Growth-correlative effect of the root on the apical part of the epicotyl in pea seedlings regarding the IAA content and L-tryptophan aminotransferase and L-tryptophan dehydrogenase activities

The effect of fluridone (or flurochloridone), the inhibitor of carotenoid, chlorophyll and abscisic acid synthesis, on the abscission of Ligustrum vulgare leaves was investigated. Both forms of fluridone inhibited the abscission of petioles when they were applied as a 1.0% concentration in lanolin to the leaf blade. Fluridone was capable of inhibiting abscission even when it was applied to the petiole as late as 7 hours after the blade was cut off. Fluridone applied in lanolin to the apical part of intact pea seedlings or on the cut surface after decapitation of the epicotyl caused albinism of the stipules. The degree of albinism decreased according to the concentration of applied flurodine (from the highest – 0.5% to the lowest – 0.03%) and was higher in intact than in decapitated plants. Albinism also appeared in pea seedlings grown from seeds swollen in differently concentrated solutions of flurochloridone, particularly in the basal part of the shoots. Complete albinism occurred only in plants cultivated from seeds swollen in high concentrations (0.06–0.12%). The lowest concentration (0.007%) stimulated root growth of the pea seedlings, but inhibited epicotyl growth.

Pea seeds were swollen in solutions with different concentrations of cytokinin benzyladenine (BA). As compared with controls, a low concentration of BA (0.05 mg/l) did not affect the growth of stems and roots of twelve-day-old seedlings but concentrations of 0.20–4.00 mg/l showed an inhibiting effect; this inhibition was proportional to the concentration of BA. In controls the ratio between the lengths of stems and roots was equal to 1.14, while in seedlings influenced by BA in concentration of 0.20 mg/l its value was significantly lowered to 0.91. Also in six-day-old pea seedlings cultivated under in vitro conditions, BA concentrations of 0.70 and 3.30 mg/l inhibited the growth of roots and epicotyls and significantly increased the production of ethylene (by 150 and 330%, respectively). Three hours after the amputation of the root of five-day-old pea seedlings the level of cytokinin trans-zeatin increased in the apical part of the stem. Within the interval of 48 hours after the amputation of the root the concentration of this plant hormone gradually decreased again. The initial increase in the zeatin level in the stem indicates that the absence of the root induces a reduction of growth-inhibiting effects of roots, which negatively influences the zeatin level in the stem apex at the beginning of germination.

Lectin histochemistry was used to perform in situ characterization of the glycoconjugates present in boar testis and epididymis. Thirteen horseradish peroxidase- or digoxigenin-labelled lectins were used in samples obtained from healthy fertile boars. The acrosomes of the spermatids were stained intensely by lectins with affinity for galactose and N-acetyl-galactosamine residues, these being soybean, peanut and Ricinus communis agglutinins. Sertoli cells were stained selectively by Maackia ammurensis agglutinin. The lamina propria of seminiferous tubules showed the most intense staining with fucose-binding lectins. The Golgi area and the apical part of the principal cells of the epididymis were stained intensely with many lectins and their distribution was similar in the three zones of the epididymis. On the basis of lectin affinity, both testis and epididymis appear to have N- and O-linked glycoconjugates. Spermatozoa from different epididymal regions showed different expression of terminal galactose and N-acetyl-galactosamine. Sialic acid (specifically alpha2,3 neuraminic-5 acid) was probably incorporated into spermatozoa along the extratesticular ducts. These findings indicate that the development and maturation of boar spermatozoa are accompanied by changes in glycoconjugates. As some lectins stain cellular or extracellular compartments specifically, these lectins could be useful markers in histopathological evaluation of diseases of boar testis and epididymis.

Background: One of the main sources of ROS in stress conditions is the mitochondria. Excessive generation of ROS leads to oxidation of thiol groups of proteins, peroxidation of membrane lipids and swelling of the mitochondria. In this regard, there is a need to search for preparationsadaptogens that increase the body's resistance to stress factors. Perhaps, antioxidants can serve as such adaptogens. This work aims at studying the effect of antioxidant; the potassium anphen in a wide range of concentrations on the functional state of 6 day etiolated pea seedlings mitochondria (Pisum sativum L). Methods: The functional state of mitochondria was studied per rates of mitochondria respiration, by the level of lipid peroxidation and study of fatty acid composition of mitochondrial membranes by chromatography technique. Results: Potassium anphen in concentrations of 10-5 - 10-8 M and 10-13-10-16 prevented the activation of LPO in the mitochondrial membranes of pea seedlings, increased the oxidation rates of NAD-dependent substrates and succinate in the respiratory chain of mitochondria that probably pointed to the anti-stress properties of the drug. Indeed, the treatment of pea seeds with the preparation in concentrations of 10-13 M prevented the inhibition of growth of seedlings in conditions of water deficiency. Conclusion: It is assumed that the dose dependence of the biological effects of potassium anphen and the manifestation of these effects in ultra-low concentrations are due to its ability in water solutions to form a hydrate containing molecular ensembles (structures).

The mechanical discontinuities in the upper crust (i.e., faults and related fractures) lead to the uprising of geothermal fluids to the Earth’s surface. If fluids are enriched in Ca2+ and HCO3-, masses of CaCO3 (i.e., travertine deposits) can form mainly due to the CO2 leakage from the thermal waters. Among other things, fissure-ridge-type deposits are peculiar travertine bodies made of bedded carbonate that gently to steeply dip away from the apical part where a central fissure is located, corresponding to the fracture trace intersecting the substratum; these morpho-tectonic features are the most useful deposits for tectonic and paleoseismological investigation, as their development is contemporaneous with the activity of faults leading to the enhancement of permeability that serves to guarantee the circulation of fluids and their emergence. Therefore, the fissure ridge architecture sheds light on the interplay among fault activity, travertine deposition, and ridge evolution, providing key geo-chronologic constraints due to the fact that travertine can be dated by different radiometric methods. In recent years, studies dealing with travertine fissure ridges have been considerably improved to provide a large amount of information. In this paper, we report the state of the art of knowledge on this topic refining the literature data as well as adding original data, mainly focusing on the fissure ridge morphology, internal architecture, depositional facies, growth mechanisms, tectonic setting in which the fissure ridges develop, and advantages of using the fissure ridges for neotectonic and seismotectonic studies.

This article discusses the peculiarities of mineral composition and a fluid inclusions (FIs further in the text) study of the Kholtoson W and Inkur W deposits located within the Dzhida W-Mo ore field (Southwestern Transbaikalia, Russia). The Mo mineralization spatially coincides with the apical part of the Pervomaisky stock (Pervomaisky deposit), and the W mineralization forms numerous quartz veins in the western part of the ore field (Kholtoson vein deposit) and the stockwork in the central part (Inkur stockwork deposit). The ore mineral composition is similar at both deposits. Quartz is the main gangue mineral; there are also present muscovite, K-feldspar, and carbonates. The main ore mineral of both deposits is hubnerite. In addition to hubnerite, at both deposits, more than 20 mineral species were identified; they include sulfides (pyrite, chalcopyrite, galena, sphalerite, bornite, etc.), sulfosalts (tetrahedrite, aikinite, stannite, etc.), oxides (scheelite, cassiterite), and tellurides (hessite). The results of mineralogical and fluid inclusions studies allowed us to conclude that the Inkur W and the Kholtoson W deposits were formed by the same hydrothermal fluids, related to the same ore-forming system. For both deposits, the fluid inclusion homogenization temperatures varied within the range ~195–344 °C. The presence of cogenetic liquid- and vapor-dominated inclusions in the quartz from the ores of the Kholtoson deposit allowed us to estimate the true temperature range of mineral formation as 413–350 °C. Ore deposition occurred under similar physical-chemical conditions, differing only in pressures of mineral formation. The main factors of hubnerite deposition from hydrothermal fluids were decreases in temperature.
