MicroRNAs are small regulatory RNAs involved in several processes in plants ranging from development and stress responses to defense against pathogens. In order to accomplish their molecular functions, miRNAs are methylated and loaded into one ARGONAUTE (AGO) protein, commonly known as AGO1, to stabilize and protect the molecule and to assemble a functional RNA-induced silencing complex (RISC). A specific machinery controls miRNA turnover to ensure the silencing release of targeted-genes in given circumstances. The trimming and tailing of miRNAs are fundamental modifications related to their turnover and, hence, to their action. In order to gain a better understanding of these modifications, we analyzed Arabidopsis thaliana small RNA sequencing data from a diversity of mutants, related to miRNA biogenesis, action, and turnover, and from different cellular fractions and immunoprecipitations. Besides confirming the effects of known players in these pathways, we found increased trimming and tailing in miRNA biogenesis mutants. More importantly, our analysis allowed us to reveal the importance of ARGONAUTE 1 (AGO1) loading, slicing activity, and cellular localization in trimming and tailing of miRNAs.

Seven compounds (1-7) were isolated from the marine sponge Aaptos aaptos living in the Vietnamese sea. Their structures were determined as 2 hours, 5 H,7 H,9 H-9 S-hydroxy-imidazo[1,5- α]pyridine-1,3-dione (1), 3-([9-methylhexadecyl]oxy)propane-1,2-diol 2, 2,3-dihydro-2,3-dioxoaaptamine (3), indol-3-aldehyde (4), methyl indole-3-carboxylate (5) 4-hydroxy-5-(indole-3-yl)−5-oxo-pentan-2-one (6), and thymidine (7) by extensive analysis of HR-ESI-MS, 1D, and 2D NMR spectral data, as well as by comparison of the spectral data with those reported in the literature. In addition, the absolute configuration of 1 was determined from the experimental ECD spectrum and comparison of this with the theoretical ECD calculations using the TDDFT method. Compounds 1 and 2 were isolated from nature for the first time. Compound 3 induced cytotoxic activity against SK-LU-1, MCF-7, HepG2, and SK-Mel-2 cell lines with IC50 values of 41.27 ± 2.63, 40.70 ± 2.65, 34.31 ± 3.43, and 36.63 ± 1.40 µM, respectively.

A new δ-truxinic acid sucrose diester and a new ε-truxillic acid sucrose diester (named trigohonbanosides E and F) were isolated from the leaves of Trigonostemon honbaensis. Their chemical structures were determined by extensive analysis of their HR-ESI-MS and NMR spectra. At a concentration of 20 µM, trigohonbanosides E and F exhibited weak inhibitory effects on NO production in LPS-activated RAW264.7 cells with inhibitory percentages of 22.7% ± 1.1% and 18.5% ± 1.4%, respectively.

Abstract This paper deals mainly with some of the difficulties the translator might encounter when translating some culturally bound pieces of information. These would include the translation of some idioms, and some culturally bound concepts. The paper starts with definitions of translation, language and culture followed by an extensive analysis of the examples provided. All the examples are drawn from Arabic and English. The examples include the translation of some idioms which violate truth conditions, which are easily recognizable, and some others which may be translated either literally or idiomatically with obviously different results. Then the analysis moves to the translation of some culturally bound expressions from both Arabic and English. Here, we find examples that cannot be translated into the other language simply for lack of cultural equivalents. The skill and the intervention of the translator are most needed in this respect because above all translation is an act of communication. Résumé Cet article traite principalement de certaines difficultés que le traducteur peut rencontrer quand il traduit des textes d’information qui présentent un aspect culturel. Ces difficultés ont trait à certaines locutions idiomatiques et concepts culturels. L’article commence par définir la traduction, la langue et la culture, puis analyse en détail les exemples fournis. Tous les exemples sont tirés de l’arabe et de l’anglais. Ces exemples comprennent la traduction de certaines locutions idiomatiques qui trahissent les conditions de vérite et sont facilement reconnaissables, et de quelques autres qui peuvent etre traduites soit litteralement, soit de manière idiomatique, mais avec bien sur des résultats différents. Puis l’analyse passe à la traduction de certaines expressions de nature culturelle, en arabe et en anglais. Nous y trouvons des exemples qu’il est impossible de traduire dans l’autre langue, tout simplement parce qu’il leur manque des équivalents culturels. L’habileté et l’intervention du traducteur sont des plus nécéssaires dans ce cas, parce que la traduction est avant tout un acte de communication.

The Haines Index is an operational tool for evaluating the potential contribution of dry, unstable air to the development of large or erratic plume-dominated wildfires. The index has three variants related to surface elevation, and is calculated from temperature and humidity measurements at atmospheric pressure levels. To effectively use the Haines Index, fire forecasters and managers must be aware of the climatological and statistical characteristics of the index for their location. However, a detailed, long-term, and spatially extensive analysis of the index does not currently exist. To meet this need, a 40-year (1961–2000) climatology of the Haines Index was developed for North America. The climatology is based on gridded (2.5° latitude × 2.5° longitude) temperature and humidity fields from the NCEP/NCAR reanalysis. The climatology illustrates the large spatial variability in the Haines Index both within and between regions using the different index variants. These spatial variations point to the limitations of the index and must be taken into account when using the Haines Index operationally.

Recently, many states have reversed the decades-long trend of facilitating ballot access by enacting a wave of laws requesting or requiring identification from registrants before they vote. Identification laws, however, are not an entirely new phenomenon. We offer new theoretical insights regarding how changes in political power influence the adoption of identification laws. In the most extensive analysis to date, we use event history analysis to examine why states adopted a range of identification laws over the past several decades. We consistently find that the propensity to adopt is greatest when control of the governor’s office and legislature switches to Republicans (relationships not previously identified), and that this likelihood increases further as the size of Black and Latino populations in the state expands. We also find that federal legislation in the form of the Help America Vote Act seems to enhance the effects of switches in partisan control.

The Asiatic wild dog (Cuon alpinus), restricted today largely to South and Southeast Asia, was widespread throughout Eurasia and even reached North America during the Pleistocene. Like many other species, it suffered from a huge range loss towards the end of the Pleistocene and went extinct in most of its former distribution. The fossil record of the dhole is scattered and the identification of fossils can be complicated by an overlap in size and a high morphological similarity between dholes and other canid species. We generated almost complete mitochondrial genomes for six putative dhole fossils from Europe. By using three lines of evidence, i.e., the number of reads mapping to various canid mitochondrial genomes, the evaluation and quantification of the mapping evenness along the reference genomes and phylogenetic analysis, we were able to identify two out of six samples as dhole, whereas four samples represent wolf fossils. This highlights the contribution genetic data can make when trying to identify the species affiliation of fossil specimens. The ancient dhole sequences are highly divergent when compared to modern dhole sequences, but the scarcity of dhole data for comparison impedes a more extensive analysis.

Abstract Construction of a cross-country pipeline involves lifting the pipeline off the skids and lowering it into the trench (lifting and lowering-in). This can introduce the highest stress magnitude that the pipe may experience over its service life. If not managed properly, overly high stresses may cause integrity issues during construction and/or service. If the girth welds are qualified and accepted using alternative flaw acceptance criteria, such as those in API 1104 Annex A and CSA Z662 Annex K, these stresses must be kept below a preset level during lifting and lowering-in to satisfy the requirements of those standards. This paper covers the development and usage of a stress analysis tool for the continuous lifting and lowering-in of pipe strings without a concrete coating or river weights. The outcome of the stress analysis can be used to develop lifting and lowering-in plans for construction crews. The core functionality of the application tool is to calculate the stresses from bending in the vertical and horizontal planes. The stresses from vertical bending are derived from an extensive analysis of continuous lifting and lowering-in processes. The stresses from horizontal bending are calculated using closed-form analytical solutions. The tool provides a graphical interface that interprets the background stress analysis results and displays information necessary for the development of lifting and lowering-in plans. The tool can be used to evaluate what-if scenarios for various tentative lifting and lowering-in scenarios. The process of using the tool to develop lifting and lowering-in plans is demonstrated in this paper through an example problem. The number of sidebooms and other lifting and lowering-in parameters such as sideboom spacing and lifting height range are changed to make the lifting and lowering-in plan easy to use for the laying contractors. Such tradeoffs can be addressed proactively with construction contractors to ensure that a mutually acceptable approach to lifting and lowering-in is taken.
