By selective dissociation of histones with the ionic detergent sodium deoxycholate, we have demonstrated that these basic chromosomal polypeptides, which are effective inhibitors of transcription, are more tenaciously bound to DNA in mitotic than in S-phase chromatin. Evidence is presented which suggests that cell-cycle-stage-specific non-histone chromosomal proteins can account for such variations in the association of histones with DNA. When chromatin is reconstituted with DNA and histones are pooled from S-phase and mitotic cells and either S-phase or mitotic non-histone chromosomal proteins, a preferential extraction of histones with sodium deoxycholate from chromatin reconstituted with S-phase rather than mitotic non-histone chromosomal proteins is observed. In contrast, the extractability of histones with sodium deoxycholate from nucleohistone complexes reconstituted with DNA pooled from S-phase and mitotic cells and either S-phase or mitotic histones is identical. Since non-histone chromosomal proteins rather than histones are responsible for the differences in chromatin template activity during S-phase and mitosis, we propose that non-histone chromosomal proteins may modify gene expression during the cell cycle by mediating the binding of histones to DNA.

Diploid nuclei of different cell types of the granulocytopoietic and erythropoietic series showed different Feulgen values. The highest values were found in the most immature nuclei, and the lowest values in the most mature cells. Extraction of acid-soluble nuclear proteins brought the different values to the same level. Examples of similar findings in other cell types, as described in the literature, are discussed. The Feulgen reaction and its colored product depend on the functional state of the chromatin in such a way that nuclei with high template activity of deoxyribonucleic acid show high Feulgen values whereas mature, dense nuclei with low template activity have lower Feulgen values, although both kinds of nuclei contain identical amounts of deoxyribonucleic acid.

Deoxyribonucleic acid (DNA) synthesis as reflected in the specific activity of DNA nucleotides 12 hr after intraperitoneal injection of P32 has been compared after partial hepatectomy and after unilateral nephrectomy. Thirty-six hours after partial hepatectomy, a maximum uptake 11-fold greater than in controls was found. The maximum after unilateral nephrectomy occurred 48 hr after operation and was twofold greater than in controls. Subsequent to these maxima, P32 uptake declined after both operations to a minimum eight days postoperatively which was followed by a second rise on the 9th day. These data support the concept that similar basic mechanisms regulate growth and cell division after the two types of operation. In another experiment, P32 uptake in kidney DNA nucleotides showed no increase after partial hepatectomy; this lends weight to the hypothesis that the mitosis-stimulating substances which appear in the serum after partial hepatectomy and after unilateral nephrectomy are organ specific.
