BACKGROUND: Fertility is affected by both the reproductive organs and external factors (genetics, hormones, radiation exposure, use of insecticides, and nutrition). The histological structure of the ovaries is an indicator of reproductive function. Insect repellent use with pyrethroid active ingredients and its impact on health has become a discussion in the medical sector for years. Disruption of the reproductive system homeostasis may cause several issues, from disruption of ovarian function to infertility. γ-Oryzanol has higher antioxidants than vitamin E. It is found mostly in rice bran oil (RBO). Researchers have investigated the effectiveness of RBO and γ-Oryzanol, but the number of studies focusing on the reproductive system is very limited. Results of in silico showed anti-inflammatory potential, and nitric oxide γ-Oryzanol is stronger than the antioxidant activity. It also showed γ-Oryzanol bond with Foxo3a and Growth Differentiating Factor 9 (GDF9), indicating the γ-Oryzanol potential for reproductive health (women). Studies also reported that γ-Oryzanol administration caused anti-inflammatory and antioxidant activity compared to RBO in improving ovarian physiological function using tumor necrosis factor α (TNF-α) levels, Foxo3a expression, and GDF9 expression exposed to one push transfluthrin as the parameter. AIM: We aimed to investigate antioxidant and anti-inflammatory activity of γ-Oryzanol compared to rice bran oil to repair ovarian histological structure from one push transfluthrin exposure effect. METHODS: Experimental research, post-test only control group design approach, with a completely randomized design, consisted of 6 (six) groups of Wistar strain female rats. They were exposed to one push with the active ingredient of transfluthrin 21.3%, by inhalation for 6 hours, RBO 0.3 mg/g/ body weight (BW)/day, γ-Oryzanol 3.75 ml/g/BW/day. Statistical analysis was done with the Mann Whitney’s posthoc Kruskal Wallis test with IBM SPSS version 25 software. RESULTS: γ-Oryzanol had more potent anti-inflammatory and antioxidant activity than RBO in improving the ovarian histology structure (including maintaining ovarian weight, increasing follicular growth, and suppressing follicular abnormalities) through decreased TNF-α levels and decreased Foxo3a expression, and increased GDF9 expression. CONCLUSION: The administration of γ-Oryzanol improves the ovarian histological structure from free radicals effects, namely exposure to one push of transfluthrin.

Background and Objectives: For a long time, Asian women have known to use rice water, rice bran for skin care, lightening dark spots and slowing down the aging process. Gamma Oryzanol, an ester mixture of trans-ferulic acid with phytosterols and triterpene alcohols extracted from rice bran oil, has been shown to have beneficial effects on the skin, especially its antioxidant and anti-aging effect. The aims of this research were to formulate Gamma Oryzanol-loaded anti-aging cream and to evaluate the product quality. Materials and Methods: Gamma Oryzanol was imported from Japan. Some formulation parameters such as emulsifi-ers, performance modifiers were investigated and the product quality was evaluated according to the criteria of drugs and cosmetics. Results: The excipients were chosen including 1% (w/w) of Lunamer with 3% (w/w) of Prolipid as emulsifiers and 8% (w/w) of mango seed butter as a state modifier in the formulation of Gamma Oryzanol anti-aging cream. The cream is ivory-white, thick, smooth, fragrant-smelling and satisfied all criteria of developed specification. Conclusion: The study was successful in the preparation of Gamma Oryzanol anti-aging cream. Key words: Gamma Oryzanol, anti-aging cream, Franz cells.

Background: Hypercholesterolemia is a risk factor for developing coronary artery disease. Lifestyle modification including an intake of healthy food as well as medication have approved effect in lowering serum cholesterol. Objective: The primary objective of the present study was to determine the impact of a gamma-oryzanol-enriched rice bran oil, a product of Thailand, on serum cholesterol level. Materials and Methods: A total of in 54 hypercholesterolemic patients were divided into two groups; RBOh (20,000 ppm of gammaoryzanol, n = 27), and RBOn (5,000 ppm gamma-oryzanol, n = 27). The assigned RBO (15 ml) was intake each day for 8 weeks. Fasting serum lipids were measured at baseline and at the 4th and 8th weeks of the intervention. All patients were advised about lifestyle modifications. Results: When compared to the baseline, subjects received RBOh showed a significant difference in 2 parameters including a reduction of cholesterol level at 8th weeks (p-value = 0.0101), and decrease in LDL-C level at the end of 8th weeks (p-value = 0.0013). In the group treated with RBOn, a significant increase in HDL-C level at the end of 8th weeks (p-value = 0.0303) without any effect on total cholesterol or LDL was observed. No sign of toxic effect on liver or renal functions was seen in both treatment groups. Conclusion: RBO with gamma-oryzanol-enriched could decrease cholesterol and LDL-C level in hypercholesterolemic patients. Therefore, gamma-oryzanol-enriched RBO is a functional food that may reduce cardiovascular disease risk factor. Keywords: Hypercholesterolemia, Rice bran oil, Gamma-oryzanol, Oryza sativa

Chronic inflammation is a pathological process where cells of the mesenchymal lineage become a major source of inflammatory mediators. Platelet-rich fibrin (PRF) has been shown to possess potent anti-inflammatory activity in macrophages, but its impact on mesenchymal cells has not been investigated. The aim of this study was, therefore, to expose mesenchymal cells to inflammatory cytokines together with lysates generated from liquid platelet-poor plasma (PPP), the cell-rich buffy coat layer (BC; concentrated-PRF or C-PRF), and the remaining red clot layer (RC), following centrifugation of blood. Heating PPP generates an albumin gel (Alb-gel) that when mixed back with C-PRF produces Alb-PRF. Membranes prepared from solid PRF were also subjected to lysis. We report here that lysates of PPP, BC, and PRF decreased the cytokine-induced expression of interleukin 6 (IL6) and nitric oxide synthase (iNOS) in the bone marrow-derived ST2 cells. Consistently, PPP, BC, and PRF greatly decreased the phosphorylation and nuclear translocation of p65 in ST2 cells. The inflammatory response caused by Pam3CSK4 was reduced accordingly. Moreover, PPP, BC, and PRF reduced the enhanced expression of inflammatory mediators IL6 and iNOS in 3T3-L1 pre-adipocyte mesenchymal cells, and iNOS and CCL5 in murine calvarial cells. Surprisingly, PRF lysates were not effective in reducing the inflammatory response of human gingival fibroblasts and HSC2 epithelial cells. The data from the present study suggest that both liquid PRF and solid PRF exert potent anti-inflammatory activity in murine mesenchymal cells.

Abstract Background Macrophages play a crucial role in inflammation. Astilbe chinensis is one of perennial herbs belonging to the genus Astilbe. Plants in the genus have been used for pain, headaches, arthralgia, and chronic bronchitis. However, the effect of A.chinensis on inflammation remains unclear. To study the anti-inflammatory action of A.chinensis ethanol extract (ACE), we investigated the effect of ACE on the production of pro-inflammatory mediators and cytokines in macrophages. Methods We evaluated the effectiveness of ACE in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages and thioglycollate (TG)-elicited peritoneal macrophages from male C57BL/6 mice. We measured the levels of pro-inflammatory mediators and cytokines, and examined the anti-inflammatory actions of ACE on nuclear factor κB (NF-κB) pathway in the macrophages. Western blot analysis and immunofluorescence microscopy were used to determine protein level and translocation, respectively. Results ACE suppressed the output of nitric oxide (NO), prostaglandin E2 (PGE2), and pro-inflammatory cytokines in stimulated macrophages via inhibiting the expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) proteins. ACE suppressed mRNA expression of pro-inflammatory cytokines such as interleukin (IL)-6 and tumor necrosis factor-alpha (TNF-α). We examined the efficacies of ACE on NF-κB activation by measuring the expressions including IκB kinase (IKK), inhibitor of κB (IκB), and nuclear p65 proteins. In addition, the inhibition of NF-κB p65’s translocation was determined with immunofluorescence assay. Conclusion Our findings manifested that ACE inhibited LPS or TG-induced inflammation by blocking the NF-κB signaling pathway in macrophages. It indicated that ACE is a potential therapeutic mean for inflammation and related diseases.

Background. Different pharmacological properties, such as antioxidant, antiproliferative, and anti-inflammatory properties, have been described among natural products. We previously described that the Bougainvillea xbuttiana (Variety Orange) ethanolic extract (BxbO) has an anti-inflammatory effect; however, this action is not fully understood. In this study, the action of the BxbO extract on the secretion of inflammatory mediators in two experimental models, in vitro and in vivo, after LPS challenge was evaluated. Methods. Peritoneal macrophages were obtained from female BALB/c mice and LPS-challenged with or without the BxbO extract. For the evaluation of mediators, the supernatants at 0, 12, 24, 36, and 48 hours were collected. For in vivo estimation, groups of female BALB/c mice were first intraperitoneously injected with different amounts of LPS and later administered the oral BxbO extract (v.o.) for 144 hours. To understand the mechanism of action, sera obtained from mice were collected at 0, 2, 4, 8, 12, and 24 hours after LPS challenge (with or without BxbO) for the detection of mediators. Results. The results showed that, in both peritoneal macrophages and sera of mice treated with the BxbO extract 1 hour before or together with LPS challenge, proinflammatory cytokines and nitric oxide release were unquestionably repressed. In contrast, in both systems studied here, the IL-10 levels were elevated to 5 to 9 times. At lethal doses of LPS, the BxbO extract treatment was found to protect animals from death. Conclusions. The results revealed that the inhibitory, protective, and benign effects of the BxbO extract were due to its capacity to balance the secretion of mediators.

The aim of this study was to design and evaluate novel cyclodextrin (CD)-based aggregate formulations to efficiently deliver nepafenac topically to the eye structure, to treat inflammation and increase nepafenac levels in the posterior segment, thus attenuating the response of inflammatory mediators. The physicochemical properties of nine aggregate formulations containing nepafenac/γ-CD/hydroxypropyl-β (HPβ)-CD complexes as well as their rheological properties, mucoadhesion, ocular irritancy, corneal and scleral permeability, and anti-inflammatory activity were investigated in detail. The results were compared with a commercially available nepafenac suspension, Nevanac® 3 mg/mL. All formulations showed microparticles, neutral pH, and negative zeta potential (–6 to –27 mV). They were non-irritating and nontoxic and showed high permeation through bovine sclera. Formulations containing carboxymethyl cellulose (CMC) showed greater anti-inflammatory activity, even higher than the commercial formulation, Nevanac® 0.3%. The optimized formulations represent an opportunity for topical instillation of drugs to the posterior segment of the eye.
