Production of cell wall degrading enzymes (CWDEs) polygalacturonase (PG), pectate lyase (PL), and xylanase was studied in chickpeas ( Cicer arietinum L. ‘P-2245’) inoculated with Fusarium oxysporum f. sp. ciceris (Padwick) Matuo & K. Sato races 0 (mildly virulent, causing a yellowing syndrome) and 5 (highly virulent, causing a wilting syndrome) by the water-culture method. These CWDEs were similarly produced in both syndromes. PG and PL were the only enzymes occurring in roots and stems and attained the highest specific activity, this being generally higher for race 5 than for race 0. Gel filtration chromatography revealed a similar complement of in planta expressed pectinase isoforms, dominated by an endo-PG and two endo-PLs, the endo-PLs being differentially expressed by the two races. CWDE activities in roots and stems were positively correlated with development of yellowing and wilting. Exceptions to this were PG in stems, which was negatively correlated with the development of yellowing, and PG in roots, which showed a negative trend with development of either syndrome. The levels of CWDEs that significantly correlated with disease development were adequately described by exponential functions of disease progress. Results have implications for the role played by CWDEs in the early and later stages of pathogenesis in chickpea fusarium wilt.

This study aimed to disentangle the structure, composition, and co-occurrence relationships of the banana (cv. Dwarf Cavendish) root endophytome comparing two phenological plant stages: mother plants and suckers. Moreover, a collection of culturable root endophytes (>1000) was also generated from Canary Islands. In vitro antagonism assays against Fusarium oxysporum f.sp. cubense (Foc) races STR4 and TR4 enabled the identification and characterization of potential biocontrol agents (BCA). Eventually, three of them were selected and evaluated against Fusarium wilt of banana (FWB) together with the well-known BCA Pseudomonas simiae PICF7 under controlled conditions. Culturable and non-culturable (high-throughput sequencing) approaches provided concordant information and showed low microbial diversity within the banana root endosphere. Pseudomonas appeared as the dominant genus and seemed to play an important role in the banana root endophytic microbiome according to co-occurrence networks. Fungal communities were dominated by the genera Ophioceras, Cyphellophora, Plecosphaerella, and Fusarium. Overall, significant differences were found between mother plants and suckers, suggesting that the phenological stage determines the recruitment and organization of the endophytic microbiome. While selected native banana endophytes showed clear antagonism against Foc strains, their biocontrol performance against FWB did not improve the outcome observed for a non-indigenous reference BCA (strain PICF7).

Striga species cause significant yield loss in maize varying from 20 to 100%. The aim of the present study was to screen and identify maize genotypes with partial resistance to S. hermonthica (Sh) and S. asiatica (Sa) and compatible with Fusarium oxysporum f. sp. strigae (FOS), a biocontrol agent. Fifty-six maize genotypes were evaluated for resistance to Sh and Sa, and FOS compatibility. Results showed that FOS treatment significantly (p < 0.001) enhanced Striga management compared to the untreated control under both Sh and Sa infestations. The mean grain yield was reduced by 19.13% in FOS-untreated genotypes compared with a loss of 13.94% in the same genotypes treated with FOS under Sh infestation. Likewise, under Sa infestation, FOS-treated genotypes had a mean grain yield reduction of 18% while untreated genotypes had a mean loss of 21.4% compared to the control treatment. Overall, based on Striga emergence count, Striga host damage rating, grain yield and FOS compatibility, under Sh and Sa infestations, 23 maize genotypes carrying farmer preferred traits were identified. The genotypes are useful genetic materials in the development of Striga-resistant cultivars in Tanzania and related agro-ecologies.

Thirty-four isolates of Fusarium oxysporum, obtained in China from cucumber plants showing either Fusarium wilt (F. oxysporum f. sp. cucumerinum) or root and stem rot (F. oxysporum f. sp. radicis-cucumerinum) symptoms, were characterized by pathogenicity, vegetative compatibility, and random amplified polymorphic DNA (RAPD). Of these, 23 isolates were identified by pathogenicity as F. oxysporum f. sp. cucumerinum, and one as F. oxysporum f. sp. radicis-cucumerinum, while 10 isolates were avirulent on cucumber, melon, sponge gourd, and pumpkin. The Chinese isolates of F. oxysporum f. sp. cucumerinum were assigned to RAPD groups III and XXI and to vegetative compatibility group (VCG) 0183, four new VCGs, 0184 to 0187, and a single-member VCG included in the artificial VCG 018-. The Chinese isolate of F. oxysporum f. sp. radicis-cucumerinum was assigned to RAPD group I and bridging VCG 0260/0261. The occurrence of F. oxysporum f. sp. radicis-cucumerinum on cucumber is reported for the first time in China.

ABSTRACT A type II arabinogalactan-degrading enzyme (FoGal1) was purified from Fusarium oxysporum 12S, and the corresponding cDNA was isolated. FoGal1 had high similarity to enzymes of glycoside hydrolase family 5. Treatment of larch wood arabinogalactan with the recombinant enzyme indicated that FoGal1 is a β-1,6-galactanase that preferentially debranches β-1,6-galactobiose from the substrate.
