BACKGROUND: Red raspberry (Rubus idaeus L.), a natural dietary source of (poly)phenols, has been used as medicine for centuries. OBJECTIVE: The purpose of this study is to determine the effect of a red raspberry (poly)phenolic extract (RPE) on diet-induced obesity, hepatic steatosis and insulin resistance, and elucidate the underlying molecular mechanisms. METHODS: Male specific pathogen-free C57BL/6J mice were randomly divided into three groups (n = 12 per group), and fed with low-fat diet (10% fat energy), high-fat diet (HFD, 45% fat energy), or HFD supplemented with RPE of 150 mg/kg body weight by intragastric administration for 14 weeks. Obesity-related biochemical indexes and hepatic gene expression levels were determined. The statistical analyses were conducted using one-way analysis of variance (ANOVA) followed by Duncan’s multiple range test. RESULTS: The body weight gain, steatosis grade scores and insulin resistance index in the RPE group decreased by 34.48% (P = 0.00), 58.82% (P = 0.00), and 53.77% (P = 0.00), respectively, compared to those in the HFD group. Moreover, RPE supplement significantly changed the expression profile of the genes involved in lipid metabolism and fibroblast growth factor 21 signaling pathway. CONCLUSIONS: This study demonstrated that RPE protected from diet-induced obesity and related metabolic disorders by improving the lipid metabolism and fibroblast growth factor 21 resistance.

Background: Insulin resistance (IR) is an independent cardiovascular risk factor. IR predisposes to metabolic syndrome and diabetes. Meanwhile, little evidence exists about the effect of levothyroxine replacement treatment (LRT) on IR in hypothyroid patients. Objective: To investigate metabolic and anthropometric indices of IR in hypothyroid patients before and after 8 weeks of LRT. Methods: This pre-post study evaluated the 8 weeks outcomes of LRT on 66 patients with recently diagnosed hypothyroidism. Outcome measures included body mass index (BMI), waist circumferences (WC), waist to hip ratio (WHR), waist to height ratio (WHtR), body fat percent (BF%), free thyroxin (FT4), triglyceride (TG), low density lipoprotein (LDL), fasting plasma levels of glucose (FPG) and insulin. Sex- specific cut offs of two metabolic indices i.e. the triglyceride-glucose (TyG) and the homeostasis model assessment (HOMA) were used for IR diagnosis. The changes in TyG and HOMA were also compared after LRT. Results: Participants were overt and subclinical hypothyroidism 71% and 29%, respectively. After LRT the mean values of the following anthropometric indices significantly decreased: weight (79.61 vs. 78.64), BMI (29.53 vs. 29.2), WC (98.25 vs. 97.39) and BF% (35.34 vs. 34.95). After LRT the HOMA and TyG had no significant changes relative to their initial values. Also, IR that was determined on the basis of these metabolic indices more commonly observed in participants. Conclusion: Despite decreasing some anthropometric indices, the diagnosis of IR based on metabolic indices increased following 8 weeks LRT in hypothyroid cases.

Edible bird’s nest (EBN) is used traditionally in many parts of Asia to improve wellbeing, but there are limited studies on its efficacy. We explored the potential use of EBN for prevention of high fat diet- (HFD-) induced insulin resistance in rats. HFD was given to rats with or without simvastatin or EBN for 12 weeks. During the intervention period, weight measurements were recorded weekly. Blood samples were collected at the end of the intervention and oral glucose tolerance test conducted, after which the rats were sacrificed and their liver and adipose tissues collected for further studies. Serum adiponectin, leptin, F2-isoprostane, insulin, and lipid profile were estimated, and homeostatic model assessment of insulin resistance computed. Effects of the different interventions on transcriptional regulation of insulin signaling genes were also evaluated. The results showed that HFD worsened metabolic indices and induced insulin resistance partly through transcriptional regulation of the insulin signaling genes. Additionally, simvastatin was able to prevent hypercholesterolemia but promoted insulin resistance similar to HFD. EBN, on the other hand, prevented the worsening of metabolic indices and transcriptional changes in insulin signaling genes due to HFD. The results suggest that EBN may be used as functional food to prevent insulin resistance.

The group (totally156 postmenopausal women) used for the study of ‘standard’ (S) and ‘associated’ (A) genetic markers of potential sensitivity to metformin (MF) consisted of 37 healthy females, 32 — with diabetes (DM) without cancer, 64 cancer patients with DM, and 23 cancer patients without DM. No significant difference in carrying of S-polymorphisms was found between DM patients without and with cancer. In cancer patients without DM most characteristic data regarding potential MF-response were detected with polymorphisms of STK11 gene while data on OCT1_rs622342 and OCT1_R61C variants showed opposite trends. In regard of A-markers, the tendency to the more often finding of GC genotype of OLR1_G501C in DM patients carrying ‘MF-positive’ variant of OCT1_R61C deserves to be underlined. In patients with new-onset diabetes who carried S-markers of potential response to MF higher insulin resistance (OCT1_R61C and OCT1_rs622342) as well as lower estradiolemia (STK11 and C11orf65) were discovered. Thus, according to genetic S-criteria of sensitivity to MF, DM patients with and without cancer differ in lesser degree than they differ from cancer patients without DM. It can not be excluded, that The efficiency of such criteria might be increased due to combination with A-markers and certain hormonal-metabolic indices.
