Paclitaxel is one of the cancer drugs that often used. These drug kills cancer cells byinhibiting mitotic cycle. The efficiency of paclitaxel is increased by the use ofnanomaterials as a carrier of paclitaxel. Nanomaterials can enhance encapsulationefficiency, improve the drug release to the target cell following nanomaterialdegradation, and improve local accumulation of drug in the cell through endocytosisreceptor. Nanomaterial that often used forencapsulation of paclitaxel is a polymerderived from natural resources such as cellulose. The advantages of cellulose as acarrier of paclitaxel are nontoxic, biodegradable, and very abundant from varioussources. The mitotic cycle can be arrested in the apical summit of vegetative terminal buds of Tradescantia paludosa by restricting the level of nitrogen or light available to the plant. Cells in this portion of the bud are much more sensitive to these stress conditions than those in the subjacent portion of the meristem. This differential response induced the establishment of a quiescent "central zone" which is distinguished from the rest of the meristem by the apparent absence of mitosis and DNA synthesis, larger nuclear volume, and a lower histone content of chromatin. These features are identical with those imposed by apical dominance in apices of inhibited lateral buds.The results support the view that competition for nutrients is an important causal factor in apical dominance. They suggest also that competition for nutrients within the terminal bud meristem is important in the regulation of growth in vegetative shoots in respones to conditions of the environment.

ABSTRACT Ciliated protozoa contain two types of nuclei, germ line micronuclei (Mic) and transcriptionally active macronuclei (Mac). During sexual reproduction, the parental Mac degenerates and a new Mac develops from a mitotic product of the zygotic Mic. Macronuclear development involves extensive endoreplication of the genome. The present study shows that endoreplication of macronuclear DNA in Tetrahymena is an example of endocyling, a variant of the mitotic cycle with alternating S and G phases in the absence of cell division. Thus, endocycling is conserved from ciliates to multicellular organisms. The gene ASI2 in Tetrahymena thermophila encodes a putative signal transduction receptor. ASI2 is nonessential for vegetative growth, but it is upregulated during development of the new Mac. Cells that lack ASI2 in the developing Mac anlagen are arrested in endoreplication of the DNA and die. This study shows that ASI2 is also transcribed in the parental Mac early in conjugation and that transcription of ASI2 in the parental Mac supports endoreplication of the DNA during early stages of development of the Mac anlagen. Other molecular events in Mac anlage development, including developmentally regulated DNA rearrangement, occur normally in matings between ASI2 knockouts, suggesting that ASI2 specifically regulates endocycling in Tetrahymena.

With the use of the light and electron microscopes, the chromosomes of Fritillaria lanceolata and Scilla sibirica are shown to differ in respect of the heterochromatin they contain. In root meristems of the former, the heterochromatic regions (H-segments) were recognizable at all phases of the mitotic cycle by their slighter opacity to electrons than that of euchromatic parts. This was due both to less tight packing of the chromatin fibrils and lower opacity of the fibrils themselves, even though both had the same diameter, about 3 nm. In root tips of the Scilla, the heterochromatin was invariably of similar opacity to euchromatin and thus only recognizable at telophase and interphase as large chromocentres. The opacity to electrons in the heterochromatin of the 2 species, was at all times closely paralleled by the staining behaviour seen with the light microscope in sections (0.07-0.5 µm in thickness) stained with toluidine blue. The disparity in the Fritillaria, as seen in sections with the light microscope, in respect of the stainability of the hetero- and euchromatin, was masked in Feulgen squash preparations of root tips from plants grown at 18-20 °C; at metaphase by the thickness of the chromosomes and at interphase by the density of the chromocentres. When, on the other hand, the plants were grown for 4 days at 2 °C, the masking effect of thickness was circumvented in metaphase chromosomes by differential super-contraction in euchromatin. The implications of these findings in respect to previous interpretations of the differential reactivity of H-segments to low temperature, as well as the phenomenon of enhanced and reduced fluorescence in these segments with fluorochromes are discussed.
