+50% MH (T3); 25% NPH+75% MH (T4); and 100% MH (T5). A local concentrate mixture [300 g dry matter (DM)/hd/d] was fed to all animals. Crude protein (CP) concentration of the basal diet increased as proportion of MH in the ration increased (P<0.05). Intake of DM and nutrients, and nutrient digestibility coefficients increased significantly (P<0.05) as proportion of MH in the roughage component of the ration increased. Final body weight, average daily bodyweight gain, feed conversion efficiency and most carcass parameters measured were significantly (P<0.05) higher as proportion of MH increased from 0 to 100% in the basal diet. Based on the biological performance of the experimental lambs, performance of sheep in the region could be enhanced significantly by incorporating MH with native pasture hay and concentrate supplement in feeding rations. Economic assessments would reveal the optimal combinations of native pasture and MH for feeding to achieve particular outcomes. Other improved grass and legume species may fill the same role and should be investigated in differing environments.

Two experiments were conducted to investigate the effect of arginine (Arg); the combination of Arg and glutamine (Gln); as well as an amino acid-based solution (MIX) containing Arg, Gln, threonine (Thr), and grape extract, on performance, intestinal permeability, and expression of selected mechanistic genes. Using 240 male Ross 308 off-sex broiler chickens, four experimental treatments were replicated six times with 10 birds per replicate. The experimental treatments included 5 g/kg Arg, 2.5 g/kg Arg and 2.5 g/kg Gln, and 1 g/kg MIX added to a basal diet as control. In the second study, the four dietary treatments were then given to 24 birds with or without a synthetic glucocorticoid, dexamethasone (DEX), as a gut dysfunction model. Feed conversion ratio was improved by all the supplemented treatments from day 7 to 35 of age (p < 0.001). DEX injections increased (p < 0.001) the intestinal permeability in all treatments, which tended to be reversed by Arg or MIX. Additional Arg, Arg-Gln, and MIX suppressed (p < 0.05) the overexpression of IL-1β generated by DEX. Feeding birds with MIX treatment increased (p < 0.05) expression of SGLT-1 and glutathione synthetase. In conclusion, tested amino acid supplements were effective in improving feed efficiency and restraining intestinal inflammation caused by DEX through IL-1β pathway.

The present study was conducted to evaluate the effect of different levels of methionine supplementation on feed intake, nutrient digestibility and growth performance of growing rabbit. Sixteen weaned crossbred New Zealand White (NZW) growing rabbits (30-35 d) were distributed into four treatment groups having four replications in each group using a Completely Randomized Design (CRD). Basal diet composed of green grass (dhal grass) and concentrate mixture which was offered ad libitum basis for 56 days period. Four levels of methionine such as 0.0% (control), 0.15%, 0.25%, and 0.35% were supplemented randomly to rabbits. Results showed that supplementation of methionine did not affect green grass intake. Cumulative as well as daily concentrate and DM intake were significantly (p<0.05) higher for all methionine groups than control group. Final body weight gain as well as daily, weekly and cumulative body weight gains were improved significantly with increasing level of methionine. It was found that methionine had significant (p<0.01) effect on digestibility of DM, CP, NFE and EE but CF digestibility did not differ significantly. Digestibility was improved with increasing the level of methionine. Feed conversion ratio also decreased significantly with methionine supplementation, and 0.25% methionine group showed the best performance among the four treatments. The effect of the dietary inclusion level of a three-species probiotic on broiler performance, nutrient digestibility, caecal microbiota composition and volatile fatty acid (VFA) pattern was evaluated. Day-old Cobb broilers (n = 448) were allocated in four treatments for 6 weeks. Each treatment had four replicates (two per gender) of 28 broilers each. Depending on the type of addition per kg basal diet, treatments were C (no other addition), PL (108 colony forming units of probiotic), PH (109 colony forming units of probiotic) and A (2.5 mg avilamycin). Overall bodyweight gain was better (P = 0.002) in PL and PH than in the control (2082 g) by 8.7% and 7.5%, respectively, while treatment PL did not differ from A (2341 g), which showed the highest bodyweight gain. The ileal and total-tract apparent digestibility of DM and the apparent metabolisable energy content corrected for N improved linearly (P ≤ 0.05) with the probiotic level. Fluorescent in situ hybridisation analysis showed caecal Bifidobacterium levels to increase linearly (P = 0.006) with the probiotic level. Probiotic administration resulted in altered caecal VFA patterns compared with the control. Gender effects (P ≤ 0.05) were noted for caecal levels of C. histolyticum group, Bacteroides fragilis group and Streptococcus spp., while interactions (P ≤ 0.05) of treatment with gender were seen for Bifidobacterium and all VFA components, except for acetate. In conclusion, beneficial effects on bodyweight gain, DM digestibility, apparent metabolisable energy content corrected for N, caecal Bifidobacterium levels and VFA patterns were noted with both probiotic inclusion levels.

Two experiments were carried out to investigate the effect of “on top” addition of different enzyme complexes, the enzyme α-galactosidase and three sources of the enzyme phytase available on the market, in broiler diets. In the first experiment, 1260 one-day-old Cobb 500® chicks were distributed into seven treatments in a completely randomized design (CRD) with six replicates and 30 birds/replicate. Treatments consisted of combinations of different enzyme complexes, namely, complex A (phytase, protease, xylanase, ß-glucanase, cellulase, amylase, pectinase), complex B (protease and cellulase) and complex C (xylanase, amylase and protease); isolated α-galactosidase (GAL); and three sources of phytase (P1, P2 and P3) in the diet. The treatments were formulated as follows: T1 - basal diet (BD); T2 - BD + enzyme complex A + enzyme complex B (BDAB); T3 - BDAB + GAL; T4 - BD + complex A + GAL; T5 - BD + complex C + P1 + GAL (BDCG); T6 - BDCG + P2; and T7 - BDCG + P3. The following variables were measured in the experimental period of 42 days: feed intake (FI), weight gain (WG), average final weight (AFW), feed conversion (FC), and carcass yield. Significant differences occurred for AFW, WG and FC in the pre-starter phase. In the second experiment, 112 Cobb 500® chicks aged 25 days were distributed into seven treatments in a CRD with four replicates and four birds/replicate. Treatments were the same as in the first experiment. Nutrient digestibility was evaluated in an experimental period of seven days. Differences were found in the metabolism coefficient of ether extract (MCEE). Dietary inclusion of enzyme complexes improves the AFW and WG of chickens from 1 to 7 days of age and MCEE in the grower phase.

Incremental levels of the phytase, OptiPhos® Plus, were fed to 150 weaned piglets (five treatments of 30 pigs) to determine the effect on piglet performance, nutrient digestibility and bone mineralisation. Piglets were fed a basal diet from four to six weeks and a pre-starter diet from six to ten weeks of age. The basal diets contained reduced digestible phosphorus (P) and calcium (Ca) (1.5 g/kg digestible P, and Ca 5.5 g/kg and 5.6 g/kg for weaner and pre-starter, respectively) to provide a negative control. Phytase was added at 0, 125, 250, 500 and 1000 phytase units (FTU)/kg feed. Faecal samples were collected at nine weeks of age and analysed for P, Ca and crude protein (CP) content to calculated digestibility. Metacarpal bone IV mineralisation was assessed by ash content and the weight and length of the bone. Incremental increases of phytase improved final body weight (BW) (P<0.001), average daily gain (ADG) (P<0.001) and feed conversion ratio (FCR) (P<0.01). The statistical model predicted a linear increase for daily feed intake (DFI), ADG and BW with increased phytase increments. BW at the end of the trial increased by 824 g per increase of 250 FTU/kg (P<0.001). ADG and DFI increased by 19 g/day (P<0.001) and 18 g/day (P<0.05) respectively, per increase of 250 FTU/kg. As the effect on ADG and final BW was higher at lowest doses, a quadratic effect for these parameters was observed, within the measured range of phytase levels (P=0.029 and 0.028, respectively). A reduction of 0.03 in FCR was observed for each 250 FTU/kg increase (P<0.01). For each increase in 250 FTU/kg, bone dry weight increased by 131 g (P<0.05) and bone ash concentration by 1.9% of fat free dry matter (P<0.001). The results showed that the addition of phytase to piglet diets low in P led to better utilisation of phytate-P and consequently better piglet performance.

The study aimed to evaluate the effect of inulin and mannan-oligosaccharides (MOS) vs zinc-bacitracin (ZnB) on rabbit growth performance, nutrient digestibility, and blood profiles. In total, 108 rabbits 32 days of age were divided into four homogeneous groups fed the same basal diet and submitted to the following dietary treatments: control group, without supplementation; inulin group, supplemented with inulin (0.25 g/rabbit.day); MOS group, supplemented with MOS (0.083 g/rabbit.day); and ZnB group, supplemented with ZnB (0.083 g/rabbit.day). The supplements were administered 2 days per week from 32 to 81 days of age. At 32, 56 and 81 days, the rabbits were weighed and feed intake was measured. Mortality was recorded daily. Nutrient digestibility was measured at 65 days by an ingesta–excreta balance. Samples of blood were collected at 53, 67 and 81 days of age. The control group showed the highest (P < 0.01) mortality rate throughout the trial (17.0%), followed by ZnB (8.18%), inulin (3.7%) and then the MOS group, which had the lowest mortality (1.6%; P < 0.01). Addition of MOS led to a more favourable (P < 0.05) feed conversion ratio than addition of ZnB at 32–56 days of age and inulin for the entire trial period. The MOS group showed increased (P < 0.01) digestibility of dry matter, organic matter, ash and crude fibre relative to the control group. All supplements increased (P < 0.01) protein digestibility relative to the control. The level of alanine aminotransferase was reduced due to the use of feed supplements, and MOS supplementation gave the lowest (P < 0.01) value. Administration of prebiotics to growing rabbits as an alternative to ZnB can lead some advantages compared with both antibiotic and unsupplemented diets; MOS was more effective than inulin.

This study was conducted to investigate the effects of dietary supplementation with compound probiotics and berberine (CPB) on growth performance, nutrient digestibility and fecal microflora in weaned piglets. A total of 200 piglets 35 days old were randomly allocated to 5 groups, 4 replications in each group, and 10 piglets in each replication. Group A was the basal diet; group B was supplemented with antibiotics and zinc oxide; groups C, D and E were supplemented with 0.06%, 0.12% and 0.18% CPB, respectively. The experimental period was 42 d. The results indicated that there were no significant differences in average daily feed intake (ADFI), average daily gain (ADG) and feed conversion rate (FCR) among five groups (p > 0.05). However, mortality, diarrhea and rejection rates in the control group were higher than that in other groups. CPB could increase protein digestibility and serum IgG content (p < 0.05), while it could decrease serum urea nitrogen content and alkaline phosphatase activity (p < 0.05). Analysis of fecal microbiota showed that the relative abundances of Bacteroides and Firmicutes were increased, while the relative abundances of opportunistic pathogens such as Spirochaetae and Protebactreria were dramatically decreased in piglets fed with CPB or antibiotics, compared with the control group. Furthermore, CPB intervention increased the relative abundances of Prevotella_9, Megasphaera and Prevotella_2, while decreased the relative abundance of Prevotellaceae_NK3B31_group. Correlation analysis revealed that there was good correlation between serum indexes and fecal microbiota. It was suggested that CPB might be a promising antibiotic alternative for improving piglet health and immunity, decreasing mortality by positively altering gut microbiota.

Objective: This study was conducted to evaluate the effects of diets containing different wet rice distillers’ by-product (RDP) levels on growth performance, nutrient digestibility, blood profiles and gut microbiome of weaned piglets.Methods: A total of 48 weaned castrated male crossbred pigs, initial body weight 7.54±0.97 kg, and age about 4 wks, were used in this experiment. The piglets were randomly allocated into three iso-nitrogenous diet groups that were fed either a control diet, a diet with 15% RDP, or a diet with 30% RDP for a total of 35 days. Chromium oxide was used for apparent digestibility measurements. On d 14 and d 35, half of the piglets were randomly selected for hemato-biochemical and gut microbiota evaluations.Results: Increasing inclusion levels of RDP tended to linearly increase (p≤0.07) average daily gain on d 14 and d 35, and decreased (p = 0.08) feed conversion ratio on d 35. Empty stomach weight increased (p = 0.03) on d 35 while digestibility of diet components decreased. Serum globulin concentration decreased on d 14 (p = 0.003) and red blood cell count tended to decrease (p = 0.06) on d 35, parallel to increase RDP levels. Gene amplicon profiling of 16S rRNA revealed that the colonic microbiota composition of weaned pigs changed by inclusion of RDP over the period. On d 14, decreased proportions of Lachnospiraceae_ge, Ruminococcaceae_ge, Ruminococcaceae_UCG-005, and Bacteroidales_ge, and increased proportions of Prevotellaceae_ge, Prevotella_2, and Prevotella_9 were found with inclusion of RDP, whereas opposite effect was found on d 35. Additionally, the proportion of Lachnospiraceae_ge, Ruminococcaceae_ge, Ruminococcaceae_UCG-005, and Bacteroidales_ge in RDP diets decreased over periods in control diet but increased largely in diet with 30% RDP.Conclusion: These results indicate that RDP in a favorable way modulate gastrointestinal microbiota composition and improve piglet performance despite a negative impact on digestibility of lipids and gross energy.

The objective of the present study was to determine the effect of dietary supplementation of neem oil (Azadirachtaindica) on the growth performance and nutrient digestibility of weaned rabbits. A total of 50 weaned male cross bred rabbits between 5-6 weeks with an average weight of 565.4g-566.8gwere divided into five dietary groups of ten (10) weaned rabbits each in a completely randomized design. The dietary treatments include a control, T1 (basal) diet with no neem oil (NOL), T2, T3, T4 and T5 were fed basal diet supplemented with NOL at 0.1%, 0.2%, 0.3% and 0.4% respectively. Feed and water were offered ad libitum throughout the experiment which lasted for 12 weeks. The data obtained was used to evaluate the growth performance: weight gain (WG), feed intake (FI), feed conversion ratio (FCR), mortality and nutrient digestibility (dry matter, crude protein, crude fibre and ether extract). WG, ADFI, TFI were significantly (P˂0.05) different among the treatments. T5 had the highest weight gain (755.90 g) followed by T4(734.0g), T3(705.90g), T2(705.0g) and T1(621.80g) respectively. Highest mortality was recorded in T1 (2%) followed by T2 (1%), none was recorded in T3, T4 and T5. Neem oil significantly influenced (P˂0.05) all the parameters measured. It could be concluded thatneem oil contains some essential nutrients and bioactive chemicals and could be supplemented in the diet of rabbit at 0.4 % without any deleterious effect on the general performance of the animal.
