Producing milk with uniform high selenium concentrations on commercial dairy farms

Six herds on five commercial dairy farms were involved in the production of high selenium (Se) milk. The farms had a range of herd sizes, herd structures, feeding systems and milk production per cow. On all farms, pelleted concentrate supplements containing Se yeast were fed twice daily in the dairy for 16 days. The objectives were to: (1) produce milk with Se concentrations exceeding 225 μg/kg on the five farms for pilot-scale production of a high protein milk powder; (2) validate a predictive relationship between Se intake and milk Se concentration developed in research; and (3) examine the time taken from the introduction of Se yeast to steady-state concentrations of Se in milk under a range of commercial farming conditions. We hypothesised that the relationship between Se intake and its concentration in milk found in research would apply on commercial farms. Daily Se intake, which was primarily from Se yeast in the pelleted concentrates, varied from 35 to 51 mg Se/cow. Grazed pasture and conserved forage contributed less than 1 mg Se/cow on all farms. The time taken from the introduction of pellets containing Se yeast to steady-state milk Se concentrations was 4–7 days. The steady-state Se concentrations in milk varied from 166 to 247 µg/kg, but these concentrations were only 55–72% of predicted values. All the milk produced from the five farms on the last 2 days of feeding of Se-enriched pellets was used to produce a milk protein concentrate with a Se concentration of 5.4 mg/kg. Factors that might have affected Se incorporation into milk and the implications of these results for commercial production of high Se milk or milk products are discussed.

For pilot-scale production of chito-oligosaccharides, it must be cost-effective to prepare designable recombinant chitosanase. Herein, an efficient method for preparing recombinant Bacillus chitosanase from Escherichia coli by elimination of undesirable substances as a precipitate is proposed. After an optimized culture with IPTG (Isopropyl β-d-1-thiogalactopyranoside) induction, the harvested cells were resuspended, disrupted by sonication, divided by selective precipitation, and stored using the same solution conditions. Several factors involved in these procedures, including ion types, ionic concentration, pH, and bacterial cell density, were examined. The optimal conditions were inferred to be pH = 4.5, 300 mM sodium dihydrogen phosphate, and cell density below 1011 cells/mL. Finally, recombinant chitosanase was purified to >70% homogeneity with an activity recovery and enzyme yield of 90% and 106 mg/L, respectively. When 10 L of 5% chitosan was hydrolyzed with 2500 units of chitosanase at ambient temperature for 72 h, hydrolyzed products having molar masses of 833 ± 222 g/mol with multiple degrees of polymerization (chito-dimer to tetramer) were obtained. This work provided an economical and eco-friendly preparation of recombinant chitosanase to scale up the hydrolysis of chitosan towards tailored oligosaccharides in the near future.

By only applying a after the event exam in the quality control of the batch production is not enough to meet the needs of modern large-scale production. To a certain extent, modern quality control is a dynamic process of the steady-state judge and adjustment. A simple and reliable steady-state judge rule and method is the premise to guarantee the normal operation. This paper provides a quantitative method to evaluate production process steady-state by analyzing influence factors based on mathematical statistics. The method is both suitable for simple production process and complex production process with sub-processes.

A pilot-scale fixed-biofilm reactor (FBR) was established to treat textile wastewater to evaluate the feasibility of replacing conventional treatment processes that involve activated sludge and coagulation units. A kinetic model was developed to describe the biodegradation of textile wastewater by FBR. Batch kinetic tests were performed to evaluate the biokinetic parameters that are used in the model. FBR column test was fed with a mean COD of 692 mg/L of textile wastewater from flow equalization unit. The influent flow rate was maintained at 48.4 L/h for FBR column test. Experimental data and model-predicted data for substrate effluent concentration (as COD), concentration of suspended biomass in effluent and the amount of carbon dioxide (CO2) produced in the effluent agree closely with each other. Microscopic observations demonstrated that the biofilm exhibited a uniform distribution on the surface of polyurethane foam sponge. Under a steady-state condition, the effluent COD from FBR was about 14.7 mg COD/L (0.0213 Sb0), meeting the discharge standard (COD < 100 mg/L) that has been set by the government of Taiwan for textile wastewater effluent. The amount of biofilm and suspended biomass reached a maximal value in the steady state when the substrate flux reached a constant value and remained maximal. Approximately 33% of the substrate concentration (as COD) was converted to CO2 during biodegradation in the FBR test. The experimental and modeling schemes proposed in this study could be employed to design a full-scale FBR to treat textile wastewater.

ABSTRACT Fatty acid ethyl esters (FAEEs) were produced in this study by the use of an engineered Escherichia coli p(Microdiesel) strain. Four fed-batch pilot scale cultivations were carried out by first using glycerol as sole carbon source for biomass production before glucose and oleic acid were added as carbon sources. Cultivations yielded a cell density of up to 61 ± 3.1 g of cell dry mass (CDM) per liter and a maximal FAEE content of 25.4% ± 1.1% (wt/wt) of CDM.

The Aim of this study was to develop an optimized method for manufacturing process of traditional Algerian Jben cheese, using response surface methodology (RSM). In order to develop the objective method of making this traditional cheese, several factors have been studied and a Plackett-Burman statistical design was applied. The effects of the four screened factors (enrichment with milk powder, 10 - 20 g/l; pH of milk, 5.75 - 6.75, enzymatic extract dose, 0.5 - 1.5 ml and coagulation temperature 40 - 60 °C) on the response were investigated, using a Box-Behnken statistical design. Multiple regression analysis was used so that experimental data fits to a second-order polynomial equation. This multiple analysis showed that the model explains about 90.73% of the variation. Based on statistical results, it can be noticed that enrichment with milk powder and pH of milk (Ë‚0.0001***) were highly significant factor influincing cheese yield. The optimal production parame-ters that maximized cheese product (20 g/l enrichment with milk powder, 5.75 pH of milk, 1.29 ml enzymatic extract dose and 60°C coagulation temperature) and the maximal predicted cheese yield (52.68 % ) were found out through response surface methodology. Under these conditions, a verification experiment was carried out and cheese yield was found to be 49.46 %. The overall percentage of agreement for the experimental results (more than 93 % validity) with the predicted values indicates the validation of the statistical model and the success of the optimization process.
