A new structural phase change at −60 °C and atmospheric pressure was observed in the gel phase of aqueous DMPC dispersions. This gel–gel phase transition exhibits first-order characteristics and is equivalent to the phase transition observed at 2.6 kbar and 30 °C. The decrease in temperature has analogous effects on the ordering of the acyl chains in DMPC as has the increase in pressure. The rigid interchain reorientational fluctuations in the gel phase are gradually damped with decreasing temperature and below −60 °C the interchain orientation becomes highly ordered.

Phosphoinositides (PIPs) are present in very small amounts but are essential for cell signaling, morphogenesis, and polarity. By mass spectrometry, we demonstrated that some PIPs with stearic acyl chains were strongly disturbed in apsi1ΔSaccharomyces cerevisiaeyeast strain deficient in the specific incorporation of a stearoyl chain at thesn-1 position of phosphatidylinositol. The absence of PIPs containing stearic acid induced disturbances in intracellular trafficking, although the total amount of PIPs was not diminished. Changes in PIPs also induced alterations in the budding pattern and defects in actin cytoskeleton organization (cables and patches). Moreover, when thePSI1gene was impaired, a high proportion of cells with bipolar cortical actin patches that occurred concomitantly with the bipolar localization of Cdc42p was specifically found among diploid cells. This bipolar cortical actin phenotype, never previously described, was also detected in abud9Δ/bud9Δ strain. Very interestingly, overexpression ofPSI1reversed this phenotype.

The temperature induced modifications in the lipid bilayers in water were studied by using 2D-FTIR Correlation Spectroscopy. The spectral range analysed in this paper corresponds to symmetric and asymmetric stretching vibrations of the �CH2 groups from the acyl chains of DPPC. Autocorrelation peaks at 2916 cm-1 and 2848.5 cm-1 and asynchrone modifications in the spectral ranges 2916-3000 cm-1 and 2848-3000 cm-1 were evidenced. The revealed modifications indicate changes in the conformers of the hydrocarbon chains in the phase transition from the gel to liquid crystalline phases, as well as the variations in the concentration of the two phases with temperature increasing.
