Background: The prevalence of inflammatory bowel disease is rapidly increasing around the world. Quercetin is a flavonoid commonly found in vegetables and fruits and has been reported to exert numerous pharmacological activities such as enhancing antioxidant capacity or suppressing inflammation. Objective: We aimed to explore whether quercetin was effective for IBD and the underlying mechanism of quercetin for the ameliorative effects on the DSS-induced colitis in mice. Methods: Thirty-six mice were randomly assigned to three treatments, including the control group (Ctr), DSS-induced colitis group (DSS) and DSS-induced colitis supplemented with 500 ppm quercetin (DQ500). Colitis was induced by DSS intake, and body weight was recorded every day. After six days administration of DSS, intestinal permeability was measured, and the liver was taken for antioxidant enzyme tests. Colonic tissue was taken for the histopathlogical score and RNA-sequencing analysis. Results: In this experiment, dietary quercetin for 500ppm alleviated the DSS-induced colitis, possibly by strengthening intestinal integrity, liver antioxidant capacity. Based on the results of the transcriptome of colon tissue, several key genes were modulated by quercetin. ERK1/2-FKBP pathway and RXR-STAT3 pathway were involved in the development of IBD, furthermore, in the down-regulation of S100a8/9, FBN2 contributed to lowering the risk of colongenesis. Conclusion: We demonstrated that dietary quercetin alleviated the DSS-induced colitis in mice. This is most likely due to its beneficial effects on intestinal integrity and modulation of several key pathways. Based on our research, quercetin was a promising candidate for IBD and its pharmaceutical effects on both IBD and colongenesis need further research.

Abstract We previously found that i.m. injection of GRA, similar to in-feed antibiotics, improves measures of intestinal integrity, nutrient digestibility, and overall growth performance in nursery pigs. In addition, we have demonstrated that the mitigating effects of GRA on the stress of weaning is predominantly done by subsiding the inflammatory response that often accompanies early weaning. The objective of the current study was to explore the most practical methods for delivering GRA to newly weaned pigs. One-hundred and sixty-seven PIC pigs were weaned at 25 ±1.0 days of age (BW 7 ±1.2 kg), and assigned to fourteen treatments (3-4 pigs /pen; 3 pens/treatment). Seven treatments were tested within two sexes (gilts vs. barrows). The treatments were: 1) i.m. injection of GRA (two injections, 0.2 mg/kg BW at -1 and 3 d post-weaning), 2) two levels of in-water GRA (0.8 and 1.6 ppm), 3) in-feed GRA (2.5 and 5.0 ppm), 4) in-feed antibiotic (ANT, 100 ppm tylosin), and 5) control. Dexamethasone was used as GRA. Pigs had free access to in-feed or in-water GRA or ANT during the 1st-week post-weaning. Parameters of growth performance were measured weekly until the end of the nursery phase. A completely randomized design and repeated measurement ANOVA (PROC MIXED) in SAS were used for statistical analyses. In-water, in-feed, and i.m. GRA treatments resulted in growth performance outcomes comparable to that of in-feed ANT. Relative to CON pigs, both ANT and GRA improved ADG and G: F during the nursery phase (P < 0.05). Among the delivery methods tested, 2.5 ppm of in-feed GRA resulted in superior ADG (GRA vs. CON; 0.29 vs. 0.20 kg/d) and G: F (0.63 vs. 0.40 ± 0.05), compared to the control group (P < 0.02). Collectively, these results suggested that in-feed treatment is the best method for delivering GRA to newly weaned pigs.

Abstract Beneficial effects of SCFA in modulating gut health stimulated interest on dietary strategies to increase intestinal microbial activity and digesta SCFA. Amylose has lower apparent ileal digestibility (AID) than amylopectin. In the large intestine, undigested starch is fermented by microbes producing SCFA. The objective was to determine effects of increasing dietary amylose on starch flow and metabolite profile along the intestinal tract in weaned pigs. Weaned pigs (n=32; initial BW, 8.4 kg) were randomly allocated to 4 diets containing 67% starch with 0, 20, 35, or 70% amylose in a randomized complete block design. On day 21, pigs were euthanized to collect digesta and feces for evaluating starch digestion and metabolite profiles. Apparent hindgut fermentation (AHF) was calculated as apparent total tract digestibility minus AID. Feed intake was 12% lower (P < 0.05) and growth rate was 18% lower (P < 0.05) for pigs fed 70% amylose than pigs fed 0, 20, or 35% amylose. Feed efficiency was greatest (P < 0.05) for pigs fed with 35% amylose. The AID of starch was 44% lower (P < 0.05) in pigs fed 70% amylose. Starch was completely digested by the proximal colon in pigs fed 0, 20, or 35% amylose, but AHF of starch was 14% greater (P < 0.05) in pigs fed 70% amylose. Increasing dietary amylose did not alter digesta SCFA in the small intestine, but increased (P < 0.05) digesta SCFA in the cecum, specifically acetate and total SCFA, and increased (P < 0.05) propionate and valerate in all sections of the colon. In conclusion, increasing dietary amylose in weaned pigs stimulated hindgut fermentation of starch with a corresponding increase in digesta total SCFA in the cecum and colon. Optimizing dietary amylose may exert its effect as dietary prebiotic while promoting an optimal growth rate in young pigs.

Puerarin is a well-known traditional Chinese medicine which has been used for the treatment of cardiovascular diseases. Recently, a new advantageous crystal form of puerarin, puerarin-V, has been developed. However, the cardioprotective effects of puerarin-V on myocardial infarction (MI) heart failure are still unclear. In this research, we aim to evaluate the cardioprotective effects of puerarin-V on the isoproterenol (ISO)-induced MI mice and elucidate the underlying mechanisms. To induce MI in C57BL/6 mice, ISO was administered at 40 mg/kg subcutaneously every 12 h for three times in total. The mice were randomly divided into nine groups: (1) control; (2) ISO; (3) ISO + puerarin injection; (4–9) ISO + puerarin-V at different doses and timings. After treatment, cardiac function was evaluated by electrocardiogram (ECG), biochemical and histochemical analysis. In vitro inflammatory responses and apoptosis were evaluated in human coronary artery endothelial cells (HCAECs) challenged by lipopolysaccharide (LPS). LPS-induced PPAR-Υ/NF-κB and subsequently activation of cytokines were assessed by the western blot and real-time polymerase chain reaction (PCR). Administration of puerarin-V significantly inhibits the typical ST segment depression compared with that in MI mice. Further, puerarin-V treatment significantly improves ventricular wall infarction, decreases the incidence of mortality, and inhibits the levels of myocardial injury markers. Moreover, puerarin-V treatment reduces the inflammatory milieu in the heart of MI mice, thereby blocking the upregulation of proinflammatory cytokines (TNF-α, IL-1β and IL-6). The beneficial effects of puerarin-V might be associated with the normalization in gene expression of PPAR-Υ and PPAR-Υ/NF-κB /ΙκB-α/ΙΚΚα/β phosphorylation. In the in vitro experiment, treatment with puerarin-V (0.3, 1 and 3 μM) significantly reduces cell death and suppresses the inflammation cytokines expression. Likewise, puerarin-V exhibits similar mechanisms. The cardioprotective effects of puerarin-V treatment on MI mice in the pre + post-ISO group seem to be more prominent compared to those in the post-ISO group. Puerarin-V exerts cardioprotective effects against ISO-induced MI in mice, which may be related to the activation of PPAR-γ and the inhibition of NF-κB signaling in vivo and in vitro. Taken together, our research provides a new therapeutic option for the treatment of MI in clinic.

Alginate is a natural polysaccharide present in various marine brown seaweeds. Alginate oligosaccharide (AOS) is a degradation product of alginate, which has received increasing attention due to its low molecular weight and promising biological activity. The wide-ranging biological activity of AOS is closely related to the diversity of their structures. AOS with a specific structure and distinct applications can be obtained by different methods of alginate degradation. This review focuses on recent advances in the biological activity of alginate and its derivatives, including their anti-tumor, anti-oxidative, immunoregulatory, anti-inflammatory, neuroprotective, antibacterial, hypolipidemic, antihypertensive, and hypoglycemic properties, as well as the ability to suppress obesity and promote cell proliferation and regulate plant growth. We hope that this review will provide theoretical basis and inspiration for the high-value research developments and utilization of AOS-related products.

In chronic kidney disease (CKD), oxidative stress (OS) plays a central role in the development of cardiovascular diseases. This pilot program aimed to determine whether an intradialytic aerobic cycling training protocol, by increasing physical fitness, could reduce OS and improve other CKD-related disorders such as altered body composition and lipid profile. Eighteen hemodialysis patients were randomly assigned to either an intradialytic training (cycling: 30 min, 55%–60% peak power, 3 days/week) group (EX; n = 8) or a control group (CON; n = 10) for 3 months. Body composition (from dual-energy X-ray absorptiometry), physical fitness (peak oxygen uptake and the 6-minute walk test (6MWT)), lipid profile (triglycerides (TG), total cholesterol, high-density lipoprotein, and low-density lipoprotein (LDL)), and pro/antioxidant status (15-F2α-isoprostanes (F2-IsoP) and oxidized LDL in plasma; superoxide dismutase, glutathione peroxidase, and reduced/oxidized glutathione in erythrocytes) were determined at baseline and 3 months later. The intradialytic training protocol did not modify body composition but had significant effects on physical fitness, lipid profile, and pro/antioxidant status. Indeed, at 3 months: (i) performance on the 6MWT was increased in EX (+23.4%, p < 0.001) but did not change in CON, (ii) plasma TG were reduced in EX (–23%, p < 0.03) but were not modified in CON, and (iii) plasma F2-IsoP concentrations were lower in EX than in CON (–35.7%, p = 0.02). In conclusion, our results show that 30 min of intradialytic training, 3 times per week for 3 months, are enough to exert beneficial effects on the most sensitive and reliable marker of lipid peroxidation (IsoP) while improving CKD-associated disorders (lipid profile and physical fitness). Intradialytic aerobic cycling training represents a useful and easy strategy to reduce CKD-associated disorders. These results need to be confirmed with a larger randomized study.
